Objective: The objective of this search was to synthesize a new naproxen analogues having a 1,2,4-triazole-3-thiol heterocyclic ring, and preliminary pharmacological assessment of the anti-inflammatory activity of the synthesized compounds. Methods: The synthesis of naproxen analogues that having 1,2,4-triazole-3-thiol heterocyclic ring occur through esterification of naproxen, and then its reaction with hydrazine hydrate, and carbon disulfide, finally different aromatic aldehydes reacted with triazole derivatives of naproxen containing amino group to produce schiff bases.Results: In vivo acute anti-inflammatory activity of the synthesize compounds (Va-Vd) was evaluated in rats using egg-white induced edema model of inflammation in a dose equivalent to (50 mg/kg) of naproxen. All tested compounds were produced a significant reduction in paw edema with respect to the effect of propylene glycol 50% v/v (control group). Compound Vd produced superior anti-inflammatory activity compared to naproxen.Conclusion: The results obtained in this work give evidence about the valid synthesis of 1,2,4 triazole-3-thiol derivatives of naproxen, which reacted with different aldehydes to yield several schiff bases. The incorporation of benzaldehyde possess para-electron donating group (para-hydroxyl benzaldehyde) will increase the anti-inflammatory activity of naproxen.

This research includes the synthesis of some new different heterocyclic derivatives of 5-Bromoisatin. New sulfonylamide, diazine, oxazole, thiazole and 1,2,3-triazole derivatives of 5-Bromoisatin have been synthesized. The synthesis process started by the reaction of 5-Bromoisatin with different reagents to obtain schiff bases of 5-Bromoisatin intermediate compounds(1, 8, 19) by using glacial acetic acid as a catalyst in three routes. The first route, 5-Bromoisatin reacted with p-aminosulfonylchloride to product compound(1), then converted to sulfonyl amide derivatives(2-7) by the reaction of compound(1) with different substituted primary aromatic amine in absolute ethanol. The second route includes the reaction of 5-Bromoisatin reacted with ethyl glycinate to give 5-bromo-3-(Ethyl imino acetate)-2-oxo indole(8), which undergo react with hydrazine hydrate 80% to obtain hydrazine derivatives(9) that react with different acid anhydrides to obtain diazine derivatives(10-14). Also compound(8) reacts with urea and thiourea to give compounds(15,16) which undergo cyclization with p-bromophenacylbromide in absolute ethanol as a solvent to obtain oxazole (17) and thiazole (18), respectively. The third route included the reaction of 5-Bromoisatin with p-phenylenediamine in ethanol to obtain compound(19) which is converted to new substitutes 1,2,3-triazole derivatives(22,23) by diazotation of compound(19) and treating the resulted salt(20) with sodium azid, then acetylaceton or ethylacetoacetate, respectively. Newly synthesized compounds were identified by spectral methods. (FTIR, 1H-NMR, 13C-NMR) and measurements of some of its physical properties and also some specific reactions. Furthermore the effects of the synthesized compounds were studied on some strains of bacteria.

The major products in the controlled benzylation (NaH/PhCH2Cl) of allyl and but-3-enyl β-D-glucopyranosides were the 2,4,6-tri-O-benzyl ethers. Subsequent glycosidations of these tribenzyl ethers gave derivatives of allyl and but-3-enyl β- laminaribioside, the latter being subsequently transformed into 3,4-epoxybutyl β- laminaribioside.

An alternate synthetic route to the important anticancer drug suberoylanilide hydroxamic acid (SAHA) from its α,ß-didehydro derivative is described. The didehydro derivative is obtained through a cross metathesis reaction between a suitable terminal alkene and N-benzyloxyacrylamide. Some of the didehydro derivatives of SAHA were preliminarily evaluated for anticancer activity towards HeLa cells. The administration of the analogues caused a significant decrease in the proliferation of HeLa cells. Furthermore, one of the analogues showed a maximum cytotoxicity with a minimum GI50 value of 2.5 µg/mL and the generation of reactive oxygen species (ROS) as some apoptotic features.

CD data of all four diastereoisomers of the permethylaryl ether 3- O-acetyl derivatives of a series of flavan-3-ols permit assignment of the absolute configuration at the stereocentres of the heterocyclic ring.
