Modeling fast stimulus–response association learning along the occipito-parieto-frontal pathway following rule instructions

Learned habitual responses to environmental stimuli allow efficient interaction with the environment, freeing cognitive resources for more demanding tasks. However, when the outcome of such actions is no longer a desired goal, established stimulus–response (S-R) associations or habits must be overcome. Among people with substance use disorders (SUDs), difficulty in overcoming habitual responses to stimuli associated with their addiction in favor of new, goal-directed behaviors contributes to relapse. Animal models of habit learning demonstrate that chronic self-administration of drugs of abuse promotes habitual responding beyond the domain of compulsive drug seeking. However, whether a similar propensity toward domain-general habitual responding occurs in humans with SUDs has remained unclear. To address this question, we used a visuomotor S-R learning and relearning task, the Hidden Association between Images Task, which employs abstract visual stimuli and manual responses. This task allows us to measure new S-R association learning and well-learned S-R association execution and includes a response contingency change manipulation to quantify the degree to which responding is habit-based, rather than goal-directed. We find that people with SUDs learn new S-R associations as well as healthy control participants do. Moreover, people with an SUD history slightly outperform controls in S-R execution. In contrast, people with SUDs are specifically impaired in overcoming well-learned S-R associations; those with SUDs make a significantly greater proportion of perseverative errors during well-learned S-R replacement, indicating the more habitual nature of their responses. Thus, with equivalent training and practice, people with SUDs appear to show enhanced domain-general habit formation.

1. The supplementary eye field (SEF) has been viewed as a premotor cortical field for the selection and control of saccadic eye movements. Drawing on studies of the neighboring premotor cortex, we hypothesized that if the SEF participates in the selection of action based on arbitrary stimulus-response associations, then task-related activity in the SEF should change during the learning of such associations. 2. Rhesus monkeys were operantly conditioned to make a saccadic eye movement to one of four targets (7 deg up, down, left, and right from center) in response to a foveal instruction stimulus (IS). One and only one of those four possible responses was arbitrarily designated "correct" for each IS. The monkeys responded to familiar ISs, four stimuli that remained unchanged throughout training and recording, as well as to novel ISs, which the monkeys had not previously seen. The monkeys initially chose responses to novel stimuli by trial and error, with near chance levels of performance, but quickly learned to select the correct saccade. 3. We studied 186 SEF cells as monkeys learned new visuomotor associations. Neuronal activity was quantified in four task periods: during the presentation of the IS, during an instructed delay period (i.e., after the removal of the IS but before a trigger or "go" stimulus), just before the saccade, and after the saccade during fixation of the target location. The discharge rate in each task period was considered a separate case for analysis, compared with that in a reference period preceding the IS, and eliminated from further analysis if not significantly different. 4. We observed two main categories of activity change during learning, which we termed learning selective and learning dependent. Learning-selective cases showed a significant evolution in activity as the monkeys learned which saccade was instructed by a novel IS, but had no significant modulation during trials with familiar ISs. Many of these cells were virtually inactive on trials with familiar ISs. However, they initially showed dramatic modulation when tested with a novel IS. As the monkey chose the correct saccade (or target) with increasing reliability, the modulation often decremented until the cell was again relatively unmodulated, as observed during familiar-IS trials. These cells usually remained relatively inactive until the monkeys were challenged to start learning another new stimulus-response association. Learning-selective activity was observed in all task periods, and 33 (18%) of the 186 adequately tested SEF cells showed learning-selective activity in one or more task periods.(ABSTRACT TRUNCATED AT 400 WORDS)

The use of computer monitors to provide technology-based written feedback during clinical sessions, referred to as “bug-in-the-eye” (BITi) feedback, recently emerged in the literature with preliminary evidence to support its effectiveness (Carmel, Villatte, Rosenthal, Chalker & Comtois, 2015; Weck et al., 2016). This investigation employed a single-subject, sequential A-B design with two participants to observe the effects of implementing BITi feedback using a smartwatch on the clinical behavior of student clinicians (SCs). Baseline and treatment data on the stimulus-response-consequence (S-R-C) contingency completion rates of SCs were collected using 10 minute segments of recorded therapy sessions. All participants were students enrolled in a clinical practicum experience in a communication disorders and sciences (CDS) program. A celeration line, descriptive statistics, and stability band were used to analyze the data by slope, trend, and variability. Results demonstrated a significant correlative relationship between BITi feedback with a smartwatch and an increase in positive clinical behaviors. Based on qualitative interviews and exit rating scales, SCs reported BITi feedback was noninvasive and minimally distracting. Preliminary evidence suggests BITi feedback with a smartwatch may be an effective tool for providing real-time clinical feedback.
