Penelitian ini bertujuan mengetahui pertumbuhan dan perkembangan in vitro jamur Beauveria bassiana mutan serta virulensinya terhadap hama pengisap polong kedelai, R. linearis. Penelitian dilakukan di LaboratoriumBioteknologi Pertanian Fakultas Pertanian Universitas Lampung dan dilaksanakan bulan Januari - Juni 2017. Uji pertumbuhan B. bassiana secara in vitro menggunakan Rancangan Acak Lengkap (RAL) dengan 3 perlakuan dan 5 ulangan. Uji virulensi jamur B. bassiana terhadap R. linearis menggunakan analisis probit. Virulensi diindikasikan dengan LT 50 atau lethal time 50, yaitu waktu yang dibutuhkan jamur ini untuk mematikan 50% larva uji. Hasil penelitian menunjukkan bahwa isolat Bbyf22 dan Bbyf24 (mutan) mampu tumbuh dan berkembangdengan normal seperti isolat Bbyf (wildtype). Jamur B. bassiana terbukti virulen terhadap hama R. linearis dengan LT 50 = 3,7 hari (isolat Bbyf22, mutan); 4,9 hari (isolat Bbyf24, mutan); dan 3,5 hari (isolat Bbyf, wildtype).

Kepik Hijau (Nezara viridula L.) merupakan hama penting bagi beberapa tanaman pangan seperti kedelai,  padi  dan  jagung.  Salah  satu  upaya  untuk  mengendalikan  hama  adalah  dengan pemanfaatan  entomopatogen,  seperti  virus,  bakteri  maupun  jamur.  Salah  satu  jamur entomopatogen  yang berpotensi untuk dieksploitasi adalah Beauveria bassiana yang mempunyai berbagai jenis inang. Belum diketahui patogenesitas B. bassiana yang diisolasi dari beberapa jenis inang terhadap hama kepik hijau (N. viridula), untuk itulah penelitian ini dilakukan dengan tujuan untuk mengetahui patogenesitas B. bassiana yang diisolasi dari   inang yang berbeda terhadap hama kepik hijau (N. viridula). Penelitian ini dilakukan di Laboratorium Agens Hayati (LAH) Kalasey, Balai Perlindungan dan Pengujian Mutu Tanaman Pangan dan Hortikultura (BPPMTPH), Dinas Pertanian dan Peternakan Daerah Provinsi Sulawesi Utara.  Penelitian dilakukan secara in- vitro dengan menggunakan Rancangan Acak Lengkap (RAL) dengan 6 perlakuan yaitu (Kontrol negatif , N1: suspensi B. bassiana isolat WBC (wereng batang coklat), N2: suspensi B. bassiana isolat Jati Sari sebagai kontrol positif (rekomendasi Kementan), N3: suspensi B. bassiana isolat Paraeucosmetus pallicornis (kepik hitam), N4: suspensi B. bassiana isolat Leptocorisa acuta (walang sangit) dan N5: suspensi B. bassiana isolat Scotinopara coarctata (kepinding tanah). Perlakuan menggunakan konsentrasi atau kerapatan konidia 109/ml air, dengan 5 ulangan. Data dianalisis dengan Uji Anava 95% dan analisis probit untuk menentukan LT50  (Lethal Time 50). Hasil penelitian menunjukkan bahwa jamur B. bassiana isolat Jati Sari memiliki patogenisitas tertinggi dengan membunuh kepik hijau (N. viridula) sampai 100% pada hari ke-5 HSP (hari setelah perlakuan), sedangkan isolat lain semuanya pada hari ke-6 HSP. Nilai  LT50  (waktu yang dibutuhkan untuk membunuh 50% serangga uji) terdapat pada isolat P. pallicornis yaitu  2,1 hari. Persentasi mortalitas pada hari ke-3 HSP tertinggi terdapat pada perlakuan isolat P. pallicornis dengan rata-rata mortalitas sebesar 78% diikuti isolat L. acuta dan S. coarctata masing-masing sebesar 68%   dan isolat Jati sari sebesar 58%, sedangkan mortalitas terendah terdapat pada perlakuan isolat WBC yaitu sebesar 32%. Tidak ada perbedaan patogenisitas yang signifikan diantara isolat Jati Sari, P. pallicornis, L. acuta maupun S. coarctata kecuali isolat WBC.Kata kunci: Beauveria bassiana; Entomopatogen; Nezara viridula Pathogenicity of Beauveria bassiana (Bals.) (Hemiptera: Pentatomidae) ABSTRACTGreen Ladybugs (Nezara viridula L.) are important pests for several food crops such as soybeans, rice and corn. One of the efforts to control this pest is the use of entomopathogens, such as viruses, bacteria and fungi. One of the entomopathogenic fungi that has the potential to be exploited is the fungus Beauveria bassiana. The fungi B. bassiana has various of hosts. There are no data on the pathogenicity  of  B.  bassiana  isolated  from  several  host  species  against  green  ladybugs  (Nviridula), for that reason this study was conducted with the aim of knowing the pathogenicity of B. bassiana isolated from different hosts against green ladybugs (N. viridula).. This research was conducted at Laboratory of Biological Control Agents, the Center for the Protection and Quality Testing of Food Crops and Horticulture, the Regional Agriculture and Livestock Service Office of North Sulawesi Province. The research was conducted in-vitro using a completely randomized design (CRD) with 6 treatments, namely (Negative control, N1: suspension B. bassiana, WBC isolate (brown planthopper), N2: B. bassiana suspension, Jati Sari isolate as a positive control (Ministry of Agriculture Recommendation), N3 : B. bassiana suspension isolate Paraeucosmetus pallicornis (black ladybug), N4: suspension B. bassiana isolate Leptocorisa acuta (Rice ear bug) and N5: suspension B. bassiana isolate Scotinopara coarctata (rice black bug) Treatment using conidia concentration or density 109  / ml water, with 5 replications. Data were analyzed with Anava test 95% and probit analysis to determine LT50 (Lethal Time 50). The results showed that the fungus B. bassiana Jati Sari isolate had the highest pathogenicity, because this isolate was able to kill green ladybugs (N viridula) to 100% on the 5th day of DAT (the day after treatment), while the other isolates were all on the 6th DAT. While the value of LT50  (time needed to kill 50% of the insect test) was found in P. pallicornis isolates which were 2,1 days of DAT. Because LT50 in all isolate treatments was between the 2nd and 3rd day of DAT, the Anava test for mortality was carried out on the 3rd day of DAT. The highest percentage of mortality on the 3rd day of DAT was found in the treatment of P. pallicornis isolates with an average mortality of 78% followed by L. acuta and S. coarctata isolates each at 68% and Jati sari isolates at 58%, while the lowest mortality was found in the treatment of WBC isolates, namely 32%. There was no significant difference in pathogenicity between Jati Sari, P. pallicornis, L. acuta and S. coarctata isolates except for WBC isolates.Keywords: Beauveria bassiana; Entomopatogen;  Nezara viridula

Ctenocephalides felis is an ectoparasitic flea species commonly found on dogs and cats. The current study verified the in vitro virulence of conidia of the entomopathogenic fungus Beauveria bassiana produced under different color LED light (red, blue, purple, green, yellow, and white) to adults of C. felis. The fungal isolates were cultivated on malt extract agar (MEA). Bioassay treatments used aerial conidia in test tubes. Adult fleas were obtained from a house cat in Chiang Mai province, Thailand. The experiments were composed of one control and eleven treatment groups. All of the treatments with B. bassiana conidia caused adult mortality after an exposure of 12 h. Among the conditions used in this study, B. bassiana cultured under red LED and fluorescent light were the most effective in causing mortality (100 %) in adult fleas after 36 h. The experimental results indicate that these aerial conidia of B. bassiana have promising potential for use in control of C. felis adult stages.

Virulence of entomopathogenic fungi <em>Metarhizium anisopliae</em> and <em>Beauveria bassiana</em> were tested against Arabian Rhinoceros Beetle, <em>Oryctes agamemnon arabicus</em> larvae. Four concentrations (1×10<sup>5</sup>, 1×10<sup>7</sup>, 1×10<sup>9</sup> and 1×10<sup>11</sup> conidia/mL<sup>–1</sup>) of two locally isolated entomopathogenic fungi spore suspensions were used in this study via larval direct spraying. Results revealed that both isolates can cause high mortality rate reaching 100% after 29 days. However, <em>Beauveria bassiana</em> scored higher mortality rate in short time especially at the concentration of 1×10<sup>11</sup> conidia/ mL<sup>–1</sup> with lethal time (LT)<sub>50</sub> 12.75 and LT<sub>90</sub> 20.00; while, <em>Metarhizium anisopliae</em> caused the higher percentage of malformed adults. Moreover, both isolates affected insect’s life cycle particularly in the pupal stage which was reduced remarkably by almost 50% in comparison with the control treatment.

O objetivo deste trabalho foi avaliar a eficiência de Beauveria bassiana (Bb), terra diatomácea (TD) e microrganismos eficazes (EM-4), associados ou não, no controle de Alphitobius diaperinus, e o efeito de EM-4 no desenvolvimento de B. bassiana. Os agentes de controle (Bb, TD e EM-4), em diferentes concentrações e combinações, foram aplicados em uma mistura de cama-de-frango e ração, em que os insetos foram acondicionados por dez dias. Avaliaram-se, in vitro: a germinação, as unidades formadoras de colônia (UFC), o crescimento vegetativo e a produtividade de conídios de Bb em contato com suspensão aquosa de EM-4 (1%) não filtrada e filtrada. Os maiores índices de mortalidade foram observados nos tratamentos TD + Bb + EM-4 e TD + Bb, nas maiores concentrações. Verificou-se efeito não-aditivo sinérgico para TD + Bb nas três concentrações. Nos testes com EM-4 filtrado, não houve diferença em relação à testemunha quanto à germinação e às UFC, entretanto, o crescimento vegetativo e a produtividade de conídios foram negativamente afetados. O uso conjunto de B. bassiana e terra diatomácea, para o manejo de populações de A. diaperinus, pode reduzir o uso de produtos químicos.

The in vitro fungitoxic effect of three biofertilizers, E.M.-4, Multibion <FONT FACE=Symbol>Ô</FONT> and Supermagro used in organic agriculture and the neem oil (Azadirachta indica A. Juss) on the entomopathogenic fungi Metarhizium anisopliae and Beauveria bassiana was studied. These products were mixed in a medium where the two fungi were inoculated, and germination, vegetative growth and conidiogenesis were assessed. The biofertilizers Supermagro and E.M.- 4 showed to be less toxic for the two fungi whereas Multibion<FONT FACE=Symbol>Ô</FONT> caused major inhibition on M. anisopliae, with reductions in germination (-37.74%), colony diameter (-30.26%) and conidiogenesis (-42.62%). Neem oil promoted a larger negative effect on B. bassiana, inhibiting germination (-45.27%), colony diameter (-36.62%) and conidiogenesis (-84.93%).

Microbial control in integrated pest management (IPM) programs of coffee plantations is an important factor for the reduction of pest population densities. The use of selective pesticides can be associated with entomopathogens, increasing the efficiency of the control and reducing the use of required insecticides. The in vitro fungitoxic effect of insecticide formulations of Thiamethoxam, Cyfluthrin, Deltamethrin, Alpha-Cypermethrin, Triazophos, Chlorpyrifos, Fenpropathrin and Endosulfan and Beauveria bassiana (CG 425 strain) was evaluated at three concentrations (FR = average field recommendation; 0.5 ´ FR and 2 ´ FR). Effects of these products on conidia germination, vegetative growth and sporulation were compared. Only five insecticides, at the FR concentration, promoted conidia viability higher than 60%. Viability should be considered the most important factor to be evaluated since it is the first step of the infection process. The insecticide formulations of Alpha-Cypermethrin, Thiamethoxam and Cyfluthrin caused the lower inhibition level on conidia germination at the two lower concentrations, with no difference in relation to the control. With respect to vegetative growth analysis, Thiamethoxam at the two lower concentrations was not found to cause radial growth inhibition. Thiamethoxam caused the smallest inhibition level with regard to conidia production. The use of Alpha-Cypermethrin and Thiamethoxam formulations in coffee IPM programs for a B. bassiana inoculum conservation strategy are recommended, since these products were compatible with the entomopathogenic fungus Beauveria bassiana (CG 425), an important natural control agent of the coffee berry borer, Hypothenemus hampei.

Research highlights: The pine-tree lappet moth, Dendrolimus pini, can cause serious needle defoliation on pines with outbreaks occurring over large geographical areas. Under laboratory conditions, the promising potential of the naturally occurring entomopathogenic fungus Beauveria bassiana was tested against D. pini larvae as a biological control method. Background and objectives: The aim of this study was to investigate the most effective concentration and treatment dose of B. bassiana conidial suspension and how it affected the survival and feeding behavior of the pest. Materials and methods: The first experiment applied the fungal suspension directly on the back of selected larvae, and in the second experiment, sporulating cadavers obtained in the first experiment were placed into Petri dishes with healthy individuals. Different doses per larvae [µL] and spore suspension concentration [spores/µL]) were used. The second experiment was designed to investigate the horizontal transmission of fungi by exposing individual caterpillars to a cadaver covered in B. bassiana mycelia. Mortality rates were analyzed by Chi-squared tests using absolute values for total mortality and B. bassiana- attributed mortality. The lethal time and feeding-disruption speed were analyzed with parametric and non-parametric tests with the aim to determine whether statistically significant differences were observed between treatments. Results: Analysis of lethal time revealed that the 20 µL dose and 7.9 × 104 concentration yielded highest mortality, but also the average dieback rate of 9.4 days was significantly faster compared to all other treatments. In order to see whether insects stopped feeding after infection with B. bassiana, larvae weight loss was examined. Results showed that effective treatments induced change in the feeding behavior of infected larvae. The feeding disruption caused by the 20 µL treatment occurred on average 5.5 days after exposure or 3.9 days before the final dieback of larvae.

Botrytis cinerea causes substantial losses in tomato and chili pepper crops worldwide. Endophytes have shown the potential for the biological control of diseases. The colonization ability of native endophyte strains of Beauveria bassiana and their antifungal effect against B. cinerea were evaluated in Solanaceae crops. Root drenching with B. bassiana was applied, and endophytic colonization capacity in roots, stems, and leaves was determined. The antagonistic activity was evaluated using in vitro dual culture and also plants by drenching the endophyte on the root and by pathogen inoculation in the leaves. Ten native strains were endophytes of tomato, and eight were endophytes of chili pepper. All strains showed significant in vitro antagonism against B. cinerea (30–36%). A high antifungal effect was observed, and strains RGM547 and RGM644 showed the lowest percentage of the surface affected by the pathogen. Native strains of B. bassiana colonized tomato and chili pepper tissues and provided important levels of antagonism against B. cinerea.
