Gold labeling of two antigens (double labeling) is often done on two section surfaces separated by section thickness. Whether labeling is done on both sides of the same section or on two parallel surfaces separated by section thickness (PSSST), comparable results are dependent on an equal number of epitopes being exposed at each surface. We propose a method to study protein labeling within the same field of proteins, by examining two directly adjacent surfaces that were split during sectioning. The number of labeling sites on adjacent surfaces (AS) were compared to sites on PSSST surfaces in individual bacteria.Since each bacteria needed to be recognizable in all three section surfaces, one-hole grids were used for labeling. One-hole grids require a supporting membrane and excessive handling during labeling often ruptures the membrane. To minimize handling, a labeling chamber was designed that is inexpensive, disposable, minimizes contamination, and uses a minimal amount of solution.

In this study, an effort is made to model the thermal conduction and mass diffusion phenomena in perspective of Buongiorno’s model and Cattaneo-Christov theory for 2D flow of magnetized Burgers nanofluid due to stretching cylinder. Moreover, the impacts of Joule heating and heat source are also included to investigate the heat flow mechanism. Additionally, mass diffusion process in flow of nanofluid is examined by employing the influence of chemical reaction. Mathematical modelling of momentum, heat and mass diffusion equations is carried out in mathematical formulation section of the manuscript. Homotopy analysis method (HAM) in Wolfram Mathematica is utilized to analyze the effects of physical dimensionless constants on flow, temperature and solutal distributions of Burgers nanofluid. Graphical results are depicted and physically justified in results and discussion section. At the end of the manuscript the section of closing remarks is also included to highlight the main findings of this study. It is revealed that an escalation in thermal relaxation time constant leads to ascend the temperature curves of nanofluid. Additionally, depreciation is assessed in mass diffusion process due to escalating amount of thermophoretic force constant.

Biodiesel produced by transesterification process from vegetable oils or animal fats is viewed as a promising renewable energy source. Now a day’s diminishing of petroleum reserves in the ground and increasing environmental pollution prevention and regulations have made searching for renewable oxygenated energy sources from biomasses. Biodiesel is non-toxic, renewable, biodegradable, environmentally benign, energy efficient and diesel substituent fuel used in diesel engine which contributes minimal amount of global warming gases such as CO, CO2, SO2, NOX, unburned hydrocarbons, and particulate matters. The chemical composition of the biodiesel was examined by help of GC-MS and five fatty acid methyl esters such as methyl palmitate, methyl stearate, methyl oleate, methyl linoleate and methyl linoleneate were identified. The variables that affect the amount of biodiesel such as methanol/oil molar ratio, mass weight of catalyst and temperature were studied. In addition to this the physicochemical properties of the biodiesel such as (density, kinematic viscosity, iodine value high heating value, flash point, acidic value, saponification value, carbon residue, peroxide value and ester content) were determined and its corresponding values were 87 Kg/m3, 5.63 Mm2/s, 39.56 g I/100g oil, 42.22 MJ/Kg, 132oC, 0.12 mgKOH/g, 209.72 mgKOH/g, 0.04%wt, 12.63 meq/kg, and 92.67 wt% respectively. The results of the present study showed that all physicochemical properties lie within the ASTM and EN biodiesel standards. Therefore, mango seed oil methyl ester could be used as an alternative to diesel engine.

Abstract Atomic force probing (AFP) uses very sharp tungsten tips (100nm in radius) which wear out rather quickly, even with the greater durability of tungsten as compared to silicon. This paper demonstrates how worn tips that no longer image and probe properly can be reconditioned using the focus ion beam (FIB) tool. The method works best for tips that are under approx. 750nm in diameter and are not bent. It works well for freshly manufactured tips that do not work properly due to mishandling or improper storage which allowed particulates/oxide to build up on the tip. The method also works well for fresh tips that have been worn down (or slightly bent) after several hours of scanning and probing. This method is straightforward and requires a minimal amount of time. Typically, four probe tips can be reconditioned in about 30 minutes on the FIB.
