The RNA of clover yellow mosaic virus (CYMV) contains a RNases-resistant oligonucleotide which was determined to be m7GpppGp and located at the 5′ end of RNA. This RNA also contains polyadenylic acid sequences of variable sizes (75–100 residues). These sequences were isolated by RNases T1 and A digestion, by binding to a poly(U)-Sepharose column, and by subsequent electrophoresis in 12% polyacrylamide gels in 7 M urea. The results of specific labelling of the 3′ -OH end of the RNA with 32PCP in the presence of RNA ligase and the ratio of radioactivity present in the poly(A) to that of the total RNA indicated that the poly(A) sequences are located at the 3′-OH end of CYMV-RNA.
