The thiosulfate–citrate–bile salts–sucrose agar (TCBS) used in the most-probable-number method for detecting Vibrio parahaemolyticus cannot differentiate growth of V. parahaemolyticus from Vibrio vulnificus or Vibrio mimicus. This study examined the selectivity and specificity of Bio-Chrome Vibrio medium (BCVM), a chromogenic medium that detects V. parahaemolyticus on the basis of the formation of distinct purple colonies on the medium. A panel consisting of 221 strains of bacteria, including 179 Vibrio spp. and 42 non-Vibrio spp., were examined for their ability to grow and produce colored colonies on BCVM. Growth of Salmonella, Shigella, Escherichia coli, Enterobacter cloacae, Yersinia enterocolitica, and Aeromonas was inhibited by both BCVM and TCBS. All 148 strains of V. parahaemolyticus grew on BCVM, and 145 of them produced purple colonies. The remaining 31 Vibrio spp., except one strain of Vibrio fluvialis, were either unable to grow or produced blue-green or white colonies on BCVM. Bio-Chrome Vibrio medium was capable of differentiating V. parahaemolyticus from other species, including V. vulnificus and V. mimicus. Further studies are needed to evaluate the sensitivity and specificity of BCVM for detecting V. parahaemolyticus in foods.

Budidaya udang vaname (Litopenaus vanamei) secara intensif telah dilakukan di kabupaten Kendal, sebagai upaya peningkatan produksi. Salah satu kendala yang sering terjadi pada budidaya udang intensif adalah serangan penyakit bakteri, termasuk pula vibriosis. Penelitian ini bertujuan untuk mengkaji keanekaragaman bakteri yang berasosiasi dengan udang yang terserang vibriosis secara intensif beserta gejala klinisnya. Udangvaname yang menunjukkan gejala vibriosis diperoleh dari pertambakan intensif di kabupaten Demak. Sebanyak 22 isolat bakteri (SMC 01 – SSD 10) berhasil diisolasi dari hepatopacreas udangvaname pada mediumThiosulfate Citrate Bile Salts Sucrose (TCBS). Berdasarkan perfomance morphologi dari keua puluh dua isolat (bentuk, warna dan karakter koloni) dipilih 5 isolat ( SMC 01,SMC 04, SSD 01, SSD 03, dan SSD 07) untuk uji selanjutnya yaitu karakterisasi secara morfologi dan uji biokimia . Uji sensitivitas terhadap antibiotik yang beredar hanya dilakukan pada 3 isolat terpilih(SMC 01, SMC 04, dan SSD 03). Gejala klinis udang yang terserang vibriosis adalah : tubuh (carapace) memerah, melanosis pada kulit, nekrosis pada ekor, kaki renang dan kaki jalan memerah serta hepatopankreas yang memerah cenderung gelap. Hasil karakterisasi dengan pendekatan secara morfologi dan biokimia diperoleh pula bahwa bakteri yang berasosiasi dengan udangvaname (L. vanamei) dari tambak intensif kabupaten Kendal adalah Vibrio vulnificus (SMC 01), V. mimicus (SMC 04), V.damsella (SSD 01), V. parahaemolytics ( SSD 03) , V. fluvialis (SSD 07). Hasil Uji sensitivitas menunjukkan bahwa ketiga isolat terpilih (SMC 01, SMC 04, dan SSD 03) tidak sensitif terhadap beberapa antibiotik..Kata kunci: vibriosis, sensitifitas, salinitas, udangvaname.

The worldwide distribution of qnr genes found on plasmids and their presence on the chromosomes of aquatic bacteria, like Vibrio vulnificus, one of the suspected sources, suggests an origin before the development of synthetic quinolones. However, their native function remains unknown. Previous work indicated that expression of qnrVv in V. vulnificus was induced by cold shock. To investigate its role further we constructed single in-frame deletion mutants in qnrVv and cspA (the gene for cold shock protein), and a double mutant in qnrVv and cspA in V. vulnificus ATCC 17562 to evaluate the response to different environmental conditions and stresses and to exposure to various DNA damaging agents. We found that qnrVv is involved in resistance to ciprofloxacin, levofloxacin, and mitomycin C, and in the cold shock response in V. vulnificus. Moreover, ΔqnrVv and ΔcspA mutants showed a lower growth when they were treated with bile salts at 37°C and then shifted to 15°C (cold shock) without bile salts in the media, with the effect being higher in the double mutant. This transition may mimic what happens when V. vulnificus is ingested into the gastrointestinal tract and released in its natural environment. Cold shock and bile salts induced the expression of cspA and DNA gyrase and topoisomerase IV genes. However, no induction was found in the ΔqnrVv mutant, suggesting that the qnrVv gene is involved in the response to DNA damage and nucleic acid secondary structure.

ABSTRACT An improved selective medium, cellobiose-colistin (CC) agar, gave a significantly higher (P < 0.05) isolation rate ofVibrio vulnificus from water and sediment samples than did modified cellobiose-polymyxin B-colistin (mCPC) agar. In a total of 446 alkaline peptone water preenrichments amended with polymyxin B,V. vulnificus was isolated from 154 preenrichments (35%) with mCPC agar and from 179 preenrichments (40%) with CC agar. CC agar gave a higher plating efficiency of V. vulnificuscells than did cellobiose-polymyxin B-colistin (CPC) agar, mCPC agar, or thiosulfate-citrate-bile salts-sucrose (TCBS) agar; the only significant difference was observed with TCBS agar, which gave much lower plating efficiencies than the other selective media. Determination of MICs demonstrated that the concentrations of colistin and polymyxin B in CPC agar inhibit growth of a proportion ofV. vulnificus strains.
