Monitoring and health risk of mycotoxins in imported wines and beers consumed in Thailand

The presence of mycotoxins has been reported in a wide range of agricultural commodities including wine and beer. Due to the widespread consumption of these beverages, risk assessment of contamination by toxins is required for consumer health. In the present study, multi-residue analysis for the determination of mycotoxins was undertaken to survey the incidence of mycotoxins in imported wines and beers being commercialised in Thailand, and to assess the consumer health risk. Samples of red wines (100) and beers (100) were collected randomly from supermarkets and retail shops in Bangkok, Thailand and were extracted using the dispersive liquid-liquid micro-extraction procedure. The quantitation and confirmation of 19 mycotoxins were performed using liquid chromatography tandem-mass spectrometry with an electro-spray ionisation interface in the multiple-reaction monitoring mode. The results demonstrated that the wine and beer samples were contaminated with mycotoxins at 35 and 13%, respectively. The most prominent mycotoxins found in wines were Alternaria toxins, ochratoxin A, and fumonisins, whereas zearalenone and deoxynivalenol were the most prevalent mycotoxins found in contaminated beers. However, ochratoxin A levels were far below the maximum limits established by the European Union. As indicated by the risk assessment, mycotoxin exposure via imported wines and beers were at safe levels in the Bangkok urban area.

Abstract A reliable and rapid method was developed to determine benzobicyclon residue in different soil and sediment samples. After extraction via a modified quick, easy, cheap, effective, rugged, and safe method, samples were purified by SPE cleanup with HLB cartridges. Quantitative determination was performed by ultra-HPLC (UPLC)-tandem MS (MS/MS) in electrospray positive ionization and multiple reaction monitoring modes. When samples were fortified at concentrations of 5, 50, and 500 µg/L, recoveries of 80.2 to 114.5% were obtained, with the repeatability (intraday RSDr) and reproducibility (interday RSDR) <14.1 and <21.4%, respectively. The instrumental LODs and LOQs for matrix-matched standards and the method LOQs for sample test were 0.19–1.34 μg/L, 0.64–4.48 μg/L, and 0.32–2.24 μg/kg, respectively. The linear range was 5–1000 μg/L (R2 > 0.99). The established UPLC-MS/MS method was applied in the detection of benzobicyclon in real soil samples, which were collected during the supervised field trial. Results showed that the maximum concentration of benzobicyclon in the soil was 4.87 mg/kg and its degradation half-life (t0.5) was 6.7 days. Generally, the proposed method could be an effective tool for controlling and monitoring the risks posed by benzobicyclon to human health and environmental safety.

Oxylipins play important roles in various biological processes and are considered as mediators of inflammation for a wide range of diseases such as rheumatoid arthritis (RA). The purpose of this research was to study differences in oxylipin levels between a widely used collagen induced arthritis (CIA) mice model and healthy control (Ctrl) mice. DBA/1J male mice (age: 6-7 weeks) were selected and randomly divided into two groups, namely, a CIA and a Ctrl group. The CIA mice were injected intraperitoneally (i.p.) with the joint cartilage component collagen type II (CII) and an adjuvant injection of lipopolysaccharide (LPS). Oxylipin metabolites were extracted from plasma for each individual sample using solid phase extraction (SPE) and were detected with high performance liquid chromatography/tandem mass spectrometry (HPLC-ESI-MS/MS), using dynamic multiple reaction monitoring (dMRM). Both univariate and multivariate statistical analyses were applied. The results in univariate Student’st-test revealed 10 significantly up- or downregulated oxylipins in CIA mice, which were supplemented by another 6 additional oxylipins, contributing to group clustering upon multivariate analysis. The dysregulation of these oxylipins revealed the presence of ROS-generated oxylipins and an increase of inflammation in CIA mice. The results also suggested that the collagen induced arthritis might associate with dysregulation of apoptosis, possibly inhibited by activated NF-κB because of insufficient PPAR-γligands.

AbstractIntroduction: In the European Union, the use of thyreostatic drugs for fattening slaughter animals has been banned since 1981 under Council Directive 81/602/EEC. For protection of consumer health against unwanted residues and in compliance with Directive 96/23, each EU country must monitor thyreostats in samples of animal origin. This paper presents the results of research on thyreostatic residues carried out in Poland in 2011–2017.Material and Methods: The material for testing was urine (n = 3,491), drinking water (n = 127), and muscle samples (n = 349) officially collected by Veterinary Sanitary Inspectors in slaughterhouses and farms throughout the country in accordance with the national residue control plan. The samples were examined for the presence of tapazole, thiouracil, methylthiouracil, propylthiouracil, and phenylthiouracil using liquid chromatography tandem mass spectrometry through an accredited method.Results: In four bovine and three porcine urine samples, the permissible thiouracil concentration was exceeded. In one sample of porcine urine, methyl- and propylthiouracil were found. The presence of thiouracil and its derivatives in urine samples is most likely due to feeding animals diet containing cruciferous plants.Conclusions: The results of research indicate that thyreostats are not used for anabolic purposes in slaughter animals in Poland.

Low fertility in rice caused by Chilo suppressalis has led to the use of diazinon to control this pest. Residue of pesticide could penetrate products and also food which can affect public health. The aim of this research was to determine health risk assessment of organophosphorus (OP) pesticide in rice, a strategic crop in Iran. Ninety rice samples were collected from 30 points during harvesting seasons from Rasht Area, Guilan Province, Iran from which 30 samples were prepared. The concentration of diazinon, the most common pesticide used in the study area, was determined by high-performance liquid chromatography. The result indicated that the total average of diazinon in rice samples (31.91 mg/kg) is by far higher than the maximum residue limit recommended by the European Union. According to the results, EDAI was 0.051 mg/kg day, while health risk index in rice was 10.2. Results showed that there is a health risk associated with the lifetime consumption of rice polluted by OP pesticide in the study area.

Abstract Groundwater samples of Ile-Ife, Osun State, Nigeria were investigated for their organochlorine pesticides (OCPs) levels. Probable sources of the OCPs and health risks associated with their consumption along with the water were determined in order to establish the potability of the groundwater samples. Quantitative determination of the OCPs was carried out by Gas Chromatography coupled with Electron Capture Detector (GC-ECD) after liquid-liquid extraction with dichloromethane (DCM). Results indicated that all the analyzed OCPs except p,p'-dichlorodiphenylethane were detected with high concentrations in the groundwater. Heptachlor (14.60±3.60 µg L-1) and methoxychlor (12.60±2.20 µg L-1) showed dominant concentrations that were higher than 0.02 ng L-1 maximum residue levels (MRLs) recommended by the European Union. Levels of the OCPs in the samples followed the decreasing trend: cyclodienes > diclorophehylethanes > chlorinated cyclohexanes, while the predominant source of the analyzed pesticides could be ascribed to aerial transportation from fresh applications in homes within the community. The carcinogenic health risk assessment also revealed consistent higher values of HQ and CR in children as opposed to adults, indicating that children are the more vulnerable population to the analyzed environmental contaminants.

Background: Despite huge advances in science, technology, and medical equipment, numerous concerns grow over healthcare workers (HCWs) since they are exposed to a wide range of biological hazards due to the nature of their work. Objectives: The current study aimed at developing and validating an index to assess the risk of occupational safety and health (OSH) in hospitals and healthcare settings. Methods: In the current cross-sectional study, an index called the hospital occupational safety, and health risk assessment (HOSHRA) was developed and validated through the face and content validity as well as internal consistency and inter-rater reliability. Ultimately, the developed index was utilized to assess OSH risks in 36 teaching hospitals affiliated to Shiraz University of Medical Sciences, Shiraz, Iran. Results: The mean score of the HOSHRA index among the selected hospitals was 71.53 ± 5.93, indicating that the OSH risks were at a moderate level. According to the HOSHRA action levels, OSH risk level for ergonomic hazards was high, but it was moderate for physical, chemical, ergonomic, and psychological ones. In terms of physical hazard subcategories, the risk of electrical hazard, as well as fire and explosion, was high, but it was moderate for fall and slip, and radiation. Conclusions: The developed observation-based method showed an acceptable content validity and reliability for OSH risk assessment of hospitals. The HOSHRA index could also be used for hospitals as an applicable measure to improve their OSH.
