The Lower Silesian Coal Basin in south-western Poland was a region of intensive coal mining until the late 1990s. After mining was stopped and the mines were closed, the underground workings were flooded. This created an underground reservoir of waters that can be economically managed to supply energy, using heat pumps, to the town of Nowa Ruda. The article analyses the energy potential of these waters. It was shown that open loop with reinjection system based on mine shafts can provide 10 GWh of geothermal energy while reducing emissions from 4.27 tCO2 to 0.22 tCO2. In addition, 2.09 GWh can be extracted from water flowing out of mine shafts via adits. As a final result, this will lead to improve air quality as well as increase the environmental value of the region. These results can act as a reference tool for local government, specialists in energy policy at the local scale. They are also the basis for future work aimed at acquiring external funds to carry out detailed studies of the condition of mine shafts, geothermal profiling of water in sunken shafts and test pumping, which will make it possible to determine the real quantities of water to be used.

Healthy ageing and disease prevention depends on nutritional and lifestyle changes. Available evidence accentuates the influence of genetic, metabolic, and host gut microbiota characteristics on how individuals respond to each nutrient leading the way for the stratification of dietary guidelines. In recent days, state-of-the-art changes are pursued in therapeutic nutrition services integrating information and communication technologies (ICT) such as big data analytics, artificial intelligence (AI), and deep learning. Furthermore, we live in the ‘omics' era, where individuals' dietary needs can be tailored according to their gene pattern. Digital twin typifies virtual representation that provides real-time digital technology counterpart of a physical process or object. This chapter discusses how to converge genetic information with other biochemical parameters to conceptualize a digital replica of oneself that may well be a reference tool to guide personalized nutrition.

Life is a set of relationships in which countless factors interact and go on together. Each of the interacting factors is handled and used as a different discipline when the time comes. Depending on the spirit of the times and the structure of societies, the importance of some discipline areas may change and their place in the agenda may decline. However, this does not apply to mathematics. Because mathematics has served as a constant reference tool for the development of all other scientific fields. Activities that are put forward by ignoring this relationship of mathematics with other disciplines, turn out to be a waste of time and energy. This situation is also valid for educational activities. When mathematics is isolated from other disciplines, ambiguity of meaning and purpose emerges, loses its importance, and becomes a source of concern. The resulting confusion causes mathematics to be perceived as an academic obstacle to be overcome, rather than as a useful tool. It is in the hands of teachers to eliminate this perception and to teach mathematics as the common language of all disciplines. To achieve this, understanding must be changed, and mathematics must be taught based on an interdisciplinary approach.

The aim of this paper is to present the utility of the Gorazd: An Old Church Digital Hub for scholars working with Old Romanian and Slavonic texts written on the territory of today’ s Romania. The Gorazd Project was realized during the years 2016–2020 and it includes an Old Church Slavonic Card Index and three Old Church Slavonic lexical databases, among which the largest one is represented by the digitized and updated version of the monumental Lexicon linguæ palæoslovenicæ (vol. I–IV, 1958–1997) composed by the Institute of Slavonic Studies of the Czech Academy of Sciences. As the Gorazd Project uses English as meta-language, its application is not limited to narrowly specialized Slavic philologists, but it is also open for scholars of neighbouring fields. The dictionaries within the Gorazd Digital Hub can serve as a reference tool not just for the oldest attested Slavonic vocabulary and its semantics, but also for the biblical concordance of the Slavonic oldest Bible redaction and the oldest attested Old Church Slavonic morphological forms.

In this paper, we updated our 2018 systematic review aimed to identify and compare ad hoc designed frameworks for genetic testing evaluation. Overall, we identified 30 frameworks (29 in the first systematic review and one in the update): they were mainly based on the ACCE model, whereas a minority were adjustments of the more traditional Health Technology Assessment (HTA) approach. After discussing the strengths and weaknesses of the retrieved frameworks, this perspective calls for consensus on the assessment of genetic testing. In line with the recent European recommendations that encouraged the generation of comparable evidence across Member States, we believe that the time has come to align all the ideas that have emerged over the last few decades and find a sustainable and sharable tool for the evaluation of genetic and genomic applications. Therefore, we suggest stopping the evaluation of such technologies using ad hoc strategies–affected by validation, implementation, and adoption issues–and we propose to use a general HTA approach, particularly the European reference tool for the assessment of health technologies, the EUnetHTA HTA core model, that is built on solid theoretical and methodological principles and provides a comprehensive assessment of the technologies value.

Azotobacter vinelandii is a nitrogen-fixing free-living soil microbe that has been studied for decades in relation to biological nitrogen fixation (BNF). It is highly amenable to genetic manipulation, helping to unravel the intricate importance of different proteins involved in the process of BNF, including the biosynthesis of cofactors that are essential to assembling the complex metal cofactors that catalyze the difficult reaction of nitrogen fixation. Additionally, A. vinelandii accomplishes this feat while growing as an obligate aerobe, differentiating it from many of the nitrogen-fixing bacteria that are associated with plant roots. The ability to function in the presence of oxygen makes A. vinelandii suitable for application in various potential biotechnological schemes. In this study, we employed transposon sequencing (Tn-seq) to measure the fitness defects associated with disruptions of various genes under nitrogen-fixing dependent growth, versus growth with extraneously provided urea as a nitrogen source. The results allowed us to probe the importance of more than 3800 genes, revealing that many genes previously believed to be important, can be successfully disrupted without impacting cellular fitness. Importance These results provide insights into the functional redundancy in A. vinelandii , while also providing a direct measure of fitness for specific genes associated with the process of BNF. These results will serve as a valuable reference tool in future studies to uncover the mechanisms that govern this process.

Research on the black soldier fly (Hermetia illucens (L.) Diptera: Stratiomyidae) is steadily increasing, with a focus on its larvae and applications thereof. The egg stage of this species has received less research. This study describes the phases of embryo development for BSF eggs and the potential of delaying embryo development by using a chilling protocol. The study found that when chilling eggs for 24 hours at 10 °C and 80% relative humidity, the development is paused for a full day. This study presents a window of opportunity for embryo ages, approximately 38-58 hours post oviposition, where the hatch success is the least affected (>80% hatching when compared to control). Outside of this window, the hatch success decreases to 70% and lower. The description of embryo development can be used as a reference tool for quality control purposes. The effect of treatments on eggs can be determined by identifying key processes in development. Furthermore, the chilling of embryos and thus pausing of development can be applied in a production setting.
