Soybean mosaic virus (SMV) is one of the most destructive pathogens of soybean (Glycine max (L.) Merr.) worldwide. In this study, 184 F7:11 recombinant inbred line (RIL) populations derived from Kefeng No. 1 × Nannong 1138-2 were used to study the inheritance and linkage mapping of resistance genes against SMV strains SC7 and SC13 in Kefeng No. 1. Two independent dominant genes (designated Rsc7 and Rsc13) that control resistance to SC7 and SC13 were located on a molecular linkage group (MLG) of chromosome 2 (D1b). A mixed segregating population was developed by self-pollination of three heterozygous plants of residual heterozygous lines (RHL3-27, RHL3-30, RHL3-53) with five markers linked to the loci, and was used in fine-mapping of Rsc7 and Rsc13. In addition, Rsc7 was fine-mapped between BARCSOYSSR_02_0667 and BARCSOYSSR_02_0670 on MLG D1b. The genetic distance between the two closest markers was 0.7 cM and the physical distance of the interval was ~77 kb, which included one LRR gene and another gene containing an F-box region. Two SSR markers (BARCSOYSSR_02_0610 and BARCSOYSSR_02_0621) were closely linked to the SC13 resistance gene. The physical distance where Rsc13 was located was ~191 kb. Sequence analysis showed that there were two K-box region types of transcription factor genes; GmHSP40 and two serine/threonine protein kinase (STK) genes were the most likely candidate genes. These results will facilitate map-based cloning of the Rsc7 and Rsc13 genes and development of transgenic disease-resistant varieties, and will provide SMV-resistance breeding systems with excellent resistance germplasm.

Heritability, potential ratio, and heterosis of soybean (Glycine max [L.] Merrill) resistance to soybean mosaic virus. The use of soybean cultivars with resistance to SMV is a way for controlling soybean mosaic disease. The objective of this research was to estimate the disease severity, the narrow sense heritability, potential ratio and heterosis of resistance character and number of pithy pods, number of healthy seeds, and healthy seeds weight per plant of ten F1 populations of soybean crossing result to SMV infection. The experiment was arranged in a randomized complete block design in two replications. Observed characters were disease severity, number of pithy pods, number of healthy seeds, and healthy seeds weight per plant. The result of this research showed that 1) the crossing combinations those which were resistant to SMV (lower disease severity) were Yellow Bean x Tanggamus, Tanggamus x Orba, and Tanggamus x Taichung, 2) the narrow sense heritability of disease severity was included in medium criteria, 3) number of pithy pods belonged to high criteria, and 4) number of healthy seeds and healthy seeds weight per plant were included in low criteria. The crossing combinations that had low estimation value of heterosis and heterobeltiosis of resistance to SMV infection were Yellow Bean x Taichung, Bean x Tanggamus and Tanggamus x B3570. Disease severity or resistance to SMV is influenced by genetic and environmental factors.

Phytophthora root and stem rot, caused by Phytophthora sojae, is an important disease of soybean (Glycine max L.) in South Dakota. Because P. sojae populations are highly diverse and resistance genes deployed in commercial soybean varieties often fail to manage the disease, this study was initiated to determine P. sojae pathotype distribution in South Dakota. A total of 216 P. sojae isolates were baited from soil collected from 422 soybean fields in South Dakota in 2013-2015 and 2017. The pathotype of each isolate was determined by inoculating 10 seedlings of 13 standard soybean P. sojae differential lines using the hypocotyl inoculation technique. Of the 216 pathotyped isolates, 48 unique pathotypes were identified. The virulence complexity of isolates ranged from virulence on one Rps gene (Rps7) to virulence on 13 Rps genes and mean complexity was 5.2. Harosoy (Rps7), Harlon (Rps1a), Williams 79 (Rps 1c), William 82 (Rps1k), Harosoy 13XX (Rps1b), were susceptible to 98, 80, 78, 73, 72% of the isolates, respectively. These results highlight the highly diverse P. sojae pathotypes in South Dakota and the likely Rps genes to fail in commercial soybean varieties
