ABSTRACT Prolactin and LH concentrations were measured in the plasma of female canaries sampled during the breeding cycle and after disruption and reinitiation of incubation behaviour. The late incubation period was characterized by low LH and high prolactin concentrations, and canaries separated from their nests at this stage showed an increase in LH and a decline in prolactin within 3 h. In one experiment mean (±s.e.m.) concentrations before and 24 h after nest deprivation were: prolactin 397 ± 86 and 18 ± 5 μg/l; LH 1·04±0·21 and 2·03±0·17 μg/l. Female canaries which abandoned their nests after the eggs had been removed also showed an increase in LH together with a fall in prolactin 24 h after egg removal. When nest-deprived canaries were allowed to resume incubation, plasma prolactin increased again within 5 h and after 2 days had reached levels normal for incubating birds (398± 46 μg/l). Concentrations of LH changed more gradually but had decreased 2 days after the resumption of incubation. Thus prolactin and LH show inverse changes after the disruption and reinitiation of incubation behaviour; it is not clear, however, if the change in one is dependent on the other or if both hormones are responding to the same external stimulus. AbstractRecent experiments demonstrated that in oscine species, 1) Some special sequences of multipartite songs convey more sexual information than others and 2) Stimulus change or switching is often associated with intersexual interactions. We looked for an influence of these two song cues in common domesticated canaries. Female canaries were tested for their sexual copulation solicitation display (CSD) in response to the playbacks of different strings of song sequences from males of the same breed. In order to induce habituation, three monotonous strings were created: A.A.A.A.A.A; B.B.B.B.B.B and C.C.C.C.C.C. (repetition of the same song sequence). In order to manage disrupt of habituation by stimulus changes, four types of strings including two types of song sequences were created: B.B.B.A.A.A., A.A.A.B.B.B., C.C.C.B.B.B. and B.B.B.C.C.C. We found that a monotonous repetition of the same sequence leads to habituation when the sexual potency of the sequence is high (sequences A and B) and that song sequences may greatly differ in eliciting sexual displays (A >B>C). Subsequent stimulus change may elicit an immediate response recovery. But such a disrupt of habituation occurs only if a prefered sequence follows a non prefered one. We discuss prominent parameters that may have influenced female sensitivity and responsiveness to string organization in domesticated canaries.

Phenotype determination of sex in young canaries is very low in accuracy. This study aimed to develop a genotypic sexing method in canaries. This study used 12 canaries consisting of 3 mature males, 3 mature females and 6 one-month-old canaries. Phenotypic sexing by cloacal observation was done on all birds, continued by genotypic sexing to identification CHD1 gene using polymerase chain reaction (PCR). The PCR used blood samples for mature canaries, and feather for mature and one-month-old canaries. The results of phenotypic observations showed that all mature male canaries had prominent and pointed cloaca forms, all mature females had flat and wide, whereas all one-month-old birds had a flat cloaca. The result of PCR showed a single band (500 bp) for mature male and double bands (500 bp and 300 bp) for mature female canaries. The PCR results of one-month-old canaries showed that there were one male and five females. Based on this study, it was concluded that genotypic sexing using the PCR method is effective in the sex determination of canaries.Keywords: canary, CHD1, genotype, PCR, sexing ABSTRAKPenentuan jenis kelamin burung kenari muda secara fenotip akurasinya sangat rendah. Penelitian ini bertujuan untuk menentukan jenis kelamin burung kenari secara genotip. Penelitian ini menggunakan 12 ekor burung kenari, terdiri dari 6 ekor dewasa (3 jantan, 3 betina) serta 6 ekor umur 1 bulan. Semua burung ditentukan jenis kelaminnya dengan mengamati kloaka dan identifikasi gen CHD1 menggunakan teknik polymerase chain reaction (PCR). Sampel DNA berasal dari darah dan bulu untuk burung dewasa serta bulu untuk burung umur 1 bulan. Pengamatan fenotip menunjukkan bahwa burung kenari dewasa jantan mempunyai bentuk kloaka menonjol dan runcing, dewasa betina berbentuk datar dan lebar, sedangkan semua burung umur 1 bulan mempunya bentuk kloaka datar. Hasil identifikasi gen CHD1 diperoleh adanya 1 pita gen sekitar 500 bp dari sampel darah dan bulu semua burung kenari dewasa jantan, dan 2 pita gen sekitar 500 bp dan 300 bp dari sampel semua burung kenari betina dewasa. Hasil PCR pada sampel burung umur 1 bulan menunjukkan bahwa 1 ekor jantan dan 5 ekor betina. Berdasarkan penelitian ini dapat disimpulkan bahwa penentuan jenis kelamin secara genotip menggunakan gen CHD1 dapat dilakukan pada burung kenari.
