In vitro propagation of Althaea rosea - a valuable medicinal plant of Kashmir Himalaya

Althaea rosea, commonly known as Hollyhock is an ornamental and medicinal plant. A. rosea has been reported to contain highest amount of tannins, carbohydrates, cyanide and mucilage. The purpose of this study was to optimize conditions for multiplication of Althaea rosea using shoottip explants. Varying concentrations solo and in combinations of auxins and cytokinins were experimented. The development of micro-propagation protocol for this valuable medicinal plant can ensure continuous supply of the plant product for pharmaceutical and food industry. In this regard an efficient and reproducible micro-propagation protocol has been developed using Murashige and Skoog’s (MS) (1962) medium augmented with different phytohormonal combinations like BAP, NAA, IBA and IAA. Both direct and indirect multiplication was obtained using in vitro raised shoot tip explants. Maximum shoot number was obtained using combination of BAP and NAA and rooting of isolated shoots was obtained using IBA. 60% of the transplanted plantlets survived under laboratory conditions.

Shoot tip explants from in vitro germinated seedlings of Solanum capsicoides All. inoculated on MS containing 2 mg/l BA produced maximum shoot induction response (26 shoots per explant). Rooting of the microshoots (19.4 roots per explant) was obtained better in half strength of MS supplemented with NAA (0.5 mg/l). Shoot tip explants of Algerian Ivy Heder a canariensis were cultured on MS basal medium supplemented with a combination of salt strength and NAA and IBA. More roots per explant developed on full salt strength medium combined with NAA. The most roots per explant were obtained with a combination of IBA and 1/4 MS salt. There was an inverse relationship between an increase in IBA or NAA concentration and root length and number. Shoots proliferated better on full MS salt combined with NAA and IBA. The highest level of NAA (40 uM) and 0.1 uM TDZ produced the most shoots and roots, the longest roots, the highest rooting percentage, the largest plants with the most leaves and the best callus quality per explant. The leaves from in vitro were cultured on MS medium with varying levels of Thidiazuron (TDZ) and NAA in the presence of light produced the highest number of roots.

A simple micropropagation method from nodal and shoot tip explants is reported here for Mentha viridis, an economically important medicinal plant. High frequency of microshoots was obtained from these two explants on MS supplemented with various concentrations of BAP (1.0 - 4.0 mg/l) and Kn (1.0 -4.0 mg/l). Maximum number of shoots was obtained from nodal explants in the medium containing 3.0 mg/l BAP. The root induction was carried out by using IBA and IAA (0.5 - 2.0 mg/l). Among these, high frequency of root proliferation was achieved in the medium containing 1.5 mg/l of IBA. The rooted plantlets were hardened and transferred to the field. Plantlets were regenerated from the shoot tip explants of Vernonia cinerea (L.) Less. in MS supplimented with BA and Kn. Maximum number of shoots in BA (13.32 mg/l) and roots in IBA (7.38 mg/l) developed. A procedure was developed forin vitropropagation ofCurculigo latifoliathrough shoot tip culture. Direct regeneration and indirect scalp induction ofCurculigo latifoliawere obtained from shoot tip grown on MS medium supplemented with different concentrations and combinations of thidiazuron and indole-3-butyric acid. Maximum response for direct regeneration in terms of percentage of explants producing shoot, shoot number, and shoot length was obtained on MS medium supplemented with combination of thidiazuron (0.5 mg L−1) and indole-3-butyric acid (0.25 mg L−1) after both 10 and 14 weeks of cultures. Indole-3-butyric acid in combination with thidiazuron exhibited a synergistic effect on shoot regeneration. The shoot tips were able to induce maximum scalp from basal end of explants on the medium with 2 mg L−1thidiazuron. Cultures showed that shoot number, shoot length, and scalp size increased significantly after 14 weeks of culture. Transferring of the shoots onto the MS medium devoid of growth regulators resulted in the highest percentage of root induction and longer roots, while medium supplemented with 0.25 mg L−1IBA produced more numbers of roots.

Shoot multiplication of Bupleurum distichophyllum was achieved from the nodal and shoot tip explants of mature plants using MS with different concentrations and combinations of growth regulators. Maximum explant response was from axillary shoots and the highest number of shoots per explant was obtained on MS fortified with 1.0 mg/l BAP. The highest degree of axillary shoot proliferation was found to be 74 and 70% for nodal- and shoot tip explants, respectively on the medium containing 1.0 mg/l BAP + 0.1 mg/l NAA. The combination of BAP and GA3 was also found to be effective for both type of explants. The degree of shoot formation was affected by explant types and the exogenous hormonal regime in the medium. The regenerated shoots were successfully rooted on MS supplemented with 2.0 mg/l IBA, after sequential hardening, survival rate was 71%.
