The morphology, infraciliature and molecular phylogeny of two marine ciliated protozoans, Diophrys blakeneyensis spec. nov. and Diophrys oligothrix Borror, 1965, isolated from British salt marshes, were investigated using microscopic observations of live and protargol-impregnated specimens, and by small subunit (SSU) rRNA gene sequence analysis. Diophrys blakeneyensis spec. nov. is characterized as follows: cell oval to rectangular in outline; size variable, approximately 60–180 × 30–80 µm in vivo; adoral zone comprising about 45 membranelles; usually five frontal, two ventral, five transverse, two left marginal and three caudal cirri; five dorsal kineties with more than 10 dikinetids each; 7–23 spherical to ellipsoid macronuclear nodules in a ring-like pattern; marine biotope. The population of Diophrys oligothrix described here corresponds well with previous populations in terms of its general morphology and ciliary pattern, in particular the continuous ciliary rows on the dorsal side with loosely arranged cilia. The main differences between the present and previously reported populations are the broader buccal field and greater number of dorsal kineties in the present population, both of which are regarded as population-dependent features. Phylogenetic analyses based on SSU rRNA gene sequence data demonstrate that Diophrys blakeneyensis is most closely related to Diophrys oligothrix, and both organisms cluster with two congeners with high bootstrap support within a larger group that contain the core species of the Diophrys-complex. Cladistic analysis based on morphological and morphogenetic data broadly agree with the SSU rRNA gene sequence phylogeny. Both analyses suggest that the genus Diophrys may be polyphyletic.

AbstractThe life cycle, spore morphology, pathogenicity, tissue specificity, mode of transmission and small subunit rRNA (SSU-rRNA) gene sequence analysis of the five new microsporidian isolates viz., NIWB-11bp, NIWB-12n, NIWB-13md, NIWB-14b and NIWB-15mb identified from the silkworm, Bombyx mori have been studied along with type species, NIK-1s_mys. The life cycle of the microsporidians identified exhibited the sequential developmental cycles that are similar to the general developmental cycle of the genus, Nosema. The spores showed considerable variations in their shape, length and width. The pathogenicity observed was dose-dependent and differed from each of the microsporidian isolates; the NIWB-15mb was found to be more virulent than other isolates. All of the microsporidians were found to infect most of the tissues examined and showed gonadal infection and transovarial transmission in the infected silkworms. SSU-rRNA sequence based phylogenetic tree placed NIWB-14b, NIWB-12n and NIWB-11bp in a separate branch along with other Nosema species and Nosema bombycis; while NIWB-15mb and NIWB-13md together formed another cluster along with other Nosema species. NIK-1s_mys revealed a signature sequence similar to standard type species, N. bombycis, indicating that NIK-1s_mys is similar to N. bombycis. Based on phylogenetic relationships, branch length information based on genetic distance and nucleotide differences, we conclude that the microsporidian isolates identified are distinctly different from the other known species and belonging to the genus, Nosema. This SSU-rRNA gene sequence analysis method is found to be more useful approach in detecting different and closely related microsporidians of this economically important domestic insect.

A novel sulfate-reducing bacterium, designated C1TLV30T, was isolated from wood falls at a depth of 1693 m in the Mediterranean Sea. Cells were motile vibrios (2–4×0.5 µm). Strain C1TLV30T grew at temperatures between 15 and 45 °C (optimum 30 °C) and at pH 5.4–8.6 (optimum 7.3). It required NaCl for growth (optimum at 25 g NaCl l−1) and tolerated up to 80 g NaCl l−1. Strain C1TLV30T used as energy sources: lactate, fumarate, formate, malate, pyruvate and ethanol. The end products from lactate oxidation were acetate, H2S and CO2 in the presence of sulfate as terminal electron acceptor. Besides sulfate, thiosulfate and sulfite were also used as terminal electron acceptors, but not elemental sulfur, fumarate, nitrate or nitrite. Strain C1TLV30T possessed desulfoviridin and was piezophilic, growing optimally at 10 MPa (range 0–30 MPa). The membrane lipid composition of this strain was examined to reveal an increase in fatty acid chain lengths at high hydrostatic pressures. The G+C content of the genomic DNA was 49.6 % and the genome size was estimated at 3.5±0.5 Mb. Phylogenetic analysis of the SSU rRNA gene sequence indicated that strain C1TLV30T was affiliated to the genus Desulfovibrio with Desulfovibrio profundus being its closest phylogenetic relative (similarity of 96.4 %). On the basis of SSU rRNA gene sequence comparisons and physiological characteristics, strain C1TLV30T ( = DSM 21447T  = JCM 1548T) is proposed to be assigned to a novel species of the genus Desulfovibrio, Desulfovibrio piezophilus sp. nov.

The morphology and infraciliature of a hypotrichous ciliate collected from brackish-water biotope (salinity 5 parts per thousand) in South Korea were investigated which indicate this organism should be a new form within the genus Anteholosticha Berger, 2003. Careful morphological comparison and SSU rRNA gene sequence alignment with similar species are performed to support the validity of Anteholosticha pseudomonilata n. sp. The main diagnostic and distinguishing characteristics of the new species include: 1) 8-11 transverse cirri; 2) midventral complex composed of 10-16 pairs of zigzagging midventral cirri, extending posteriorly to slightly ahead of pretransverse cirri; 3) cortical granules colourless and pigment-like, 0.5 mu m across, longitudinally arranged in more or less short rows on whole cortex except along dorsal kineties and cirral rows; and 4) 8-12 macronuclear nodules located left of midline.

The morphologies of the three freshwater stentorid ciliates in Korea, Stentor coeruleus (Pallas, 1766); Stentor muelleri Ehrenberg, 1831, and Stentor tartari Murthy & Bai, 1974, were investigated based on live observations and protargol impregnation. The Korean population of S. tartari exhibits the following characteristics: body size 200–355 × 85–135 µm in vivo, 62–106 somatic kineties, 8–13 peristomial kineties, 110–180 adoral membranelles, mostly two macronuclear nodules and 5–18 micronuclei, reddish and colorless cortical granules and the presence of symbiotic algae. We identified S. tartari based on unique characteristics compared to close congeners. Korean populations of S. coeruleus and S. muelleri are congruent with previously described populations in most aspects of their morphologies. Here, for the first time, we report molecular gene sequence information for S. tartari. Small subunit (SSU) rRNA gene sequence-based phylogeny indicates that S. tartari, which has multiple macronuclei, forms a monophyletic group with other Stentor species having a single macronucleus. Our findings based on morphology and SSU rRNA gene sequence information corroborate the hypothesis that the elongated macronucleus evolved from the compact single or multi macronucleus state.
