In this study, a paper-integrated analytical device that combined a paper-based colorimetric assay with a paper-based cell culture platform was developed for the in situ detection of hydrogen sulfide (H2S) in three-dimensional (3D)-cultured, live prostate cancer cells. Two kinds of paper substrates were fabricated using a simple wax-printing methodology to form the cell culture and detection zones, respectively. LNCaP cells were seeded directly on the paper substrate and grown in the paper-integrated analytical device. The cell viability and H2S production of LNCaP cells were assessed using a simple water-soluble tetrazolium salt colorimetric assay and H2S-sensing paper, respectively. The H2S-sensing paper showed good sensitivity (sensitivity: 6.12 blue channel intensity/μM H2S, R2 = 0.994) and a limit of quantification of 1.08 μM. As a result, we successfully measured changes in endogenous H2S production in 3D-cultured, live LNCaP cells within the paper-integrated analytical device while varying the duration of incubation and substrate concentration (L-cysteine). This paper-integrated analytical device can provide a simple and effective method to investigate H2S signaling pathways and drug screening in a 3D culture model.

The COVID-19 pandemic has highlighted the need of eco-friendly and locally or distributed manufacturing of diagnostic and safety products. Here, we characterized five handmade papers for their potential application to make paper analytical device (PADs). The handmade papers were made from locally available plant fiber using eco-friendly method. Thickness, grammage, and apparent density of the paper samples ranged from 198 μm to 314 μm, 49 g/m2 to 117.8 g/m2, and 0.23 to 0.39 g/cm3, respectively. Moisture content, water filtration and wicking speed ranged from 5.2% to 7.1%, 35.7 to 156.7, and 0.062 to 0.124 mms-1, respectively. Further, water contact angle and porosity ranged from 76˚ to 112˚ and 79% to 83%, respectively. The best paper sample one was chosen to fabricate PADs which were used for the determination of metformin. The metformin assay on PADs followed linear range from 0.0625 to 0.5 mg/mL. The assay had limit of detection and limit of quantitation of 0.05 mg/mL and 0.18 mg/mL respectively. The new method was used to test metformin samples (n=20) collected from local pharmacies. The average amount of metformin concentration in samples was 465.6 ± 15.1mg/tablet. Three samples did not meet the regulatory standards. When compared with spectrophotometric method, PADs assay correctly predicted 18 out of 20 samples. The PADs assay on handmade paper may provide a low-cost and easy-to-use system to screening the quality of drugs and other point-of-need applications.

Rapid and sensitive detection of acrylamide in food samples is important for food safety and public health. Here, we describe a disposable origami paper-based analytical device (denoted doPAD) for colorimetric detection of acrylamide. This device uniquely exploits 3D origami folding paper for spatial control of the target recognition and signal readout, thus resulting in a positive correlation between the signals and the analytes. Under optimal conditions, the device achieved the quantitative analysis of acrylamide with a limit of detection of 1.13 μg/L within 120 min (including a derivatization time of 90 min and an assay time of 21 min). Furthermore, our method allowed the rapid and sensitive detection of acrylamide in complex food matrices. We envision that the platform described will find useful applications in the fields of food safety and environmental health.

Objective: The aim of this study was to develop Paper-based Analytical Devices (PADs) with colorimetric method as a presumptive test for detecting diazepam in urine Methods: Colorimetric method was used as a principle of this study. PADs were fabricated with wax-printing methods. Chosen colorimetric reagent was tested for selectivity with hydromorphone and codeine; and sensitivity by measuring the absorbance with UV-Vis spectrophotometer. PADs were tested for its sensitivity and stability. The intensity of color developed on PADs are measured with ImageJ. The ability of PADs to detect diazepam in urine was simulated with testing spiked urine sample to the PADs Results: Zimmermann gave the most obvious prominent color change from colorless to purple-red color out of the four reagents. PAD is selective to diazepam when tested with hydromorphone and codeine. PAD is sensitive with a cut-off concentration at 100 ppm. PAD can detect diazepam in urine with the highest recovery percent at 92.8%±4,6 Conclusion: It can be concluded that PAD is quite selective and sensitive to detect diazepam in urine and can be done easily and fast for onsite analysis

Contamination of waterways is of increasing concern, with recent studies demonstrating elevated levels of antibiotics, antidepressants, household, agricultural and industrial chemicals in freshwater systems. Thus, there is a growing demand for methods to rapidly and conveniently monitor contaminants in waterways. Here we demonstrate how a combination of paper microfluidic devices and handheld mobile technology can be used by citizen scientists to carry out a sustained water monitoring campaign. We have developed a paper-based analytical device and a 3 minute sampling workflow that requires no more than a container, a test device and a smartphone app. The contaminant measured in these pilots are phosphates, detectable down to 3 mg L-1. Together these allow volunteers to successfully carry out cost-effective, high frequency, phosphate monitoring over an extended geographies and periods.
