In order to study early events of glycogen synthesis, we have used adrenalectomized (ADX) rats fasted overnight and injected with the synthetic glucocorticoid dexamethasone (DEX) to stimulate glycogen synthesis. Rats were given DEX 0-5 hr prior to sacrifice and injected with 2 mCi 3H-galactose 1 hr prior to sacrifice. Liver was prepared for light (LM) and electron microscopic (EM) radioautography by routine procedures.The concentration of silver grains over hepatic cytoplasm was measured in LM radioautographs using a Zeiss Videoplan. The hepatocytes were categorized as unlabeled if no silver grains (gr) were present, lightly labeled (<10gr/100 μm2 cytoplasm) or intensely labeled (>10 gr/1002 μm cytoplasm). Although very few hepatocytes showed heavy labeling after 1 hr treatment with DEX, by 2 hr after DEX treatment 8% of the cells distributed throughout the lobule were intensely labeled.

ABSTRACT Adrenalectomized rats were used for the determination of the relative potency of various 6- and 9-halo substituted corticoids administered subcutaneously or by gavage using thymus weightas the endpoint. By subcutaneous injection, fluocinolone acetonide was the most active corticoid at 700 times that of cortisol. This compound was also the most active corticoid by the gavage route and was judged to be 570 times as active as the standard cortisol. The introduction of the 16,17-acetonide and 16,17-acetone 21-acetate groups into 17α,21-dihydroxy-9α,11β-dichloropregna-1,4-diene-3,20-dione increased the activity by a factor of 42 and 100, respectively. The introduction of the δ1 double bond into 6α-fluoroprogesterone 16,17-acetonide caused an increase of 10-fold in thymolytic activity assessed by the subcutaneous route

ABSTRACT Pituitary and blood corticotrophin (ACTH) concentrations were estimated in adrenalectomized rats maintained on cortisol or crude adrenocortical extract (ACE) with and without ascorbic acid. Blood ACTH levels were also determined in similarly treated animals after subjection to mild stress. Thirty days after adrenalectomy there were marked rises in pituitary and blood corticotrophin concentration which were prevented by daily treatment with small doses of cortisol or ACE. In contrast to intact animals, adrenalectomized rats exhibited markedly increased adrenocorticotrophic responses to mild stress. This pituitary hyperexcitability was not diminished by replacement therapy with corticoids alone but was considerably reduced by ascorbic acid given in conjunction with ACE. It is suggested that corticoids control the rate at which ACTH is synthesised in the pituitary gland and that other factors, including ascorbic acid, control the excitability of the pituitary gland in its adrenocorticotrophic response to stress.

In a new method for measurement of inactive rat plasma renin, the trypsin generated angiotensin I immunoreactive material, which was HPLC characterized as similar to tetradecapeptide renin substrate, is removed by a cation exchange resin before the renin incubation step. The method also corrects for trypsin destruction of endogenous angiotensinogen by the addition of exogenous angiotensinogen. When measured with this method inactive renin in rat plasma decreased after nephrectomy and increased after adrenalectomy. This is in accordance with findings in humans. A sexual dimorphism of prorenin (inactive renin) in rat plasma, similar to that reported in humans and mice, was demonstrated. Thus, inactive renin in the rat is no exception among species, and the rat might be a suitable animal model for further studies dealing with the physiology of prorenin in plasma and tissues.Key words: angiotensinogen, inactive renin, renin.

We previously reported the elevation of plasma interleukin (IL)-6 activity in response to immobilization stress in rats. To investigate the role of peripheral corticotropin-releasing factor (CRF) in this response, we examined the effects of CRF antagonists on immobilization-induced IL-6 response. Intravenous pretreatment with either [d-Phe12,Nle21,38,CαMeLeu37]-anti-human rat (h/r) CRF12—41(1.5 mg/kg) or cyclo(30—33)[d-Phe12, Nle21,38,Glu30,Lys33]-h/rCRF12—41(Astressin, 0.5 mg/kg) attenuated the IL-6 response to immobilization, which confirmed our previous finding that systemic administration of an antiserum against CRF blocked this response. In addition, an intraperitoneal injection of h/rCRF (100 μg/kg) or rat urocortin (10 and 100 μg/kg) increased the plasma IL-6 activity, mimicking the response to immobilization. An intravenous injection of h/rCRF (100 μg/kg) also elevated plasma IL-6 in adrenalectomized rats. These findings suggest that peripheral CRF mediates the plasma IL-6 elevation in response to immobilization.
