Selection of Beauveria bassiana isolates to control Alphitobius diaperinus

ABSTRACT: The fruit borer Conotrachelus humeropictus is one of the most important pests of the cupuaçu crop, and can promote yield losses of around 50%. The present study involved the selection of Beauveria bassiana isolates on Conotrachelus psidii larvae, which was used as test insect due to its taxonomic closeness to C. humeropictus. In order to assess their interaction using combined application, we studied the ability of the nematode to carry conidia of B. bassiana and Metarhizium anisopliae to soil layers. This study also aimed to establish a biological control method for cupuaçu borer with the use of the fungus B. bassiana at 10 kg/ha and the entomopathogenic nematode Steinernema brazilense at 1, 3, 9, and 27 IJ/cm2, applied as single and combined treatments. In the selection of B. bassiana strains for C. psidii, the IBCB 276 and IBCB 165 isolates were the most promising ones, causing 86 and 84% mortality, respectively. The IBCB 276 strain was applied in the field to control C. humeropictus. In the study of carriage of conidia in soil, the nematode S. brazilense was found to be capable of carrying conidia of B. bassiana to deep 7 to 10 cm soil layers. This was not observed with M. anisopliae, found only in the topsoil. In field trials against C. humeropictus larvae, B. bassiana promoted 15.6% mortality. The nematode promoted 60.0% mortality at the highest concentration used, and their combined use with B. bassiana (10 kg/ha) increased mortality to 65.6%, with an additive interaction observed between the two entomopathogens.

The cotton leafworm, Alabama argillacea (Huebner, 1818) (Lepidoptera: Noctuidae), is considered to be one of the key pests in herbaceous cotton (Gossypium hirsutum L. r. latifolium Hutch) cropping, with constant occurrence in all cotton-growing states of Brazil. In this study Metarhizium anisopliae and Beauveria bassiana isolates were screened and evaluated for pathogenicity against Alabama argillaceae. Initially, a screening of ten isolates of each fungus in a concentracion of 10(8) conidia mL-1, was carried out on 3rd instar larvae of A. argillacea. Further studies were conducted to determine the pathogenicity and virulence of six and seven isolates of M. anisopliae and B. bassiana, respectively, against 3rd instar larvae of A. argillacea and using the concentrations of 10(6), 10(7), 10(8), and 10(9) conidia mL-1. The experiments were carried out in Recife, PE, Brazil, at 27 ± 2ºC, RH 70 ± 5% and a photophase of 12 hours. Mortalities caused by M. anisopliae isolate at the different concentrations ranged from 4.5 to 91.2%, the highest mortality percentage being found for the isolate 1189 at 10(9) conidia mL-1. The isolate 645 of B. bassiana caused the highest mortality at the highest concentration, followed by isolates 634, 604, and IPA 198. The lowest lethal time for B. bassiana and M. anisopliae, was achieved by the isolates 483 (4.1 days) and 1189 (2.0 days), respectively. The isolates 1189, 1022 e 866 of M. anisopliae and 483, IPA198 and 604 of B. bassiana, at 10(8) e 10(9) conidia mL-1 are promissing for use the integrated control of A. argillacea larvae, but M. anisopliae seems more effective.

O objetivo deste trabalho foi avaliar a eficiência de Beauveria bassiana (Bb), terra diatomácea (TD) e microrganismos eficazes (EM-4), associados ou não, no controle de Alphitobius diaperinus, e o efeito de EM-4 no desenvolvimento de B. bassiana. Os agentes de controle (Bb, TD e EM-4), em diferentes concentrações e combinações, foram aplicados em uma mistura de cama-de-frango e ração, em que os insetos foram acondicionados por dez dias. Avaliaram-se, in vitro: a germinação, as unidades formadoras de colônia (UFC), o crescimento vegetativo e a produtividade de conídios de Bb em contato com suspensão aquosa de EM-4 (1%) não filtrada e filtrada. Os maiores índices de mortalidade foram observados nos tratamentos TD + Bb + EM-4 e TD + Bb, nas maiores concentrações. Verificou-se efeito não-aditivo sinérgico para TD + Bb nas três concentrações. Nos testes com EM-4 filtrado, não houve diferença em relação à testemunha quanto à germinação e às UFC, entretanto, o crescimento vegetativo e a produtividade de conídios foram negativamente afetados. O uso conjunto de B. bassiana e terra diatomácea, para o manejo de populações de A. diaperinus, pode reduzir o uso de produtos químicos.
