SUMMARY In order to study the factors involved in the initiation of allometric mammary growth, which occurs at about 3 weeks of age in the intact female rat, an attempt to induce this phase prematurely in the suckling gonadectomized animal by means of oestrogen was made. It was found, however, that the mammary glands were practically insensitive to physiological doses of this hormone during the suckling period unless anterior pituitary (a.p.) extract was also given. The relative mammary growth rate in animals treated with a.p. plus oestrogen from the 10th to 27th day of life was similar to that found in control rats receiving oestrogen alone from the 20th to 36th day, but in the former groups allometry was initiated before the 20th day of life. It is suggested that both the level of endogenous oestrogen secretion and the functional activity of the anterior pituitary may be important in determining the time of onset of allometric mammary development in the intact animal.

Abstract Estrous cycle is a repetitive phenomenon occurring during the reproductive life of a female dog. The duration of the canine estrous cycle is considerably longer than one in the most of the other animals and is broadly grouped into follicular phase (proestrus and estrus), luteal phase (diestrus) and non-seasonal anestrus. Dogs in the same stage of cycle can be inadvertently assigned to same group during routine safety and metabolic studies leading to possible erroneous interpretation of test-item related effects. This retrospective analysis was conducted by analyzing data of 86 female beagle dogs from control/placebo treated groups to correlate any possible effect of estrous stages with electrocardiography, clinical pathology and ovarian weight. Different estrous cycle stages of beagles were confirmed histologically by evaluating ovary, uterus, vagina and mammary glands. The incidence of beagles in diestrus was the highest, followed by anestrus, proestrus and estrus. No significant effect was noticed on heart rate, P–A, P–D, RR, QRS and QT intervals across different stages of estrous cycle. However, significantly higher PQ (PR) interval in dogs in proestrus stage was observed compared to dogs in anestrus and estrus. Marginally higher WBCs, neutrophils, lymphocytes, RBCs, hemoglobin, AST and lower hematocrit, lipid profile (total cholesterol, HDL, LDL, triglycerides), ALP level was evident in estrous period. Relative ovary weight was significantly higher in dogs in diestrus stage. Considering these results, one may need to exercise caution while interpreting experimental data from female beagle dogs.

17β-Estradiol (E2) exerts inhibitory actions on the nitric oxide pathway in rat adult pituitary glands. Previously, we reported that in vivo E2 acute treatment had opposite effects on soluble guanylyl cyclase (sGC) subunits, increasing α1- and decreasing β1-subunit protein and mRNA expression and decreasing sGC activity in immature rats. Here we studied the E2 effect on sGC protein and mRNA expression in anterior pituitary gland from adult female rats to address whether the maturation of the hypothalamus-pituitary axis influences its effects and to corroborate whether these effects occur in physiological conditions such as during estrous cycle. E2 administration causes the same effect on sGC as seen in immature rats, and these effects are estrogen receptor dependent. These results suggest that E2 is the main effector of these changes. Since the sGC α-subunit increases while the sGC activity decreases, we studied if other less active isoforms of the sGC α-subunit are expressed. Here we show for the first time that sGCα2 and sGCα2 inhibitory (α2i) isoforms are expressed in this gland, but only sGCα2i mRNA increased after E2 acute treatment. Finally, to test whether E2 effects take place under a physiological condition, sGC subunit expression was monitored over estrous cycle. sGCα1, -β1, and -α2i fluctuate along estrous cycle, and these changes are directly related with E2 level fluctuations rather than to NO level variations. These findings show that E2 physiologically regulates sGC expression and highlight a novel mechanism by which E2 downregulates sGC activity in rat anterior pituitary gland.
