The effect of calcium on heart rate, A-V conduction, and contractility was studied in the dog's heart-lung preparation. Normal plasma calcium concentration was 2.71 ± 0.20 mm (N = 25). Heart rate was increased by calcium in the range from 1.6 to 14.0 mm and fell at still higher levels. A-V conduction time was shortest at calcium concentration slightly above normal, and decreased at higher as well as lower concentrations. At levels above 17 mm conduction block, fibrillation or cardiac arrest, or both, occurred. Contractile force was markedly reduced at low calcium. Increase of calcium lowered ventricular filling pressure, increased the rate of tension development and of relaxation, and shortened duration of contraction. The effects of calcium were not altered by pretreatment with reserpine (2 x 0.5 mg/kg, s.c., given 48 and 24 hr prior to experiment) and were not influenced by atropine (2–5 mg). The effect of calcium on contractility was fully developed 100 sec after administration; the effect on sinoatrial rhythm was established only after 4.4 ± 0.54 min.

A bolus injection of methylene blue (1 mg), a guanylate cyclase inhibitor, or aspirin (3 mg) in the isolated rat lung preparation had little or no effect on resting perfusion pressure under normoxic condition. In contrast, methylene blue markedly potentiated hypoxic vasopressor response (4-fold) when injected before or during the alveolar hypoxic stimulation. Hemoglobin also potentiated the hypoxic pressor response. Similarly, methylene blue or aspirin augmented the pressor responses to angiotensin II (0.1–1 microgram). The increased hypoxic response induced by methylene blue was immediate and sustained. Methylene blue, when added during hypoxia in the presence of aspirin, further augmented the response to hypoxia compared with the enhanced hypoxic response observed with aspirin alone. Our results suggest that, in addition to the role of cyclooxygenase products, the pulmonary vascular bed may be regulated by endothelium-dependent factors that can be antagonized directly or indirectly by methylene blue.

The effect of indomethacin treatment of the pulmonary vasodilation caused by ventilation of the fetal lung with air was evaluated in anesthetized, exteriorized, fetal goats by means of an open-chest, pump-perfused lung preparation. The decrease in pulmonary vascular resistance that occurs when the fetal lung is ventilated with air consists of two components: 1) a rapid decrease in pulmonary vascular resistance during the first 30 s of ventilation; 2) a slower decline, which continues through the first 10-20 min or more of ventilation. Indomethacin has no effect on the first component. The second component is absent following indomethacin pretreatment. The effect of indomethacin treatment is more pronounced in immature fetuses (less than 90% gestation) than in mature fetuses. Prostaglandin synthase activity appears to be important in the pulmonary vasodilation caused by ventilation of the fetal lungs with air.
