Comparison of bacterial communities associated withXestospongia testudinaria, sediment and seawater in a Singaporean coral reef ecosystem

Despite alterations caused by anthropogenic activities in Singaporean coral reefs, the sponge communities are quite diverse andXestospongia testudinariais one of the most common sponge species. In the present study, we used 16S rRNA gene barcoded pyrosequencing to characterize and compare bacterial communities from different biotopes (sponge, seawater and sediment) and to identify dominant bacterial symbionts ofX. testudinariain a Singaporean coral reef ecosystem. Our results showed that biotope appears to affect the richness, composition and abundance of bacterial communities. Proteobacteria was the most abundant phylum in sediment and seawater whilst Chloroflexi was more abundant inX. testudinaria.Members of the order Caldilineales (fermentation of organic substrates), Chromatiales (purple sulphur bacteria), Rhodospirillales (purple non-sulphur bacteria) and Syntrophobacterales (sulphate-reducing bacteria) were relatively more abundant inX. testudinariasamples.

Abstract Herbivorous fishes play important ecological roles in coral reefs by consuming algae that can otherwise outcompete corals, but we know little about the gut microbiota that facilitates this process. This study focussed on the gut microbiota of an ecologically important coral reef fish, the convict surgeonfish Acanthurus triostegus. We sought to understand how the microbiome of this species varies along its gastrointestinal tract and how it varies between juvenile and adult fish. Further, we examined if the bacteria associated with the diet consumed by juveniles contributes to the gut microbiota. 16S rRNA gene amplicon sequencing showed that bacterial communities associated with the midgut and hindgut regions were distinct between adults and juveniles, however, no significant differences were seen for gut wall samples. The microbiota associated with the epilithic algal food source was similar to that of the juvenile midgut and gut wall but differed from the microbiome of the hindgut. A core bacterial community including members of taxa Epulopiscium and Brevinemataceae was observed across all gastrointestinal and diet samples, suggesting that these bacterial symbionts can be acquired by juvenile convict surgeonfish horizontally via their diet and then are retained into adulthood.

Composition and distribution of foraminifers are affected by human activities and have close association with coral reef ecosystem. The aims of this research were to investigate the benthic foraminifers’ composition and distribution in sediment of coral reef ecosystem. Eleven stations of Karang Bongkok, Pramuka, and Onrust Island were observed in this study. The sediments were taken from surface substrate up to 2 cm under the substrate. Samples were washed on sieve with mesh size 0,063 mm, and then dried in oven with 50°C of temperature  for two hours. After separating from the sediment, the foraminifers were laid on foraminiferal slide and indentified using binocular microscope. The highest composition of symbiont-bearing foraminiferal assemblages which associated with reef ecosystem was in East Pramuka (78.17%) and the lowest was in South Onrust (21,83%). The opportunistic type had the highest composition in South Onrust (38.67%) and the lowest was in South Karang Bongkok. In west Pramuka had the highest composition of heterotrophic type (57.17%) and the lowest was in North Onrust (11.33%). Onrust Island was dominated by opportunistic type, indicating high nutrient. The highest amount of foraminifers’ taxa was found in Karang Bongkok with good coral reef coverage, while the lowest in Onrust facing with Jakarta Bay.
