Organoleptic and biochemical attributes in the coffee bean determine the final cup quality of coffee which is a critical factor in the price determination of coffee in the market. The study aimed at determining the genetic variability of the green coffee bean. The trial sites were located at Siaya and Busia counties in Kenya. Nineteen different genotypes were established and included Arabusta coffee hybrids, backcrosses of Arabica to tetraploid Robusta, Arabica coffee, Robusta coffee, and Arabusta coffee. Randomized Complete Block Design with three replications in each site was used in conducting the experiment. The coffee beans were harvested in the year 2018 and extraction and calculation of sucrose, trigonelline, caffeine, and chlorogenic acids was carried using the recommended methods. The cupping procedure involved the use of five judges in assessing the flavor, aroma, balance, overall standard, acidity, body, and aftertaste of the roasted coffee beans. The sensory evaluation used the Specialty Coffee Association (SCA) method. There were significant variations recorded for the traits that were measured. All the traits were highly heritable registering values of > 50% for heritability whereby, caffeine and oil were highly heritable traits with 90.8% and 88.9% respectively. Oil had a high phenotypic coefficient of variation, genotypic variation, and response values when compared to the other traits. All the organoleptic traits were positively correlated with sucrose, trigonelline, and oil but the correlation with caffeine and chlorogenic acids was negative. The genotypic effects contributed largely to the high heritability recorded with a low influence from the environmental factors.

AbstractThe domestication of forest species has traditionally relied on productivity issues. However, today there are concerns about the potential responses of natural populations and new cultivars to extreme climatic conditions derived from climate change and how to incorporate this knowledge into the domestication programs. Aristotelia chilensis (Molina) Stuntz (‘Maqui’) is a widely distributed native species in Chile. Its berry is considered a “super fruit” with an increasing interest in the food industry. This study investigated the phenotypic variation of growth, fruit, and ecophysiological traits of 20 A. chilensis clones originated from six provenances along the latitudinal gradient and established in a common-garden experiment in the Mediterranean zone of central Chile (center part of the species distribution). Differences among provenances were observed for most of the  traits under study, especially between the northern and southernmost provenances (i.e., San Fernando versus Entre Lagos). Northern provenances showed higher development of vegetative tissue and fruit yield but lower intrinsic water use efficiency (WUEint) compared with southern ones. Clonal variation within provenances was found significant for the ripening index, WUEint, and fruit number and weight but not significant for traits related to the crown and leaf morphology. A genetic differentiation due to latitudinal cline was not evident in this study, but differences among provenances suggest local adaptation for some traits. The genotypic variation in productive traits must be considered in the outgoing domestication of the species and future selection programs.

Abstract Rice is a cereal staple of global fame and importance. Oryza barthii, a wild species holds significant traits and its utilization in rice breeding is rare. This study traced introgression trend of heritable traits in the offspring of O. barthii with an Africa-Asian progenitor to F8 and assessed diversity between the parents and the F8 population. Significant (P<0.05) genotypic variation existed for all the traits except tiller number, panicle/meter squared, grains/panicle and 1000 grain weight. Grains/panicle and days to 50% flowering had respective least (3.34%) and highest (96.32%) broad sense heritabilities. All traits had lower GCV compared to PCV. The least (5.28% and 8.05%) and the highest (90.8% and 98.1%) GCV and PCV were respectively from grains/panicle and tiller number. Clear variations on the panicles and grains include: variations in sizes, shapes, colours, presence or absence of awns. The total variance explained by five principal component axes was 80.1%. Plant height at maturity was the only trait with significant (p ≤ 0.01) correlation and regression between F6 and F7. Progenies resemblance to Parent 1(IRGC 104084) retrogressively declined but parent-offspring to parent 2 (TGS 25) progressively increased from F6 to F8. Three visible groups of rice type in this study were: the O. barthii (11%), O. sativa (67%) and the intermediate group (22%). This research has added to rice genetic resources; an investigation of the nutritional status of the progenies would be an interesting research.

The plant-associated microbiome has been shown to vary considerably between species and across environmental gradients. The effects of genomic variation on the microbiome within single species are less clearly understood, with results often confounded by the larger effects of climatic and edaphic variation. In this study, the effect of genomic variation on the rhizosphere bacterial communities of maize was investigated by comparing different genotypes grown within controlled environments. Rhizosphere bacterial communities were profiled by metabarcoding the universal bacterial 16S rRNA v3-v4 region. Initially, plants from the inbred B73 line and the Ancho - More 10 landrace were grown for 12-weeks and compared. The experiment was then repeated with an additional four Mexican landraces (Apachito - Chih 172, Tehua - Chis 204, Serrano - Pueb 180 and Hairnoso de Ocho) that were grown alongside additional B73 and Ancho - More 10 genotypes. In both experiments there were significant genotypic differences in the rhizosphere bacteria. Additionally, the bacterial communities were significantly correlated with genomic distance between genotypes, with the more closely related landraces being more similar in rhizosphere bacterial communities. Despite limited sampling numbers, here we confirm that genomic variation in maize landraces is associated with differences in the rhizosphere bacterial communities. Further studies that go beyond correlations to identify the mechanisms that determine the genotypic variation of the rhizosphere microbiome are required.

Given that an effective combined foliar application of iodine (I), selenium (Se), and zinc (Zn) would be farmer friendly, compared to a separate spray of each micronutrient, for the simultaneous biofortification of grain crops, we compared effectiveness of foliar-applied potassium iodate (KIO3, 0.05%), sodium selenate (Na2SeO4, 0.0024%), and zinc sulfate (ZnSO4∙7H2O, 0.5%), separately and in their combination (as cocktail) for the micronutrient biofortification of four Basmati cultivars of rice (Oryza sativa L.). Foliar-applied, each micronutrient or their cocktail did not affect rice grain yield, but grain yield varied significantly among rice cultivars. Irrespective of foliar treatments, the brown rice of cv. Super Basmati and cv. Kisan Basmati had substantially higher concentration of micronutrients than cv. Basmati-515 and cv. Chenab Basmati. With foliar-applied KIO3, alone or in cocktail, the I concentration in brown rice increased from 12 to 186 µg kg−1. The average I concentration in brown rice with foliar-applied KIO3 or cocktail was 126 μg kg−1 in cv. Basmati-515, 160 μg kg−1 in cv. Chenab Basmati, 153 μg kg−1 in cv. Kisan Basmati, and 306 μg kg−1 in cv. Super Basmati. Selenium concentration in brown rice increased from 54 to 760 µg kg−1, with foliar-applied Na2SeO4 individually and in cocktail, respectively. The inherent Zn concentration in rice cultivars ranged between 14 and 19 mg kg−1 and increased by 5–6 mg Zn per kg grains by foliar application of ZnSO4∙7H2O and cocktail. The results also showed the existence of genotypic variation in response to foliar spray of micronutrients and demonstrated that a foliar-applied cocktail of I, Se, and Zn could be an effective strategy for the simultaneous biofortification of rice grains with these micronutrients to address the hidden hunger problem in human populations.

Potassium (K) is the most abundant cation in plants, playing an important role in osmoregulation. Little is known about the effect of genotypic variation in the tolerance to osmotic stress under different K treatments in barley. In this study, we measured the interactive effects of osmotic stress and K supply on growth and stress responses of two barley cultivars (Hordeum vulgare L.) and monitored reactive oxygen species (ROS) along with enzymatic antioxidant activity and their respective gene expression level. The selected cultivars (cv. Milford and cv. Sahin-91Sahin-91) were exposed to osmotic stress (−0.7 MPa) induced by polyethylene glycol 6000 (PEG) under low (0.04 mM) and adequate (0.8 mM) K levels in the nutrient solution. Leaf samples were collected and analyzed for levels of K, ROS, kinetic activity of antioxidants enzymes and expression levels of respective genes during the stress period. The results showed that optimal K supply under osmotic stress significantly decreases ROS production and adjusts antioxidant activity, leading to the reduction of oxidative stress in the studied plants. The cultivar Milford had a lower ROS level and a better tolerance to stress compared to the cultivar Sahin-91. We conclude that optimized K supply is of great importance in mitigating ROS-related damage induced by osmotic stress, specifically in drought-sensitive barley cultivars.

Somatic embryogenesis is being piloted for the commercial production of genetically improved Norway spruce (Picea abies L. Karst) forest regeneration material in Finland. The main challenge to making the process commercially relevant is the dependence on time-consuming and highly skilled manual labor. Automation and scaling up are needed to improve cost-effectiveness. Moving from the proliferation of embryogenic tissue on semisolid media to suspension cultures could improve process scalability. In a series of four experiments (overall, with 20 cell lines, 4–9 per experiment), the suitability of proliferation in suspension culture for Norway spruce somatic embryogenesis was evaluated based on the growth rate, indicators of stress conditions, good-quality cotyledonary embryo yield, and embling survival in a greenhouse. The proliferation rate in suspension was found equal to on semisolid media, but with a remarkable genotypic variation. Embryogenic tissue matured directly without pre-treatments from suspension onto semisolid media produced lower numbers of good-quality embryos than tissue matured from semisolid media. Rinsing the suspension-grown tissue with hormone-free liquid media before maturation improved embryo yield, bringing it closer to that of semisolid-grown tissue. Decreasing 6-benzylaminopurine and 2,4-dichlorophenoxyacetic acid concentrations in suspension proliferation media to 0.5 or 0.1 times those in semisolid media did not affect tissue growth and did not improve embryo production. The hydrogen peroxide (H2O2) content and guaiacol peroxidase activity were elevated in suspension cultures compared with semisolid medium, which had the same plant growth regulator content. In one experiment out of four, the greenhouse survival of germinants was lower when proliferation was carried out in full strength suspension than on semisolid media; in other experiments the survival rates were equal.

Phenotypic variation and correlations among root traits form the basis for selecting and breeding soybean varieties with efficient access to water and nutrients and better adaptation to abiotic stresses. Therefore, it is important to develop a simple and consistent system to study root traits in soybean. In this study, we adopted the semi-hydroponic system to investigate the variability in root morphological traits of 171 soybean genotypes popularized in the Yangtze and Huaihe River regions, eastern China. Highly diverse phenotypes were observed: shoot height (18.7–86.7 cm per plant with a median of 52.3 cm); total root length (208–1663 cm per plant with a median of 885 cm); and root mass (dry weight) (19.4–251 mg per plant with a median of 124 mg). Both total root length and root mass exhibited significant positive correlation with shoot mass (p ≤ 0.05), indicating their relationship with plant growth and adaptation strategies. The nine selected traits contributed to one of the two principal components (eigenvalues > 1), accounting for 78.9% of the total genotypic variation. Agglomerative hierarchical clustering analysis separated the 171 genotypes into five major groups based on these root traits. Three selected genotypes with contrasting root systems were validated in soil-filled rhizoboxes (1.5 m deep) until maturity. Consistent ranking of the genotypes in some important root traits at various growth stages between the two experiments indicates the reliability of the semi-hydroponic system in phenotyping root trait variability at the early growth stage in soybean germplasms.
