Several problems in engineering realm pose modeling and simulation difficulty due to severe non–linear behavior and debilitating singular or stiff conditions that act as additional impediments. In many such instances advanced numerical schemes are employed to either relax or simplify the PDE that defines the physical process to obtain reasonable output from the simulation. Digressing from this traditional approach, we present an experimental similitude method in this paper to analyze non–linear systems. Combined with the use of original mapping algorithms, we discuss the benefits of empirical similarity techniques and present a heat transfer example for exposition.

<p>Adhesive and repulsive, nm-range surface forces acting between mineral grains control colloidal stability and mineral aggregation but less is known about how these forces are affected by surface reactivity and to what extent these grain-scale forces can influence various deformation processes in rocks. In this experimental work, we explore and quantify the surface forces acting between dynamic mineral surfaces that undergo recrystallization or are chemically reactive in contact with water or aqueous salt solutions. Our experimental setup consists of the surface forces apparatus (SFA) coupled with the multiple beam interferometry (MBI). This setup can excellently reproduce a typical grain contact geometry with nanometer-thin water films confined between contacting mineral grains over relatively large, micron-sized contact areas. Owing to the use of MBI, both surface growth or dissolution processes can be monitored during force measurements in real-time. As such, SFA can provide information about the links between surface reactivity and adhesive or repulsive surface forces. Using the examples of force measurements between recrystallizing or chemically reactive mineral surfaces such as carbonates, hydroxides, and silicates, we comment on the relationship between the measured surface forces and surface reactivity. We link our findings with the observed changes in mineral phases, surface topographies, or surface roughness. We also comment on how the micron-scale confinement in the SFA affects the growth and dissolution processes in contrast to less confined regions. The magnitude of the forces associated with dynamic mineral surfaces and the potential significance of these forces to macroscopic deformation processes and cohesion in rocks are discussed.</p>

Abstract A modeling and simulation approach was used for quantitative comparison of a new generation HER2 antibody drug conjugate (ADC, PF-06804103) with trastuzumab-DM1 (T-DM1). To compare preclinical efficacy, the pharmacokinetic (PK)/pharmacodynamic (PD) relationship of PF-06804103 and T-DM1 was determined across a range of mouse tumor xenograft models, using a tumor growth inhibition model. The tumor static concentration was assigned as the minimal efficacious concentration. PF-06804103 was concluded to be more potent than T-DM1 across cell lines studied. TSCs ranged from 1.0 to 9.8 µg/mL (n = 7) for PF-06804103 and from 4.7 to 29 µg/mL (n = 5) for T-DM1. Two experimental models which were resistant to T-DM1, responded to PF-06804103 treatment. A mechanism-based target mediated drug disposition (TMDD) model was used to predict the human PK of PF-06804103. This model was constructed and validated based on T-DM1 which has non-linear PK at doses administered in the clinic, driven by binding to shed HER2. Non-linear PK is predicted for PF-06804103 in the clinic and is dependent upon circulating HER2 extracellular domain (ECD) concentrations. The models were translated to human and suggested greater efficacy for PF-06804103 compared to T-DM1. In conclusion, a fit-for-purpose translational PK/PD strategy for ADCs is presented and used to compare a new generation HER2 ADC with T-DM1.
