Triterpenic compounds stand as a widely investigated class of natural compounds due to their remarkable therapeutic potential. However, their use is currently being hampered by their low solubility and, subsequently, bioavailability. In order to overcome this drawback and increase the therapeutic use of triterpenes, cyclodextrins have been introduced as water solubility enhancers; cyclodextrins are starch derivatives that possess hydrophobic internal cavities that can incorporate lipophilic molecules and exterior surfaces that can be subjected to various derivatizations in order to improve their biological behavior. This review aims to summarize the most recent achievements in terms of triterpene:cyclodextrin inclusion complexes and bioconjugates, emphasizing their practical applications including the development of new isolation and bioproduction protocols, the elucidation of their underlying mechanism of action, the optimization of triterpenes’ therapeutic effects and the development of new topical formulations.

This research is focused on cellulose and starch derivatives, carboxymethyl cellulose (CMC) and carboxymethyl starch (CMS), added to the detergent in washing reference cotton fabric in soft and hard water at 40, 60 and 90 °C. The applied polymers were analyzed through the potential of surface cellulose modification and inhibition of stain transfer from standard stain donors to modified and initial cotton fabrics. The surface modification of the cotton fabrics, characterized by the zeta potential and amounts of deposits, was coupled with the cluster analysis as well as a whiteness assessment. The obtained results of the zeta potential and degree of whiteness of the reference cotton fabrics before and after washing showed differences between CMC and CMS. The appropriateness of the cluster analysis was confirmed in assessing the potential of applied polymers for surface modification of cotton fabrics and greying inhibition.

Cyclodextrins (CDs) are a group of cyclic oligosaccharides produced from starch or starch derivatives. They contain six (αCD), seven (βCD), eight (γCD), or more glucopyranose monomers linked via α-1,4-glycosidic bonds. CDs have a truncated cone shape with a hydrophilic outer wall and a less hydrophilic inner wall, the latter forming a more apolar internal cavity. Because of this special architecture, CDs are soluble in water and can simultaneously host lipophilic guest molecules. The major advantage of inclusion into CDs is increased aqueous solubility of such lipophilic substances. Accordingly, we present studies where the complexation of natural compounds such as propolis and dietary plant bioactives (e.g., tocotrienol, pentacyclic triterpenoids, curcumin) with γCD resulted in improved stability, bioavailability, and bioactivity in various laboratory model organisms and in humans. We also address safety aspects that may arise from increased bioavailability of plant extracts or natural compounds owing to CD complexation. When orally administered, α- and βCD—which are inert to intestinal digestion—are fermented by the human intestinal flora, while γCD is almost completely degraded to glucose units by α-amylase. Hence, recent reports indicate that empty γCD supplementation exhibits metabolic activity on its own, which may provide opportunities for new applications.

Abstract This chapter describes the general method for isolation of starch from tuber crops, the separation of amylose and amylopectin, and the determination of starch content. The methods for the determination of the physical, granular, chemical, structural, rheological and thermal properties of starch, in vitro starch digestibility, and starch derivatives are discussed.

Based on the pharmacokinetics of nitazole, proprietary oral administration of the rabbit analyzes various aspects of its use as auxiliary substances in the preparation of dosage forms with controlled absorption of starch, carboxymethyl starch, dialdehyde starch, sodium alginate and chitosan. Our analysis of pharmacokinetic curves revealed a correlation between the presence and magnitude of the negative charge in the polymer of starch derivatives and an increase in the bioavailability of nitazole, the absorption rate estimated by tmax and from the equations describing the curves in the framework of the single-particle absorption model. It can be assumed that the change in the bioavailability of nitazole in the presence of starch derivatives is associated with the ion-ion or ion-dipole interaction of the positively charged imine group of nitazole and negatively charged groups of starch derivatives. Obviously, in the mechanism of absorption of nitazole, the limiting stage is the stage of dissolution of nitazole in the stomach.Since the absorption of nitazole may also depend on the microviscosity of the membranes of the cells of the walls of the stomach, the effect of the above polysaccharides on the microviscosity of the membranes of red blood cells as model cells has been studied. However, the work revealed a noticeable effect of only chitosan on the microviscosity of erythrocyte membranes, in which there was some immobilization of the conformational mobility of the lipid bilayer of the cell membranes upon binding of the oppositely charged chitosan to the surface of the erythrocyte membranes. The remaining negatively charged polysaccharides did not significantly affect the microviscosity of the membranes, apparently due to the processes of electrostatic repulsion from red blood cell cells. The totality of the data obtained has expanded our knowledge about the mechanisms of the possible effect of high molecular weight polysaccharides on the bioavailability of various drugs when creating drugs with controlled absorption.

Simple emulsions (o/w) stabilized by octenyl succinic anhydrous (OSA) starch were studied. They were characterized by the evaluation of the rheological and physicochemical properties and, the observation under the light microscope in combination with granulometric analysis after formulation. The obtained results demonstrated the effects of the amount of OSA starch on the variation of the diameter of particles, stability of emulsions and their rheological behavior. The best characteristics were found in the emulsion with a ratio 40/60 (o/w) at high OSA starch concentration, because the substitution of the oil phase by the network created by the chains of the modified polysaccharides which could be the major factor in stabilisation of emulsions.

The main applications of ionic liquids in chemistry and material research on one of the most important natural polymers—starch—are presented in this review. A brief characterization of ionic liquids and the advantages and disadvantages of using them in the modification and processing of polysaccharides is presented. The latest reports on the use of various ionic liquids as solvents or co-solvents; as media for synthesizing starch derivatives in oxidation, etherification, esterification, and transesterification, with particular emphasis on biocatalyzed reactions; and as plasticizers or compatibilizers in the processing of starch-based polymers have been investigated. The current trends, possibilities, and limitations of using this type of compound for the production of functional starch-based materials are presented.

Gold nanoparticles (AuNPs) are the most thoroughly studied nanoparticles because of their remarkable optical properties. Color changes in assays that use AuNPs can be easily observed with the naked eye, resulting in sensitive colorimetric methods, useful for detecting a variety of biological molecules. However, while AuNPs represent an excellent nano-platform for developing analytical methods for biosensing, there are still challenges that must be overcome before colloidal AuNPs formulation can be successfully translated into practical applications. One of those challenges is the ability to immobilize AuNPs in a solid support. There are many difficulties with controlling both the cluster size and the adhesion of the coatings formed. In addition, many of the techniques employed are expensive and time-consuming, or require special equipment. Thus, a simple and inexpensive method that only requires common lab equipment for immobilizing AuNPs on a surface using Starch Hydrogels has been developed. Starch hydrogels confer a 400% increase in stability to the nanoparticles when exposed to changes in the environment while also allowing for macromolecules to interact with the AuNPs surface. Several starch derivatives were tested, including, dextrin, beta-cyclodextrin and maltodextrin, being dextrin the one that conferred the highest stability. As a proof-of-concept, a SlipChip microfluidic sensor scheme was developed to measure the concentration of DNA in a sample. The detection limit of our biosensor was found to be 25 ng/mL and 75 ng/mL for instrument and naked eye detection, respectively.
