Evaluation of Four Analytical Methods to Detect Weevils in Wheat: Granary Weevil, Sitophilus granarius (L.), in Soft White Wheat

Soft white wheat was infested with adult granary weevils under controlled conditions at 3 levels of infestation, and samples were removed for testing 6 times over a total of 48 d. Three analytical methods available for use in FDA laboratories were compared along with a simple counting of the visually-detectable, insect-damaged kernels in a 100-g sample. The objective of the study was to determine how the analytical results compared with one another and with the levels of infestation. Data confirmed that the more rapid visual exam of wheat is not reliable for indicating the hidden, internally-developing weevils. Analysts should be aware of the possibility that wheat inspected visually could be passable with respect to insect-damaged kernels, but still may produce violative flour due to high insect fragment counts when processed. More research is needed to correlate insect infestation in wheat with insect fragment counts in flour. The two methods which could be used to estimate potential flour contamination caused by weevils are the x-ray exam and the cracking and flotation method, since both of these reveal the stages of weevil development inside the kernels. Tables present the results of the analyses and relate them to the number and sex of the founding adults in each level of infestation.

Almond, as with other stone fruit, possesses a highly lignified endocarp or shell. The dominant hard-shelled trait (D-) is positively associated with greater resistant to insect infestation than nuts expressing the paper-shelled (dd) trait. Hard-shelled genotypes have undesirable effects, including a lower kernel meat-to-nut crack-out ratio, greater kernel damage during mechanical shelling, and a reduction in plant energy available to kernel development. Histogenic analysis shows that the almond endocarp, unlike peach, has a tri-partite structure. Insect feeding studies have subsequently demonstrated that the inner endocarp layer, which is similar in both hard and paper-shelled types, is the most important structural barrier to insect infestation. Shell-seal integrity and X-ray studies have confirmed that discontinuities at the inner endocarp suture seal are the primary, though not the sole site of entry for insect pests. Paper-shelled almond selections with highly lignified and well-sealed inner endocarps show resistance levels comparable to hard shelled types but with crack-out ratios 30% to 40% higher. Pseudo-paper-shelled types have also been selected, in which a highly lignified outer endocarp is formed, but is retained by the fruit hull at dehiscence. An understanding of endocarp morphology and development is thus important in breeding for insect resistance as well as the commercial utilization of both kernel and hull.

Carbon steel is a preferred construction material in many industrial and domestic applications, including oil and gas pipelines, where corrosion mitigation using film-forming corrosion inhibitor formulations is a widely accepted method. This review identifies surface analytical techniques that are considered suitable for analysis of thin films at metallic substrates, but are yet to be applied to analysis of carbon steel surfaces in corrosive media or treated with corrosion inhibitors. The reviewed methods include time of flight-secondary ion mass spectrometry, X-ray absorption spectroscopy methods, particle-induced X-ray emission, Rutherford backscatter spectroscopy, Auger electron spectroscopy, electron probe microanalysis, near-edge X-ray absorption fine structure spectroscopy, X-ray photoemission electron microscopy, low-energy electron diffraction, small-angle neutron scattering and neutron reflectometry, and conversion electron Moessbauer spectrometry. Advantages and limitations of the analytical methods in thin-film surface investigations are discussed. Technical parameters of nominated analytical methods are provided to assist in the selection of suitable methods for analysis of metallic substrates deposited with surface films. The challenges associated with the applications of the emerging analytical methods in corrosion science are also addressed.

To solve the multiple practical aspects raised by the achieving of cereals stocks protection (especially wheat and corn) against granary weevil Sitophilus granarius L., the research aimed a complex study on the sexual dimorphism at this species, aspect which help to prevent the weevils damages using unpollutant methods (with the aid of sexual pheromones, ovogenesis inhibitors, etc.). Due to the summarily data in the scientific literature of the world refering to the sexual dimorphism of the species, a detailed study was made within the Entomology Laboratory at the Faculty of Agriculture of USAMV Cluj-Napoca and at the Center of Electronical Microscopy of Babeş-Bolyai University in Cluj-Napoca (Romania), during 2005-2009
