Insecticidal cry and vip genes from Bacillus thuringiensis (Bt) have been used for control of lepidopteran insects in transgenic crops. However, novel genes are required for gene pyramiding to delay evolution of resistance to the currently deployed genes. Two PCR-based techniques were employed for screening of cry2-type genes in 129 Bt isolates from diverse habitats in India and 27 known Bt strains. cry2Ab-type genes were more prevalent than cry2Aa- and cry2Ac-type genes. Correlation between source of isolates and abundance of cry2-type genes was not observed. Full-length cry2A-type genes were amplified by PCR from 9 Bt isolates and 4 Bt strains. The genes from Bt isolates SK-758 from Sorghum grain dust and SK-793 from Chilli seeds warehouse, Andhra Pradesh, were cloned and sequenced. The gene from SK-758 (NCBI GenBank accession No. GQ866915) was novel, while that from SK-793 (NCBI GenBank accession No. GQ866914) was identical to the cry2Ab1 gene. The Bacillus thuringiensis Nomenclature Committee ( http://www.lifesci.sussex.ac.uk/home/Neil_Crickmore/Bt/toxins2.html ) named these genes cry2Af2 and cry2Ab16, respectively. The cry2Af2 gene was expressed in Escherichia coli and found to be toxic towards Helicoverpa armigera. The cry2Af2 gene will be useful for pyramiding in transgenic crops.

Yellow rust is caused by Pst (Puccinia striiformis tritici), the most devastating wheat disease with continuous challenge of emerging virulences breaking vertical resistance worldwide resulting in epidemics. Vertical resistance genes incorporation is sustainable, economical and environment-friendly approach to control rust diseases. Wheat landraces (WLR) acquired vertical resistance through long time exposure of host pathogen survival competition in specific area having unique agronomic traits, genetic base and resistance against biotic and abiotic agents can be exploitable commodity for future food production. Fifty Pakistani WLRs already with known vertical resistance were screened against 7 potential Pakistani Pst races at seedling stage under glasshouse conditions to postulate resistance genes. Resistance magnitude was compared among the landraces. Six genes Yr1, Yr8, Yr9, Yr43, Yr44, and YrTr1 were successfully postulated either singly or in combination along with unidentified genes in 45 landraces. Pakistani Pst races are avirulent to Yr5, Yr10, Yr15, Yr24, Yr32, YrSp and YrTye. Most frequently postulated genes are Yr44 found in 22 genotypes, YrTr1 in 21, Yr9 in 19, Yr43 in 18, Yr8 and Yr1 in 14 wheat landraces. Multiple Yr gene pyramiding was observed in (B-74, B-281, B-530) with the presence of Yr8, Yr9, Yr43, Yr44, and YrTr1 and single gene were postulated from 12 genotypes. WLRs (B-03, B-158, B-160, B-171) reaction was immune showing presence of novel Yr genes. Study provides information regarding yellow rust resistance genes originated independently against localized Pst races with desirable agronomic traits since long and can be option for food security in changing environmental challenges.

A Bacillus thuringiensiscry1Ab endotoxint termelő transzgénikus kukorica (DK440BTY) és izogénes, toxint nem termelő kukorica (DK440) rizoszféráját jellemeztük talajbiológiai eszközökkel, illetve összehasonlító értékelést végeztünk egy vegetációs periódus három mintavételi időszakában. Telepszámlálásos módszerrel meghatároztuk néhány kitenyészthető mikrobacsoport (heterotrófok, oligotrófok, spórás mikrooganizmusok, szabadon élő N2-kötők és mikroszkopikus gombák) csíraszámát a rizoszféra talajában. Elvégeztük a mikrobiális össz-aktivitás vizsgálatát fluoreszcein-diacetát (FDA) hidrolízisének mérésével, illetve nyomon követtük a gyökérrendszer szimbiotikus struktúráinak, azaz az arbuszkuláris mikorrhiza gombák kolonizációjának a működőképességét is. A rizoszférához szorosan kapcsolódó talajrészben a szabadon élő szaprotróf Trichoderma gombák faji diverzitásának az alakulását ellenőriztük.  Megállapítottuk, hogy a belső rizoszféra (endoriza) mikroszimbiontás kolonizációja az első mintavétel során (2001. június) szignifikánsan kisebb aktivitást mutatott, és csökkent a gomba működőképességét jelző arbuszkulumok mennyisége is. A további mintavételek során (2001. augusztus és október) azonban ezek a különbségek nem jelentkeztek, a szimbiózis működőképessége helyreállt. A varianciaanalízis azonban összesített hatásban szignifikáns különbséget jelzett. A gyökérfelszín kitenyészthető mikrobacsoportjainak csíraszámában nem adódott statisztikailag igazolható különbség a kétféle növény között. Az FDA módszerrel kimutatható össz-mikrobiális aktivitást ugyanakkor mindegyik mintavételnél nagyobbnak találtuk a Bt-kukorica rizoszférájában, amiből a fiziológiai tulajdonságok megváltozására következtettünk. A transzgénikus növény gyökérhatásának távolabbi részén, a külső rizoszférában a Trichodema gombák száma és faji összetétele csak szezonális változásokat mutatott, de nem különbözött szignifikánsan a génmódosított növénynél. Eredményeink jelzik a génmódosítás közvetett hatását a rizoszférában található „nem célzott” szervezetek összetételére vagy aktivitására, és felhívják a figyelmet a további, tartamjellegű vizsgálatok szükségességére is.
