O objetivo deste trabalho foi avaliar a eficiência de Beauveria bassiana (Bb), terra diatomácea (TD) e microrganismos eficazes (EM-4), associados ou não, no controle de Alphitobius diaperinus, e o efeito de EM-4 no desenvolvimento de B. bassiana. Os agentes de controle (Bb, TD e EM-4), em diferentes concentrações e combinações, foram aplicados em uma mistura de cama-de-frango e ração, em que os insetos foram acondicionados por dez dias. Avaliaram-se, in vitro: a germinação, as unidades formadoras de colônia (UFC), o crescimento vegetativo e a produtividade de conídios de Bb em contato com suspensão aquosa de EM-4 (1%) não filtrada e filtrada. Os maiores índices de mortalidade foram observados nos tratamentos TD + Bb + EM-4 e TD + Bb, nas maiores concentrações. Verificou-se efeito não-aditivo sinérgico para TD + Bb nas três concentrações. Nos testes com EM-4 filtrado, não houve diferença em relação à testemunha quanto à germinação e às UFC, entretanto, o crescimento vegetativo e a produtividade de conídios foram negativamente afetados. O uso conjunto de B. bassiana e terra diatomácea, para o manejo de populações de A. diaperinus, pode reduzir o uso de produtos químicos.

Ctenocephalides felis is an ectoparasitic flea species commonly found on dogs and cats. The current study verified the in vitro virulence of conidia of the entomopathogenic fungus Beauveria bassiana produced under different color LED light (red, blue, purple, green, yellow, and white) to adults of C. felis. The fungal isolates were cultivated on malt extract agar (MEA). Bioassay treatments used aerial conidia in test tubes. Adult fleas were obtained from a house cat in Chiang Mai province, Thailand. The experiments were composed of one control and eleven treatment groups. All of the treatments with B. bassiana conidia caused adult mortality after an exposure of 12 h. Among the conditions used in this study, B. bassiana cultured under red LED and fluorescent light were the most effective in causing mortality (100 %) in adult fleas after 36 h. The experimental results indicate that these aerial conidia of B. bassiana have promising potential for use in control of C. felis adult stages.

The in vitro fungitoxic effect of three biofertilizers, E.M.-4, Multibion <FONT FACE=Symbol>Ô</FONT> and Supermagro used in organic agriculture and the neem oil (Azadirachta indica A. Juss) on the entomopathogenic fungi Metarhizium anisopliae and Beauveria bassiana was studied. These products were mixed in a medium where the two fungi were inoculated, and germination, vegetative growth and conidiogenesis were assessed. The biofertilizers Supermagro and E.M.- 4 showed to be less toxic for the two fungi whereas Multibion<FONT FACE=Symbol>Ô</FONT> caused major inhibition on M. anisopliae, with reductions in germination (-37.74%), colony diameter (-30.26%) and conidiogenesis (-42.62%). Neem oil promoted a larger negative effect on B. bassiana, inhibiting germination (-45.27%), colony diameter (-36.62%) and conidiogenesis (-84.93%).

Microbial control in integrated pest management (IPM) programs of coffee plantations is an important factor for the reduction of pest population densities. The use of selective pesticides can be associated with entomopathogens, increasing the efficiency of the control and reducing the use of required insecticides. The in vitro fungitoxic effect of insecticide formulations of Thiamethoxam, Cyfluthrin, Deltamethrin, Alpha-Cypermethrin, Triazophos, Chlorpyrifos, Fenpropathrin and Endosulfan and Beauveria bassiana (CG 425 strain) was evaluated at three concentrations (FR = average field recommendation; 0.5 ´ FR and 2 ´ FR). Effects of these products on conidia germination, vegetative growth and sporulation were compared. Only five insecticides, at the FR concentration, promoted conidia viability higher than 60%. Viability should be considered the most important factor to be evaluated since it is the first step of the infection process. The insecticide formulations of Alpha-Cypermethrin, Thiamethoxam and Cyfluthrin caused the lower inhibition level on conidia germination at the two lower concentrations, with no difference in relation to the control. With respect to vegetative growth analysis, Thiamethoxam at the two lower concentrations was not found to cause radial growth inhibition. Thiamethoxam caused the smallest inhibition level with regard to conidia production. The use of Alpha-Cypermethrin and Thiamethoxam formulations in coffee IPM programs for a B. bassiana inoculum conservation strategy are recommended, since these products were compatible with the entomopathogenic fungus Beauveria bassiana (CG 425), an important natural control agent of the coffee berry borer, Hypothenemus hampei.

Background: Alphitobius diaperinus, known as the lesser mealworm, is recognized as a problem for the poultry production sector. Several chemical methods have been employed in an attempt to control it, without successful results. Thus, alternative methods should be considered as an interesting approach to control this type of infestation. A recent study showed that glycerol monolaurate (GML) possesses insecticidal effect against bees. Therefore, the aim of this study was to evaluate, for the first time, the effect of GLM against the larval and adult forms of the lesser mealworm in vitro, and to verify whether GLM treated poultry litter used as soil fertilizer exerts any negative effect on its fauna.Materials, Methods & Results: In vitro tests were performed using three concentrations of GLM: 0.2, 0.4 and 1 mg mL-1, and the number of dead larvae and adults of A. diaperinus was counted on days 0, 2, 5 and 8 post-treatment. Poultry litter was pulverized with 1 mg mL-1 of GML, maintained under controlled conditions for 20 days and revolved every two days. Two ecotoxicological tests were performed using Enchytraeus crypticus. In the first test, GML was applied directly into the soil at doses of 0.5, 1 and 2 mg kg-1 of soil, while the second test used poultry litter as soil fertilizer at concentrations of 0, 2, 4 and 8 tons per hectare. Insecticidal action of GML against larvae and adults of A. diaperinus where only the 1 mg mL-1 concentration showed positive effect. GML caused 100% mortality of larvae two days after the beginning of treatment. Similarly, GML showed high efficacy to control adult forms of A. diaperinus, causing 83 and 90% of mortality on days 5 and 8 post-treatment, respectively. No difference was observed while applying GML directly at doses of 0.5, 1 and 2 mg kg-1 in the TAS when compared to the control group, similarly to what was observed using poultry litter containing 1 mg/m2 of GML at concentrations of 0, 2, 4 and 8 tons per hectare compared to the control group.Discussion: GML treatment exerted larvicidal and insecticidal action in vitro against A. diaperinus, similarly to what was observed by researchers against bees, where this product was able to eliminate 55% of the bees after 120 h of treatment. GML insecticidal mechanism of action is poorly investigated and remains unknown. However, there are evidences regarding its antimicrobial property using Staphylococcus aureus, Streptococcus spp., Enterococcus faecalis, as well as its capacity to inhibit some virulence factors linked to antimicrobial resistance. Some tests have been performed in order to reduce the use of chemical products, as Melaleuca alternifolia (tea tree oil), Ocotea odorifera (popularly known as sassafras) and Eucalyptus viminalis (popularly known as eucalypt) essential oils exerts insecticidal action against the larvae and adults of mealworms. The main chemical used to control A. diaperinus is cypermethrin, a chemical product that can damage the soil fauna due to its intensive utilization. The use of GML did not cause negative effects in the soil fauna, in disagreement other study with GML at concentrations of 50 and 100 µg mL-1 and found negative effects on the soil fauna using adults and juvenile forms of springtails (Folsomia candida) as biomarkers, showing a LD50 of 41 µg mL-1. Based on these evidences, we can suggest that the toxicity of GML to soil fauna is related to the biomarker used to determine the toxicity. Therefore, it is possible to conclude that GML possesses insecticidal action without toxic effects for the soil fauna, demonstrating potential as an alternative method to control the lesser mealworm.

Penelitian ini bertujuan mengetahui pertumbuhan dan perkembangan in vitro jamur Beauveria bassiana mutan serta virulensinya terhadap hama pengisap polong kedelai, R. linearis. Penelitian dilakukan di LaboratoriumBioteknologi Pertanian Fakultas Pertanian Universitas Lampung dan dilaksanakan bulan Januari - Juni 2017. Uji pertumbuhan B. bassiana secara in vitro menggunakan Rancangan Acak Lengkap (RAL) dengan 3 perlakuan dan 5 ulangan. Uji virulensi jamur B. bassiana terhadap R. linearis menggunakan analisis probit. Virulensi diindikasikan dengan LT 50 atau lethal time 50, yaitu waktu yang dibutuhkan jamur ini untuk mematikan 50% larva uji. Hasil penelitian menunjukkan bahwa isolat Bbyf22 dan Bbyf24 (mutan) mampu tumbuh dan berkembangdengan normal seperti isolat Bbyf (wildtype). Jamur B. bassiana terbukti virulen terhadap hama R. linearis dengan LT 50 = 3,7 hari (isolat Bbyf22, mutan); 4,9 hari (isolat Bbyf24, mutan); dan 3,5 hari (isolat Bbyf, wildtype).
