In order to detect the assembly quality of the combine harvester accurately and effectively, a method for the assembly quality inspection of the combine harvester based on the improved whale algorithm (IWOA) to optimize the least square support vector machine is proposed. Aiming at the characteristics of whale optimization algorithm’s weak search ability and easy maturity, this paper introduces the cosine control factor and the sine time-varying adaptive weight to improve it and uses the benchmark function to verify the general adaptability of the algorithm. Combined with the local mean decomposition (LMD), the assembly quality inspection model of the combine harvester was established and applied to the Dongfanghong 4LZ-9A2 combine harvester for experimental verification. The experimental results show that the IWOA proposed in this paper has better optimization ability and adaptability. The average accuracy of the IWOA model proposed in this paper reaches 90.5%, which is 4% higher than that of the WOA model, and the standard deviation of the average accuracy is reduced by 0.15%, which indicates that the IWOA model has better stability.

Endoplasmic reticulum (ER) degradation-enhancing α-mannosidase-like protein 1 (EDEM1) is a quality control factor directly involved in the endoplasmic reticulum-associated degradation (ERAD) process. It recognizes terminally misfolded proteins and directs them to retrotranslocation which is followed by proteasomal degradation in the cytosol. The amyloid-β precursor protein (APP) is synthesized and N-glycosylated in the ER and transported to the Golgi for maturation before being delivered to the cell surface. The amyloidogenic cleavage pathway of APP leads to production of amyloid-β (Aβ), deposited in the brains of Alzheimer’s disease (AD) patients. Here, using biochemical methods applied to human embryonic kidney, HEK293, and SH-SY5Y neuroblastoma cells, we show that EDEM1 is an important regulatory factor involved in APP metabolism. We find that APP cellular levels are significantly reduced after EDEM1 overproduction and are increased in cells with downregulated EDEM1. We also report on EDEM1-dependent transport of APP from the ER to the cytosol that leads to proteasomal degradation of APP. EDEM1 directly interacts with APP. Furthermore, overproduction of EDEM1 results in decreased Aβ40 and Aβ42 secretion. These findings indicate that EDEM1 is a novel regulator of APP metabolism through ERAD.

The differentially expressed genes (DEGs), key genes and metabolic pathways of the parasitic beetle, Dastarcus helophoroides (Fairmaire), were compared between the fiftieth commercially reared population and the first natural population to reveal the adaptive mechanism in response to high temperature stress. The high-throughput sequencing technique was employed for transcriptome sequencing of two populations of D. helophoroides. In total, 47,763 non-redundant transcripts with the average length of 989.31 bp and the N50 of 1607 bp were obtained. Under high temperature stress, 1108 DEGs were found in the commercial population; while there were 3946 DEGs in the natural population, which were higher than those in the commercial population (3.56 times). High temperature stress of D. helophoroides promoted the expression of heat shock proteins (HSPs) and metabolism-related genes in both populations, but metabolism synthesis and hydrolysis of natural population was much higher, allowing them to produce more resistant substances (such as HSPs, superoxide dismutase (SOD), peroxiredoxin (Prx), etc.). Therefore, HSPs may play a major role in the high temperature adaptation of a commercial population, while the natural population probably respond to heat stress with more resistant substances (such as HSPs, SOD, Prx, etc.). These results provide a reference to select and domesticate a specific ecotype with stronger adaptability to the high temperature weather in the forest and further improve the efficiency of D. helophoroides as a bio-control factor.

This study explores the relationship between planned behaviour, i.e. attitude towards behaviour, subjective norm and perceived behavioural control and coherence towards intention to enrich the literature - Vietnamese students studying abroad at university. In addition, the study also explores the factors related to family cohesion to the intention of Vietnamese students to study abroad. This study was conducted through a cross-sectional survey using a targeted sampling technique (n=350). The SEM model was used to test the hypotheses. The SEM model proved that: The decisive factor affects the intention to study abroad with the coefficient of Regression Weight = 0.222. The subjective norm factor (norm) affects the intent variable with the coefficient of Regression Weight = 0.138; The behavioural intention control factor affects the intent factor with Regression Weights = 0.276; The factor through family cohesion (cohesion) affects the intent factor with Regression Weights = 0.159. The conclusions of this study provide valuable data for study abroad managers, universities, study abroad consulting service providers and families who want to invest in their children to study abroad.

Filling mining plays an important role in controlling surface subsidence. To study the movement of overburdened rock in filling mining under thick loose layers, a numerical simulation combing field measurement in CT30101 working face in the Mahuangliang coal mine was tested. The results show that different filling rates and filling body strength have different influences on roof and surface movement. The filling rate has a greater impact, which is the main control factor. The filling stress and roof tensile stress decrease gradually with roadway filling. The filling body stress and roof tensile stress in the first and second rounds are far greater than in the fourth round. After the completion of filling, the first and second round of filling bodies mainly bear the overburden, and the total deformation of the surrounding rock of the main transport roadway is very small, and therefore the displacement of the overburdened rock is controllable. The field monitoring results also show that the overburdened rock became stable after several fillings rounds. Combing the numerical modeling and field tests results, this study can be a guideline for similar geological conditions especially for coal mining under thick loose layers and thin bedrock.

ABSTRACTThe heterotrimeric BAG6 complex coordinates the direct handover of newly synthesised tail-anchored (TA) membrane proteins from an SGTA-bound preloading complex to the endoplasmic reticulum (ER) delivery component TRC40. In contrast, defective precursors, including aberrant TA proteins, form a stable complex with this cytosolic protein quality control factor, enabling such clients to be either productively re-routed or selectively degraded. We identify the mitochondrial TA protein MAVS (mitochondrial antiviral-signalling protein) as an endogenous client of both SGTA and the BAG6 complex. Our data suggest that the BAG6 complex binds to a cytosolic pool of MAVS before its misinsertion into the ER membrane, from where it can subsequently be removed via ATP13A1-mediated dislocation. This BAG6- associated fraction of MAVS is dynamic and responds to the activation of an innate immune response, suggesting that BAG6 may modulate the pool of MAVS that is available for coordinating the cellular response to viral infection.SUMMARY STATEMENTMitochondrial antiviral-signalling (MAVS) protein is a favoured client of the cytosolic BAG6 complex. We discuss how this dynamic interaction may modulate MAVS biogenesis at signalling membranes.

We report a molecular switching ensemble whose states may be regulated in synergistic fashion by both protonation and photoirradiation. This allows hierarchical control in both a kinetic and thermodynamic sense. These pseudorotaxane-based molecular devices exploit the so-called Texas-sized molecular box (cyclo[2]-(2,6-di(1H-imidazol-1-yl)pyridine)[2](1,4-dimethylenebenzene); 14+, studied as its tetrakis-PF6− salt) as the wheel component. Anions of azobenzene-4,4′-dicarboxylic acid (2H+•2) or 4,4′-stilbenedicarboxylic acid (2H+•3) serve as the threading rod elements. The various forms of 2 and 3 (neutral, monoprotonated, and diprotonated) interact differently with 14+, as do the photoinduced cis or trans forms of these classic photoactive guests. The net result is a multimodal molecular switch that can be regulated in synergistic fashion through protonation/deprotonation and photoirradiation. The degree of guest protonation is the dominating control factor, with light acting as a secondary regulatory stimulus. The present dual input strategy provides a complement to more traditional orthogonal stimulus-based approaches to molecular switching and allows for the creation of nonbinary stimulus-responsive functional materials.
