Abstract Cellulolytic bacteria is one of the beneficial bacteria that can found in mangrove ecosystem. The purposes of study were to analyse the cellulolytic index, and to analyse the cellulase activity of bacteria isolated from soil mangrove. Qualitatively, assessment of cellulase activity were carried out at the Microbiology laboratory of Fish Quarantine Station, Quality Control and Safety of Fishery Products (SKIPM) Aceh, while quantitatively was observed in microbiology laboratory, Biology Department, IPB. Assessment of qualitative cellulase activity is performed by growing the selected pure isolate on 1% CMC medium then spilled 1% congo red to test its cellulolytic potential. Cellulolytic potential was determined by clear zone performed around the colony after congo red flooded. The quantitative cellulase enzyme activity test carried out by DNS method tested on one selective isolate. There were 21 from 39 isolates showed a clear zone isolated from mangrove soil. The cellulolytic index (CI) obtained ranged from 0.07 to 0.80 classified as low cellulolytic index criteria. The cellulolytic index was higher in bacteria isolated from mangrove rehabilitated than mangrove unrehabilitated. The highest cellulase activity and specific cellulase activity of BTMD32 was at 48 hours with the value were 0.0012 U/ml, 0.077 U/mg, respectively. The result concluded that the bacteria cellulolytic isolated from mangrove soil had low cellulolytic index, low cellulase activity, and low specific cellulase activity.

ABSTRACTPackaged beverages have become a product that is very much on the market both internationally and nationally. In the process of making packaged beverages, in order to last longer the microbes in the drink must first be inactivated in order for the drink to last longer, the microbial inactivation process in the drink is called pasteurization. Convernsional pasteurization is done by heating the drink to be in production. But the high temperature of the drink can change the taste, color, and even nutrition of the drink. This is underpinning the non-thermal pasteurization process by using high voltage impulses (Pulsed Electric Field,PEF). PEF is a technology that utilizes high voltage DC to produce a field between two electrodes. Pateurization occurs when between electrodes there is milk that is flowed with high voltage DC. High voltage generation is performed using flyback transformers with voltage output up to 30 kV. In the pasteurization process given 6 voltage variations namely 5, 10, 12,5, 15, 20, and 30 kV. Pasteurization results are then tested in fmipa microbiology laboratory to find out the success of the tools that have been made. In each voltage variation it is obtained that all treatments have reached pasteurized milk quality standards based on SNI where at 5kV voltage variation can inactivat microbes up to 83.33%, voltage variation of 10kV can inactivat microbes up to 94.5%, At a voltage variation of 12.5 kV can inactivae microbes up to 83.6%, 15kV voltage variation can inactivae microbes up to 88.8%, while voltage variation of 20 kV can inactivae microbes up to 84.2% and at voltage variation 30 kV can inactivae microbes up to 84.8%.Key Words: PEF, Flyback Transformer, ZVS Driver, Pasteurization.

Cellulolytic bacteria that produce cellulase enzymes play an essential role in degrading cellulose in their habitat. The presence of cellulolytic bacteria strongly supports the fertility and productivity in mangrove waters. The objectives of the study are to analyze the activity of cellulase enzyme qualitatively through the cellulolytic index and quantitatively through the activity and specific activity of the cellulase enzyme from bacteria isolated from the water of mangrove ecosystems in Aceh Province. The qualitative experiment of enzyme activity was carried out at the Microbiology laboratory SKIPM Aceh, and a quantitative experiment of enzyme activity was conducted at the Microbiology Laboratory, Biology Department, IPB. Isolation of cellulolytic bacteria isolated from mangrove water used Carboxy Methyl Cellulose (1% CMC) selective media and carried out by spread plate method. The ability of bacteria to produce cellulase was tested qualitatively using the spot technique, this test was carried out using 1% Congo Red. Furthermore, the quantitative testing of cellulase enzymes activity adopted the DNS spectrophotometric method. The specific activity of the cellulase enzyme can be determined by using the Lowry method. There were 21 isolates that had a clear zone and had the ability to produce cellulase enzymes from 49 isolates that were successfully purified. The highest cellulolytic index (CI) produced using BAM421 isolate with the value of 5.50 was included in the high category, followed by BAM326 and BAM132 isolates, with values of 1.55 and 1.05 were categorized into the medium category. The other isolates were in the low cellulolytic index category. The isolate with the highest CI value was further tested using the quantitative enzyme activity test. The highest cellulase enzyme activity of BAM421 occurred at 24hr (0.0029 U/ml). The highest specific cellulase activity of BAM421 was at 24hr with the value of 0.210 U/mg. The result concluded that the qualitative test showed CI values can be categorized into low, medium, and high. Soil Transmitted Helminth (STH) is the most common intestinal parasific infection and in one of the contributors to the global disease burden. Diagnosis of Soil Transmitted Helminth desease can be done using sedimentation method and natif method. This study aims to find out the differences in sedimentation methods and natif methods in deteting Soil Transmitted Helminth Type of research conducted ind true experimental form. This research was conducted on March 27-29, 2021 at the Microbiology Laboratory of Muhammadiyah Institute of Health and Technology Palembang which numbered 35 respondents. The results obtained in this study were as many as 2 positive samples and 33 negative samples. In sample 1 there are 2 types of eggs Soil Transmitted Helminth namely eggs Ascaris lumbricoides and Trichuris trichiura on sedimentation method and natif method, while in sample 5 there are eggs Ascaris lumbricoides only on the natif method. The results of the research data were conduted using an alternative test Wilcoxon with a value of P = 0.317 means there is no difference that signifikan.the conclusion of this study was obtained by the examination that there is no difference in sedimentation with natiif method.

Background: The incidence of Dengue hemorrhagic fever, Dengue shock syndrome associated with Dengue can be reduced by diagnosing Dengue early and by initiating early treatment to Dengue patients. This study was conducted to compare results of NS1 antigen rapid test and ELISA in clinically suspected dengue patients. Materials and Methods: Present study was a comparative study conducted on 100 Patients presented with clinical history of Dengue. At Microbiology Laboratory, serum of all samples was assessed for NS1 detection using antigen Rapid test and ELISA.  Sensitivity & specificity values were calculated for NS1 antigen rapid test, compared with ELISA. Results: Out of 100 serum samples collected from suspected cases of Dengue in and around Anantapuramu, 30 (30%) were positive for ELISA and 28 (28%) were positive for Rapid diagnostic test. Sensitivity & specificity when only NS1 was considered on rapid test kits when compared with ELISA were 93.33%, 98.57%, Conclusion: It is concluded that ELISA test was superior in the diagnosis of Dengue and recommended on improvement in sensitivity of RDTs.

Caisim (Brassica chinensis) is one of the vegetables that has many benefits such as protein, carbohydrates, fat and plays an important role in meeting food needs, and is good for health. Caisim is well adapted to high temperatures and the price is relatively stable and easy to cultivate. The purpose of this study was to determine the presence of Soil-Transmitted Helminths eggs in caisim (B. chinensis). The research was conducted in a descriptive observational. The research was conducted in July-August 2020 at Parasitology and Microbiology Laboratory, Faculty of Medicine, Universitas Muhammadiyah Purwokerto. Identification of Soil-Transmitted Helminths eggs from 20 caisim as the samples used NaCl flotation method and were analyzed by using a univariate test with tabulated frequency distribution. The results showed that no Soil-Transmitted Helminths eggs were found in caisim (B. chinensis).   Caisim (Brassica chinensis) merupakan sayuran yang mempunyai banyak kandungan manfaat seperti protein,karbohidrat, lemak dan berperan penting untuk memenuhi kebutuhan pangan serta baik untuk kesehatan. Caisimmampu beradaptasi dengan baik terhadap suhu tinggi serta harga relatif stabil dan mudah diusahakan. Tujuanpenelitian ini untuk mengetahui adanya telur Soil Transmitted Helminths terdapat pada caisim (B. chinensis).Penelitian dilakukan secara observasional deskriptif. Penelitian dilakukan pada bulan Juli-Agustus 2020 diLaboratorium Parasitologi Fakultas Kedokteran Universitas Muhammadiyah Purwokwerto. Identifikasi telur SoilTransmitted Helminthsdari 20 sampel caisim dengan metode Flotasi NaCl. dianalisis dengan uji univariat dengandistribusi tabulasi frekuensi. Hasil penelitian menunjukan tidak ditemukan telur Soil Transmitted Helminths padacaisim (B. chinensis)

BackgroundTotal laboratory automation (TLA) has the potential to reduce specimen processing time, optimize workflow, and decrease turnaround time (TAT). The purpose of this research is to investigate whether the TAT of our laboratory has changed since the adoption of TLA, as well as to optimize laboratory workflow, improve laboratory testing efficiency, and provide better services of clinical diagnosis and treatment.Materials and MethodsLaboratory data was extracted from our laboratory information system in two 6-month periods: pre-TLA (July to December 2019) and post-TLA (July to December 2020), respectively.ResultsThe median TAT for positive cultures decreased significantly from pre-TLA to post-TLA (65.93 vs 63.53, P<0.001). For different types of cultures, The TAT of CSF changed the most (86.76 vs 64.30, P=0.007), followed by sputum (64.38 vs 61.41, P<0.001), urine (52.10 vs 49,57, P<0.001), blood (68.49 vs 66.60, P<0.001). For Ascites and Pleural fluid, there was no significant difference (P>0.05). Further analysis found that the incidence of broth growth only for pre-TLA was 12.4% (14/133), while for post-TLA, it was 3.4% (4/119). The difference was statistically significant (P=0.01). The common isolates from CSF samples were Cryptococcus neoformans, coagulase-negative Staphylococcus, Acinetobacter baumannii, and Klebsiella pneumonia.ConclusionUsing TLA and setting up three shifts shortened the TAT of our clinical microbiology laboratory, especially for CSF samples.
