The in vitro regeneration of Plubago zeylanica exhibited that the callus was initiated in the basal medium containing BAP, NAA, 2, 4-D, and IBA.  The high amount (90%) of organic calli was induced in the basal medium supplemented with 2, 4-D, alone at 2.0 mg/l. In the subculture the adventitious shoot formation was prominently higher (83%) in the basal medium containing BAP, and NAA at 3.5 and 0.3 mg/l, respectively. IAA (1.0 mg/l)effectively produced higher percen-tage (90) of roots and root growth. After sequential hardening, survivability rate was observed to be significantly higher (80%) in the hardening medium containing garden soil, sand and vermicompost in the ratio of 1 : 1 : 1 by volume under greenhouse condition.  Key words: Plumbago zeylanica, In vitro regeneration, Medicinal plant D.O.I. 10.3329/ptcb.v18i2.3648 Plant Tissue Cult. & Biotech. 18(2): 173-179, 2008 (December)

An efficient in vitro propagation of an endangered medicinal plant Vanasushava pedata (Apiaceae) by axillary shoot proliferation from nodal segments of mature plants was designed. The medium type and growth regulators markedly influenced in vitro regeneration of V. pedata. An in vitro plantlet production system has been investigated on MS with the synergistic combination of BA (5.0 mg/l), IAA (0.1 mg/l) and 3 % sucrose which promoted the maximum number of shoots (8.6) as well as enhanced shoot lengths. Subculturing of nodal segments from in vitro derived shoots on a similar medium enabled continuous production of healthy shoots with a similar frequency. Rooting was highest (100%) on half strength MS containing IAA (2.0 mg/l). Micropropagated plants established in garden soil and forest humus (1 : 1) were uniform and identical to the donor plants with respect of growth characteristics as well as floral features. The objective of this work was to study the histological events related to the regeneration process of a medicinal plant, Nymphoides cristatum (Roxb). Leaf explants were cultured on MS medium supplemented with 0.5 mgl-1 of 6-benzyl amino purine (BAP). About 90% of explants gave rise to shoots after 15 days of incubation. The histological studies showed that the regeneration originated directly from parenchymatous cells and direct organogenesis after 20 days of culture could be observed. Buds and roots were found completely differentiated after 40 days of culture and number of shoots per explants was 30. Micorshoots were rooted in hormone - free medium and the plants obtained grew in artificial pond under green house conditions. Radermachera xylocarpa (Roxb.) K. Schum. is a rare indigenous forest tree species which is utilized for its wood and medicinal properties. Due to its overexploitation and specific habitat requirements the species is restricted to limited areas. In vitro mass propagation of tree species faces various challenges and no such efforts have yet been taken in propagation of this useful plant using these methods.  In order to overcome the hurdles and understanding an urgent need of its conservation and mass propagation present authors attempt to develop a simple effective tissue culture protocol for regeneration of R. xylocarpa. Nodal explants were cultured on MS supplemented with various concentrations of cytokinins and auxins.  Among different cytokinins, maximum bud induction and proliferation was obtained in media supplemented with Kn along with IBA and for effective root induction which is tough to obtain in tree species, 100% rooting was achieved in cultures with increasing concentrations of IBA. Field survival is a major challenge with regenerated plants of forest tree species. We report here for the first time 100% survival of plants in soil by carefully standardizing the period of hardening and acclimatization procedures. A novel and effective in vitro regeneration protocol of R. xylocarpa has been successfully standardized which can be adopted for large scale propagation, reforestation and conservation of rare Radermachera xylocarpa of medicinal importance.Plant Tissue Cult. & Biotech. 23(1): 21?29, 2013 (June)DOI: http://dx.doi.org/10.3329/ptcb.v23i1.15556
