Background. Atherosclerotic cardiovascular diseases, as well as coronary heart disease, cerebrovascular disease, and peripheral artery disease (probably caused by atherosclerosis), are the leading cause of disability and mortality in people with diabetes. Objective. The purpose of our study was to determine the histostructural features and morphometric analysis of the components of the aortic wall and its hemomicrocirculatory bed after 2 and 4 weeks of streptozotocin-induced diabetes mellitus. Methods. The samples for the histology were the sections of the wall of the ascending part, the aortic arch, and the descending part of the aorta of 26 mature white male rats weighing 100 - 160 g. For morphometric examination, a series of images of the aortic wall was taken using a Meiji MT4300 LED microscope with an x40 objective, x10 eyepiece. Results and conclusion. The measurements were carried out using the ImageJ software. The development of micro- and macroangiopathies in experimental animals with 8-week streptozotocin-induced diabetes mellitus was histologically proved. Statistical analysis revealed a significant difference of all morphometric parameters of both - the components of the aortic wall and the vessels of its hemomicrocirculatory bed after 4 weeks of experimental diabetes in comparison with the norm, control, and the 2-nd week of the experiment. An explicit dependency of the severity of destructive changes in the wall of the aorta and links of its hemomicrocirculatory bed of vessels on the duration of the experiment was determined.

Background: Recent studies have revealed that a hyperglycemia-induced overproduction of superoxide can be the first event in the activation of all pathways involved in the pathogenesis of complications of diabetes. Supplementation of garlic was found to decrease diabetes-induced oxidative stress complications. Studies shown also that melatonin attenuates diabetes‐induced oxidative stress in diabetic induced rabbits and rats. Objective: In this present study, oxidative stress in diabetic model and the effect of garlic oil or melatonin treatment were examined in both genders' male and females' mice. Methods: 96 mice were randomly divided into 5 groups including control (C), diabetic (D), melatonin 10 mg/kg (D+M), garlic extract 100 mg/kg (D+G) and combined melatonin and garlic (D+M+G). All treatments were given orally daily for 16 weeks after induction of hyperglycemia by streptozocin (STZ). Fasting blood glucose and antioxidant levels were estimated. Results: Streptozotocin induced diabetic mice, showed a significant increase of plasma glucose, lipid peroxide and uric acid. Accordingly, significant decreases in the levels of antioxidants ceruloplasmin were found in the plasma of diabetic mice. Treatment of diabetic mice with garlic oil or melatonin for 16 weeks significantly increased plasma levels of ceruloplasmin activities. Lipid peroxides, uric acid, blood glucose was decreased significantly after treatment with garlic oil or melatonin. Conclusion: The results suggest that garlic oil or melatonin may effectively normalize the impaired antioxidants status in streptozotocin induced diabetes in both males and females mice.

This study was undertaken to evaluate the remedial effect of the aqueous seed extract of Mucuna pruriens (ASEMP) on the endocrine region of pancreas of nicotinamide-streptozotocin-induced diabetes in Wistar rats. The anti-hyperglycemic efficiency of two varieties of Mucuna pruriens var utilis; IIHR Selection 3 and Arka Dhanvantari was investigated. Oral administration of ASEMP against the nicotinamide-streptozotocin-induced diabetes in the Wistar rats showed anti-hyperglycemic effect on the blood glucose level (94 ± 0.2 mg/dl ASEMP 400 mg/kg) when compared with the control group (92 ± 0.2 mg/dl glibenclamide 5 mg/kg), (248 ± 0.3 mg/dl Diabetic control) and (90 ± 0.3 mg/dl Normal control). Significant reduction in creatinine level of the nicotinamide-streptozotocin-induced diabetic rats treated with ASEMP was also recorded. Histopathology examination of the endocrine region of pancreas of the rats revealed restoration of pancreatic islet cells in the diabetic-ASEMP treated rats as the beta cell mass increased and necrotic changes was reduced significantly in contrast with the diabetic control group which showed degenerated pancreatic islet cells. Therefore, this study supports and recommends the exploration of the aqueous seed extract of M. pruriens as an alternative medicine for the management of type-2 diabetes.

Honey has several pharmacological effects, including anti-diabetic activity. However, the effectiveness of bitter gourd honey (BGH) in the treatment of diabetes mellitus (DM) is unknown. The aim of this study was to determine the antioxidant, anti-inflammatory, and anti-apoptotic properties of BGH on the kidney and liver of a streptozotocin-induced diabetes rat model. Methods: A single dose (nicotinamide 110 mg/kg, streptozotocin (STZ) 55 mg/kg, intraperitoneal (i.p.)) was used to induce DM in male rats. For 28 days, normal or diabetic rats were administered 1 g/kg/day and 2 g/kg/day of BGH orally. After the treatment, blood, liver, and kidney samples were collected and analysed for biochemical, histological, and molecular parameters. In addition, liquid chromatography–mass spectrometry (LC-MS) was used to identify the major bioactive components in BGH. Results: The administration of BGH to diabetic rats resulted in significant reductions in alanine transaminase (ALT),aspartate aminotransferase (AST), creatinine, and urea levels. Diabetic rats treated with BGH showed lesser pathophysiological alterations in the liver and kidney as compared to non-treated control rats. BGH-treated diabetic rats exhibited reduced levels of oxidative stress (MDA levels), inflammatory (MYD88, NFKB, p-NFKB, IKKβ), and apoptotic (caspase-3) markers, as well as higher levels of antioxidant enzymes (SOD, CAT, and GPx) in the liver and kidney. BGH contains many bioactive compounds that may have antioxidative stress, anti-inflammatory, and anti-apoptotic effects. Conclusion: BGH protected the liver and kidney in diabetic rats by reducing oxidative stress, inflammation, and apoptosis-induced damage. As a result, BGH can be used as a potential therapy to ameliorate diabetic complications.

According to the World Health Organization, cardiovascular diseases and diabetes mellitus occupy a significant niche in the structure of diseases with high disability and mortality impact and pose major healthcare and social issues. It should be stressed that 50-80% of fatal cases in patients with diabetes are associated with cardiovascular diseases. The purpose of this study is to determine the histostructural characteristics and to perform morphometric analysis of the components of the aortic wall and its hemomicrocirculatory bed of the aorta in 6 and 8 weeks of streptozotocin-induced diabetes mellitus. The material for the histological study included the sections of the wall of the ascending aorta, the aortic arch, and the descending aorta taken from 26 sexually mature white male rats weighing 100 - 160 g. For morphometric examination, a series of photos of the aortic wall was taken using a Meiji MT4300 LED microscope with an x40 objective, x10 ocular. The measurements were carried out using the Image J software. The development of micro - and macroangiopathies in experimental animals with 8-week streptozotocin-induced diabetes mellitus was histologically confirmed. Statistical analysis revealed a significant difference in all morphometric parameters of the components of the aortic wall and the vessels of its hemomicrocirculatory bed after 8 weeks of experimental diabetes as compared with the normal values, control values, and in values obtained at the 6-week period of the experiment. The study has demonstrated clear dependence between the severity of destructive changes in the aortic wall and sections of its hemomicrocirculatory bed and the duration of the experiment.

For the diagnosis of diabetes two major tests are done blood glucose level and Glycated hemoglobin content (HbA1c). Ayurveda is the super bank for various molecules to treat any disease. Here we have chosen the super critical fluid extract of Ginger that is Zingiber Officinale. In our study we have used Streptozotocin induced diabetes mellitus model by using rats. Our result showed promising effects in the streptozotocine induced diabetes and immunomodulatory activity was also shown by the extract. Overall in conclusion it Zingiber Officinale supercritical fluid extract can be considered as source of multitalented molecules which can be proven better for the management of diabetes and its related
