Plant response to osmotic stress is a complex issue and includes a wide range of physiological and biochemical processes. Extensive studies of known cultivars and their reaction to drought or salinity stress are very important for future breeding of new and tolerant cultivars. Our study focused on the antioxidant activity, accumulations of osmotica, and the content of abscisic acid in apple (cv. “Malinové holovouské”, “Fragrance”, “Rubinstep”, “Idared”, “Car Alexander”) and cherry (cv. “Regina”, “Napoleonova”, “Kaštánka”, “Sunburst”, “P-HL-C”) cultivated in vitro on media containing different levels of polyethylene glycol PEG-6000. Our results indicated that the studied genotypes responded differently to osmotic stress manifested as reduction in the leaf relative water content (RWC) and increment in the activities of antioxidant enzymes, proline, sugars, and abscisic acid content. Overall, cherry cultivars showed a smaller decrease in percentage RWC and enzymatic activities, but enhanced proline content compared to the apple plants cultivars. Cultivars “Rubinstep”, “Napoleonova”, and “Kaštánka” exhibited higher antioxidant capacity and accumulation of osmoprotectants like proline and sorbitol that can be associated with the drought-tolerance system.

For the Mediterranean region, climate models predict an acceleration of desertification processes, thus threatening agriculture. The present work aimed to investigate the effect of drought and salinity on Sulla coronaria (L.) Medik., a Mediterranean forage legume, for understanding plant defence systems activated by these stressors. In detail, we focused our attention on the variations on the plant redox status. Plants were subjected to suboptimal watering and irrigation with sodium chloride (NaCl) solutions. The same salt treatment was applied for in vitro tests on seedlings. Water content did not change after treatments. Salt negatively influenced seed germination and seedling development, but it did not affect photosynthesis parameters, contrary to what was observed in adult plants. Proline concentration increased in all samples, while abscisic acid level increased exclusively in seedlings. NaCl caused accumulation of superoxide anion in plants and seedlings and hydrogen peroxide only in seedlings; nevertheless, lipid peroxidation was not detected. Total phenolics, glutathione, expression level, and activity of antioxidant enzymes were assayed, revealing a complex antiradical molecular response, depending on the type of stress and development stage. Our results confirm Sulla as a drought- and salt-tolerant species and highlight its ability to counteract oxidative stress. This evidence suggests a key role for the redox components, as signal transduction messengers, in Sulla acclimation to desertification. Finally, plants and seedlings showed different acclimation capacity to salinity, revealing a greater genomic plasticity for seedlings.

The aim of this study was to investigate the accumulation of silicon in oilseed rape and to characterize the changes in chosen water balance parameters in response to drought. The following parameters were estimated: water content, osmotic and water potential, evapotranspiration, stomatal conductance and abscisic acid level under optimal and drought conditions. It was shown that oilseed rape plants accumulate silicon after its supplementation to the soil, both in the case of silicon alone and silicon together with iron. It was revealed that silicon (without iron) helps maintain constant water content under optimal conditions. While no silicon influence on osmotic regulation was observed, a transpiration decrease was detected under optimal conditions after silicon application. Under drought, a reduction in stomatal conductance was observed, but it was similar for all plants. The decrease in leaf water content under drought was accompanied by a significant increase in abscisic acid content in leaves of control plants and those treated with silicon together with iron. To sum up, under certain conditions, silicon is accumulated even in non-accumulator species, such as oilseed rape, and presumably improves water uptake under drought stress.

Mango is a commercial fruit crop in different parts of the tropical and subtropical world. Commercially important monoembryonic varieties are propagated through grafting onto rootstock seedlings of polyembryonic genotypes that plays an important role in sustained growth and production. Use of salt tolerant genotypes as rootstock to combat the adverse effect of salinity could be helpful for commercial mango production in salt affected areas. Current study was carried out to elucidate the effect of salinity stress induced by NaCl + CaCl2 (1:1 w/w) at 0, 25, 50 and 100 mM concentrations in irrigation water on candidate polyembryonic mango genotypes namely EC-95862, Bappakkai, Vellaikolamban, Nekkare, Turpentine, Muvandan, Kurukkan, Kensington, Olour, Manipur, Deorakhio, Vattam, Mylepelian, Sabre and Kitchener. We studied the morpho-physiological changes of these seedlings under salinity induced stress for determining their relative tolerance by assessing growth parameters such as plant height, number of leaves, leaf area, inter-nodal length, fresh weight of shoot, fresh weight of root, dry weight of shoot, dry weight of root, stem diameter and physiological parameters like photosynthetic rate, transpiration rate, stomatal conductance, number of stomata and stomata length and width, in addition to ABA content in leaves. Our results clarifies that the polyembryonic genotypes Turpentine, Deorakhio Olour and Bappakkai showed less reduction in terms of growth and better maintenance of gas exchange status under higher level of salinity.

In this work, specific concentrations of cadmium, copper and zinc in double combination, were supplied for 12 days to growing seedlings of the model species <em>Arabidopsis</em> <em>thaliana</em>. Metal accumulation was measured in roots and shoots. Microscopic analyses revealed that root morphology was affected by metals, and that the root and shoot levels of indole-3-acetic acid, <em>trans</em>-zeatin riboside and dihydrozeatin riboside varied accordingly. Minor modifications in gibberellic acid levels occurred in the Zinc treatments, whereas abscisic acid level did not change after the exposition to metals. Reverse transcription polymerase chain reaction analysis of some genes involved in auxin and cytokinin synthesis (<em>AtAAO</em>, <em>AtNIT</em> and <em>AtIPT</em>) revealed that their expression were not affected by metal treatments. The root morphological alterations that resulted in an increased surface area, due to the formation of root hairs and lateral roots, could be signs of the response to metal stress in terms of a functionally-addressed reorientation of root growth. The root system plasticity observed could be important for better understanding the manner in which the root architecture is shaped by environmental and hormonal stimuli.
