A protocol for micropropagation of Tribulus terrestris, an important medicinal herb was established using juvenile explants viz., leaf, node and internode. All the explants were tested for callus induction on Murashige and Skoog’s (MS) medium, supplemented with BAP, NAA and 2,4-D. Among the three explants leaf explant responded well (98%) for the callus induction in the MS medium composted with BAP and NAA (4.0 and 0.5 mg/L) followed by the nodal segments (58.75%) in the same medium. Maximum number of shoot induction from the callus of leaf derived explants (91.1%) was perceived on MS medium fortified with BAP 4.0 mg/L and NAA (0.5 mg/L). Moreover, root elongation and profuse rooting percentage (77.19%) were achieved when the well-grown shoots were cultured on MS media supplemented with IAA (2.0 mg/L) for leaf callus derived shoots. The regenerated plantlets were hardened and established at 80% survival rate in hardening media encompassed with red soil, sand and vermicompost in the ratio of 1:1:1 by volume.

In the present study the protocol for callus induction and regeneration in Azima tetracantha has been developed in culture medium. The young apical leaf explants were used for callus induction on MS medium containing BAP and NAA at 1.0 and 0.4mgl-1 respectively showed maximum callus induction (73%). The amount of callus responded for shoot formation (74%) was obtained in the MS medium containing BAP (1.5 mgl-1) and NAA (0.3mgl-1).The elongated shoots were rooted on half strength medium supplemented with IBA (1.5 mgl-1) and Kn (0.4 mgl-1) for shoots rooted. Regenerated plantlets were successfully acclimatized and hardened off inside the culture and then transferred to green house with better survival rate.

The experiment was conducted during May to December 2008 in the Biotechnology Laboratory of Bangladesh Agricultural University, Mymensingh to observe the callus induction, regeneration potentiality and to establish a suitable in vitro plantlet regeneration protocol of Corchorus olitorius. MS medium supplemented with different phytohormone concentrations and combinations were used to observe the callus induction, shoot regeneration and root formation ability of the cotyledon with attached petiole derived explant of three genotypes viz. O-9897, O-72 and OM-1. The highest callus induction (92.85%) was observed in O-9897 followed by O-72 (82.14%) in the MS media supplemented with 2.5 mg/L BAP + 0.5 mg/L IAA. Genotype O-9897 in MS media supplemented with 2.5 mg/L BAP + 0.5 mg/L IAA produced the highest percentage of shoot regenerants (83.33%) followed by O-72 (75.00%) in the media supplemented with 2.5 mg/L BAP + 0.5 mg/L IAA. The root formation from regenerants was the best on halfstrength of MS media supplemented with 0.6 mg/L IBA in genotype O-9897 (45.00%). The in vitro regenerated plantlets from the genotypes O-9897 could be established in the field. Therefore, the genotypes O-9897 of C. olitorius in MS media supplemented with 2.5 mg/L BAP + 0.5 mg/L IAA could be used for callus induction and shoot regeneration. In vitro study was carried out in an important medicinal plant Tinospora cordifolia (Willd.) Miers belonging to the family: Menispermaceae. Vegetative parts such as stem, leaf and nodal explants were excised from an elite in vivo grown mature plant and thereafter cultured on Murashige-Skoog (MS) medium supplemented with different hormonal concentrations for callus induction and organogenesis. Callus formation occurred from nodal segments, leaf and inter-node explants when planted on different combinations of hormones. Tinospora cordifolia showed response for in vitro shoot growth from the nodal segment. The best shoot growth was observed on MS medium supplemented with kinetin (1.5 mg/l). Similarly, the best result for root induction was obtained on MS medium supplemented with 6-benzylaminopurine (1.0 mg/l) and naphthaleneacetic acid (2.5 mg/l). This study reports on an improved protocol for callus induction and subsequent regeneration from nodal segment of wood apple (Aegle marmelos L.) Creamish friable competent callus was achieved from nodal segments on MS medium augmented with 4.0 mg1-1 2,4-D within two weeks of inoculation. The callus produced large number of shoots when cultured on MS medium fortified with 2.0 mgl-1 BAP+0.1 mgl-1 NAA within ten days of culture. In vitro raised shoots were rooted on half strength MS medium enriched with 1.0 mgl-1 IBA within fifteen days of culture. The rooted plantlets were successfully established with 80% survival. The procedure for Heuchera villosa ‘Caramel’ propagation was investigated, which involves shoot regeneration, rooting of regenerated shoots, and acclimation of regenerated plantlets. Petioles, as explants, were cultured on MS medium supplemented with 1-naphthylacetic acid (NAA), benzylaminopurine (BA), thidiazuron (TDZ) and callus formed on all media. Shoots were observed to proliferate from callus on media with BA and NAA, whereas no shoots regenerated on media with TDZ and NAA. On media containing 0.5 or 1.0 mg·L−1 BA in combination with NAA, the regenerated shoots showed severe hyperhydricity, whereas on media containing 0.1 mg·L−1 BA in combination with NAA, the regenerated shoots grew normally. The highest shoot induction rate, 90.6%, was obtained on media containing 0.1 mg·L−1 BA and 0.01 mg·L−1 NAA. The effects of indole-3-acetic acid (IAA), indole-3-butyric acid (IBA), and NAA on rooting of H. villosa ‘Caramel’ was explored. The highest rooting rate (95%) was obtained on 1/2 MS medium containing 0.2 mg·L−1 NAA. In the subsequent acclimation experiments, about 85% of rooted plantlets survived and grew normally.

ABSTRACT The aim of this study was to establish the best concentrations of growth regulators for in vitro cultivation of jambu for a subsequent elaboration of an efficient micropropagation protocol. After sterilized, the seeds were inoculated on different media (MS, ½MS and water-agar) for in vitro germination. Nodal segments of in vitro germinated jambu seedlings were used as explants in the micropropagation with different concentrations of 6-benzylaminopurine (BAP) (0.0; 0.125; 0.25; 0.50 and 0.75 mg L-1) and callus induction with 2.4- dichlorophenoxyacetic acid (2.4-D) (0.0; 0.25; 0.50; 0.75 and 1.0 mg L-1) on Murashige & Skoog’s (MS) medium. The highest germination rates were obtained on MS medium with better seedling development and greater height (3.7cm). In micropropagation, the best treatment was obtained on 0.125 BAP (T2), with an average of 2.2 sprouts/explant, average height of 2.4 cm and vigorous sprouts. In callus induction, all treatments with 2.4-D had developed friable calluses in 30 days and using doses of 0.25 and 0.50 mg L-1 provided greater fresh matter. The induction of friable callus and the root production occur without supplementation of exogenous growth regulator.

Efficient plant regeneration could be obtained from the derooted nodal segments of Centella asiatica with stole buds as the explant, when cultured on MS medium supplemented with BAP (10 ppm)+NAA(2ppm). Both callus and regeneration occurred simultaneously on the same medium. Profuse rooting were obtain on MS medium fortified with NAA (2ppm) from leaf explants. Shoot and root proliferation were observed on the medium supplemented with BAP(5 ppm)and NAA (2 ppm) through subculture. Mass propagation of plantlets were obtained through invitro culture.

The effect of different explants and concentrations of BAP and NAA on induction of callus and plant regeneration of brinjal cv. Jhumki were investigated. The treatment combinations were BAP (0. 2.0. 3.0, and 4.0 mg/l) and NAA (0. 0.1, 0.5, and 1.0 mg/l). The rate of callus formation varied in different treatments. The highest amount of callus (48.66%) was produced on MS medium containing 2.0 mg/l BAP and 0.5 mg/l NAA from stem, and 8.2 days required for callus induction. The highest fresh weight of callus was 1.12g from stem and 0.48g from root. The number of shoot regenerated through callus from stem containing 2.0 mg/l BAP and 0.5 mg/l NAA was 3.4 (23.287%) and days required for 38.8 days. Licorice plants, Glycyrrhiza glabra, G. uralensis, and G. inflata, were investigated for callus induction using Murashige and Skoog (MS) medium combined with auxins and cytokinins. After 4 weeks of culture, 33-100% of leaf or stem explants formed calli. Maximum of shoot induction from callus cultures was achieved by G. inflata stem explants cultured on MS medium supplemented with 1 mg/l α-naphthaleneacetic acid (NAA) and 0.5 mg/l 6-benzyladenine (BA) (67%) which also gave maximum shoot formation per explant (two shoots per explant). These results indicated that all three Glycyrrhiza species regenerated shoots from callus cultures on MS medium combined with NAA and BA or only thidiazuron (TDZ; 0.1 and 0.5 mg/l). Glycyrrhizin contents of G. uralensis calli induced using MS medium in combination with NAA and BA [(27.60 ± 8.47) μg/g DW] or TDZ alone [(36.52 ± 2.45) μg/ g DW] were higher than those found in other combinations.

To observe the possibility of producing haploid plants of Chrysanthemum, anthers of three Korean cultivars ‘Yes Morning’, ‘Hi-Maya’, and pot cultivar ‘Peace Pink’ were cultured. Callus induction among cultivars differed little, but equally good results were obtained with the basal MS medium supplemented with 1 mg/L of 2,4-D, 2 mg/L of BA, 250 mg/L of casein hydrolysate, 45 g/L of sucrose; solidified by 2.75 g/L gelrite. A pretreatment of anthers in media at 4 °C for 48h enhanced the callus induction. Calli were allowed to differentiate on basal MS medium supplemented with 2 mg/L of BA, 0.1 mg/L of NAA, 30 g/L of sucrose; solidified by 2.75 g/L gelrite.  Shoot formation from calli in that media slightly differed among cultivars. Multiple shoots elongated from calli were shifted to basal MS medium supplemented with 0.1 mg/L of NAA, 30 g/L of sucrose; solidified by 3 g/L gelrite for rooting. The plantlets with sufficient roots thus obtained were acclimatized and transferred to the soil. Fifty regenerated plantlets from each cultivar were randomly selected for ploidy observation by chromosome counting and haploid plantlet was detected for the garden cultivar ‘Yes morning’.
