Succinylcholine (succinyldicholine), a drug used to relax skeletal muscles, can be partially hydrolyzed to yield a mixture containing parent compound, succinyl-monocholine, choline, and succinic acid. Resolution of this mixture has been accomplished by paper chromatography. Thirty solvent systems were screened; ten separated all four compounds, while others achieved partial resolution. Kraut–Dragendorf and potassium iodoplatinate reagents were found to be useful to locate the compounds on the paper.

Hydrolysis of N' -(3-N-methylcarbamoyloxyphenyl)-N,N-dimethylformamidine (I) has been studied in mixture water-dioxane (4 : 1) at pH 1 to 13. The hydrolysis rates of methylcarbamoyl and dimethylformamidine groups are comparable within pH range 4 to 10, and they differ by as much as several orders of magnitude in pH ranges 1-3 and 11-13. The hydrolysis products of the whole pH range have been determined by paper chromatography, and reaction mechanism has been suggested on the basis of the measured hydrolysis rate constants.Effects of protonation and hydratation of dimethylformamidine group on the hydrolysis rate of the methylcarbamoyl group is discussed.

The method of cleavage of cellotriose by Myrothecium cellulase was determined from the distribution of C14 in the enzymic hydrolysis products of cellotriose labelled in the anhydroglucose unit at the reducing end of the chain. The substrate was prepared by the incubation of the enzyme in a concentrated solution of cellopentaose (0.14 M) and C14-labelled glucose (5 M) and the fractionation of the products, first by adsorption and elution on a charcoal column and then by preparative paper chromatography. The glucosidic linkage adjacent to the non-reducing end of the chain is preferentially hydrolyzed by the enzyme. Its rate of hydrolysis was estimated to be about five times that for the linkage adjacent to the reducing end of the chain.

SummaryThe investigations were carried out with a newly syn-thesised, original 99mTc-Unithiol complex (Unithiol-2,3-dimercaptopropansulphonate). It accumulates in the kidneys and enables their scintigraphic examination at 90—180 minutes after the administration. The study was performed on rats, rabbits and dogs. 99mTc-Unithiol complex accumulation in rat kidneys was 34,9%, 44,5% and 67,8% of dose after 1, 2 and 3 h respectively.Good reability of the scintigraphic picture of the kidneys in rabbits and dogs confirms the accumulative capability of the complex in these organs. It was found by means of ascending paper chromatography that the purity of the 99mTc-Unithiol complex exceeded 90%.

A simple and specific method using paper chromatography has been developed to detect the unconjugated cortisol in urine.

ABSTRACT Perfusion of rat liver with vanillic acid yielded only one metabolite. In paper chromatography with three different solvent systems, the substance showed the same RF-values as vanillyolglycine (3-methoxy-4-hydroxyhippuric acid) and in mixed chromatograms there was only one single spot. After separation by column chromatography, the UV- and IRspectra of the reaction product were identical with those of 3-methoxy4-hydroxy-hippuric acid. During the perfusion experiment, the kinetics of the conjugation were investigated.
