The calcitonin gene in the thyroid parafollicular cells generates two different mRNAs, which encode for the precursors of calcitonin and calcitonin gene-related peptide (CGRP). In this study, we investigated whether these two peptides are released simultaneously in the perfused dog thyroid. The isolated dog thyroid (n = 10) was once-through perfused with a Krebs-Ringer bicarbonate buffer. After elevation of the buffer Ca2+ concentration from 1.5 to 2.5 mM, a rapid, marked, and parallel secretion of both calcitonin and CGRP occurred (P less than 0.001). By stepwise increase of the buffer Ca2+ concentration, it was found that, although an elevation from 1.5 to 1.65 mM had no influence on the thyroid output of calcitonin or CGRP, elevations to 1.75, 2.0, and 2.5 mM of Ca2+ induced dose-dependent parallel increases in the thyroid output of calcitonin and CGRP. Furthermore, the stimulation with pentagastrin (100 pM) increased the thyroid output of both calcitonin and CGRP in parallel. It is concluded that the normal dog thyroid releases calcitonin and CGRP in parallel on stimulation with Ca2+ and pentagastrin.

By use of appropriate fragments of CT DNA or a CGRP DNA and SP6 polymerase system, we produced anti-sense RNA probes labeled with biotinylated 11-UTP. The labeling and specificity of the RNA probes were confirmed using dot-blot hybridization. By use of hybridocytochemistry, CT mRNA and CGRP mRNA were localized in all parafollicular cells in control and dihydrotachysterin-pre-treated rats. We concluded that all parafollicular cells simultaneously produce both CT mRNA and CGRP mRNA, either under control conditions or after stimulation by dihydrotachysterin-induced hypercalcemia.

This protocol describes the process of using Neurolucida 360 software to map the topographical organization of Calcitonin gene related peptide – immunoreactive axons and terminals in the muscular layer of mice stomach. Stomachs were removed, layers were separated and gone under immunohistochemistry as whole mounts, then scanned using confocal microscopy.

ZUSAMMENFASSUNGDie Charakterisierung von calcitonin gene-related peptide (CGRP) als Schlüsselmolekül in der Pathophysiologie der Migräne hat nicht nur unser Verständnis der Erkrankung, sondern auch die Entwicklung neuer Therapien vorangetrieben. Seit kurzem steht mit den monoklonalen Antikörpern gegen CGRP oder den CGRP-Rezeptor eine spezifische und hoch selektive Option für die medikamentöse Prophylaxe der episodischen und chronischen Migräne zur Verfügung, die in zahlreichen klinischen Studien ihre Überlegenheit gegenüber Placebo belegt hat. Hier werden Erfahrungen aus dem praktischen Behandlungsalltag zur kurz- und mittelfristigen Wirksamkeit und Verträglichkeit mitgeteilt und weitere Aspekte wie Therapiewechsel bei Non-Response, Verlauf nach Therapieende und die Frage des Wirkungsortes unter Einbeziehung bereits publizierter Daten angesprochen.
