Microbial diversity is the most abundant form of life. Next Generation Sequencing technologies provide the capacity to study complex bacterial communities, in which the depth and the bioinformatic tools can influence the results. In this work we explored two different protocols for bacterial classification and abundance evaluation, using 10 bacterial genomes in a simulated sample at different sequencing. Protocol A consisted of metagenome assembly with Megahit and Ray Meta and taxonomic classification with Kraken2 and Centrifuge. In protocol B only taxonomicclassification. In both protocols, rarefaction, relative abundance and beta diversity were analyzed. In the protocol A, Megahit had a mean contig length of 1,128 and Ray Meta de 8,893 nucleotides. The number of species correctly classified in all depth assays were 6 out of 10 for protocol A, and 9 out of 10 using protocol B. The rarefaction analysis showed an overestimation of the number of species in almost all assays regardless of the protocol, and the beta diversity analysis results indicated significant differences in all comparisons. Protocol A was more efficient for diversity analysis, while protocol B estimated a more precise relative abundance. Our results do not allow us to suggest an optimal sequencing depth at specie level.

A update financial system is a very necessary for the economy growth, insurance industry provides freedom security for the future to all its citizen. In India the government and the insurance industry works together to provide maximum benefit to its people in the form of life and non-life insurance which includes health, travel and vehicle insurance, it is a very old industry, exiting since ancient times in different form, with privatization, we see a rapid rise in insurance industry, where people are saving along with there is domestic saving, FDI has amplified to 74% which has brough in capital income. Farmers, poor and vulnerable people have been benefitted with majority of the health insurance policy in India. we also see a rise in digital insurance. In FY21 the non-insurance recorded 5.19% growth in Growth Direct premiums, where as the market share for general and health insurance has escalated from 47.19% to 48.3% for FY20. In this pandemic the main focus in on health insurance, the government is bringing in new schemes for health sector and incrementing its insurance.

The phenomenon of the development of the local leadership system in the Dayak Salako community in Nyarumkop village, Singkawang, West Kalimantan, cannot be separated from the current era development; of which the existing leadership system in the community requires dynamic aspects to make the traditional leadership system able to adjust its functions and role in the society that continues to develop. This article will analyze and explain how local leadership forms when community groups Dayak Salako still live as a Bantang community until now, which has become a village community that already has its formal and bureaucratic government leadership. Through a qualitative method with an ethnographic approach, key informants from the customary chief and other stakeholders within the community, it turns out that the leadership of the Dayak Salako customary chief has undergone several changes following the form of life of the community. Even though the global modern development impacts the community's way of life, the importance and influence of customary chief are pertinent.

Metabolism management plays an essential role in extracorporeal technologies. There are different metabolic management devices integrated to extracorporeal devices; the most commonly used and accepted metabolic target in adult patients is indexed oxygen delivery (280 mL/min/m2) and cardiac index (2.4 L/min/m2), which can be managed independently or according to other metabolic parameters. Extracorporeal membrane oxygenation (ECMO) is a temporary form of life support providing a prolonged biventricular circulatory and pulmonary support for patients experiencing both pulmonary and cardiac failure unresponsive to conventional therapy. The goal-directed perfusion initiative during cardiopulmonary bypass (CPB) reduced the incidence of acute kidney injury after cardiac surgery. On the basis of the available literature, the identified goals to achieve during CPB include maintenance of oxygen delivery > 300 mL O2/min/m2 and reduction in vasopressor use. ECMO and CPB are conceptually similar but differ in many aspects and finality; in particular, they differ in the scientific evidence for metabolic management nadirs. As for CPB, predictive target parameters have been found and consolidated, particularly in terms of acute renal injury and the prevention of anaerobic metabolism, while for ECMO management, a blurred path remains. In this context, we review the strategies for optimal goal-directed therapy during CPB and ECMO, trying to transfer the knowledge and experience from daily cardiac surgery to veno-arterial ECMO.

The article focuses on the phenomenon of daily reality as an element of public life. The author analyses the principles this notion is based on. Daily reality is a mode that states the authenticity of a man’s being. The premises which have led to the necessity of the notion’s study are defined, the latter being a fundamental rethinking of rationality’s role as a method to figure out where a man’s place in the world is. The author analyses methodology of the problem within the philosophical discourse, and gives a socio-cultural justification of daily reality structures and levels. To formulate the social and philosophical nature of daily reality clearly actual historical and public life context is considered. Potentially this approach helps investigate the practical side of being, which earlier has been considered by philosophers unessential from the standpoint of motives of a man’s life. The author argues that the rationalization of all the spheres of life is not the only way to regulate social being. Processes which go beyond rationality play a very significant role in human activity organization. Daily reality is not a form of passive, irresponsible living, it is viewed as the human mode of authentic being and one of the phases of active, creative, responsible existence constructing.

This study discusses the shifting dynamics of fan participatory cultures on social media platforms by introducing the concept of “platformized language games.” We conceive of a fan community as a “speech community” and propose that the language and discourses of fan participatory cultures are technological practices that only make sense in use and interactions as “games” on social media platform. Based on an ethnography of communication on fan communities on Weibo, we analyze the technological-communicative acts of fan speech communities, including the platformized setting, participants, topics, norms, and key purposes. We argue that the social media logic (programmability, connectivity, popularity, and datafication) articulates with fans’ language games, thus shifting the “form of life” of celebrity fans on social media. Empirically, fan participatory cultures continue to mutate in China, as fan communities create idiosyncratic platformized language games based on the selective appropriation of the social media logics of connectivity and data-driven metrics.

The purpose of the present essay is to exposit and interpret the principal contours of the phenomenology of Christianity proposed by Michel Henry in dialog with his theological critics. Against the claims commonly made about him, Henry is not a Gnostic of any sort: neither a monist, nor a dualist, nor a pantheist, nor a denier of faith, nor a world- or creation-denier or anything of the sort. He rather proposes a form of “life-idealism” according to which (i) life is the foundation of the possibility of the world, (ii) life assumes a visible, external representation (viz., the empirical body) in its activities in the world, and (iii) the meaning of the world is that it is the arena in which life pursues the goal of its own perfection and growth. Interpreted in this light, his thought is not Gnostic.
