Salinibacillus xinjiangensis sp. nov., a halophilic bacterium from a hypersaline lake

A Gram-positive, endospore-forming, rod-shaped bacterium, designated isolate J4T, was isolated from a neutral saline lake sample from Xinjiang Uyghur Autonomous Region, China, and subjected to a polyphasic taxonomic investigation. Phylogenetic analysis based on 16S rRNA gene sequence revealed that strain J4T is most closely related to Salinibacillus aidingensis 25-7T (with 96.7 % similarity), Salinibacillus kushneri 8-2T (96.5 %), Ornithinibacillus scapharcae TW25T (96.4 %), Salirhabdus euzebyi CVS-14T (96.4 %) and Ornithinibacillus californiensis MB-9T (96.2 %). Chemotaxonomic analysis showed menaquinone-7 (MK-7) to be the major isoprenoid quinone of strain J4T; diphosphatidylglycerol and phosphatidylglycerol were the major cellular polar lipids and the cell wall contained meso-diaminopimelic acid as the diagnostic diamino acid. The major cellular fatty acids were iso-C15 : 0 and anteiso-C15 : 0. The genomic DNA G+C content of strain J4T was determined to be 36.2 mol%. Strain J4T was positive for catalase activity and negative for oxidase activity. Strain J4T was observed to grow at 25–50 °C (optimal 35–42 °C), pH 6.5–8.0 (optimal 7.0–7.5) and in media containing 1–21 % (w/v) NaCl (optimal 9–12 %). Based on these data, strain J4T represents a novel species of the genus Salinibacillus and the name Salinibacillus xinjiangensis sp. nov. is proposed. The type strain is J4T ( = CGMCC 1.12331T = JCM 18732T).

A novel Gram-positive, slightly halophilic bacterium, designated strain B48T, was isolated from soil around the hypersaline lake Aran-Bidgol in Iran and characterized taxonomically using a polyphasic approach. Cells of strain B48T were non-motile rods and produced ellipsoidal endospores at a central or subterminal position in swollen sporangia. Strain B48T was a strictly aerobic bacterium, catalase- and oxidase-positive. The strain was able to grow at NaCl concentrations of 0.5–10.0 % (w/v), with optimum growth occurring at 2.5 % (w/v) NaCl. The optimum temperature and pH for growth were 35 °C and pH 7.5–8.0, respectively. On the basis of 16S rRNA gene sequence analysis, strain B48T was shown to belong to the genus Bacillus within the phylum Firmicutes and showed the closest phylogenetic similarity to the species Bacillus foraminis CV53T (97.4 %) and Bacillus purgationiresistens DS22T (96.9 %). The DNA G+C content of this new isolate was 40.1 mol%. The major cellular fatty acids of strain B48T were iso-C15 : 0 and anteiso-C15 : 0, and its polar lipid pattern consisted of diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, an aminophospholipid and two unknown phospholipids. The only quinone present was menaquinone 7 (MK-7). The peptidoglycan contained meso-diaminopimelic acid as the diagnostic diamino acid. All these features confirm the placement of isolate B48T within the genus Bacillus . DNA–DNA hybridization experiments revealed a low level of relatedness between strain B48T and Bacillus foraminis IBRC-M 10625T (8.1 %). On the basis of polyphasic evidence from this study, a new species of the genus Bacillus , Bacillus persicus sp. nov., is proposed, with strain B48T ( = IBRC-M 10115T = DSM 25386T = CECT 8001T) as the type strain.

Phylogenetic analysis based on 16S rRNA gene sequences revealed that strain A24T belonged to the genus Bacillus within the phylum Firmicutes and showed the closest phylogenetic similarity with the species Bacillus alkalitelluris BA288T (97.2 %), Bacillus herbersteinensis D-1,5aT (96.0 %) and Bacillus litoralis SW-211T (95.6 %). The G+C content of the genomic DNA of this strain was 35.9 mol%. The polar lipid pattern of strain A24T consisted of phosphatidylglycerol, diphosphatidylglycerol, phosphatidylethanolamine and two unknown phospholipids. The major cellular fatty acids of strain A24T were anteiso-C15 : 0 and iso-C15 : 0. The respiratory quinones were MK-7 (94 %) and MK-6 (4 %). The peptidoglycan contained meso-diaminopimelic acid as the diagnostic diamino acid. All these features confirm the placement of isolate A24T within the genus Bacillus . DNA–DNA hybridization experiments revealed a relatedness of 8 % between strain A24T and Bacillus alkalitelluris IBRC-M 10596T, supporting its placement as a novel species. Phenotypic characteristics, phylogenetic analysis and DNA–DNA relatedness data suggest that this strain represents a novel species of the genus Bacillus , for which the name Bacillus salsus sp. nov. is proposed. The type strain is strain A24T ( = IBRC-M 10078 T = KCTC 13816T).

A facultatively anaerobic, Gram-stain-positive, rod-shaped bacterium, designated strain KB0549T, was isolated from sesame oil cake. Cells were motile, round-ended rods, and produced central or terminal spores. The cell wall peptidoglycan contained meso-diaminopimelic acid as the diamino acid. The major fatty acids were anteiso-C15 : 0 and anteiso-C17 : 0. The DNA G+C content of strain KB0549T was 51.9 mol%. On the basis of 16S rRNA gene sequence phylogeny, strain KB0549T was affiliated with the genus Paenibacillus in the phylum Firmicutes and was most closely related to Paenibacillus cookii with 97.4 % sequence similarity. Strain KB0549T was physiologically differentiated from P. cookii by the high content of anteiso-C17 : 0, inability to grow at 50 °C, spore position, and negative Voges–Proskauer reaction. Based on these unique physiological and phylogenetic characteristics, it is proposed that the isolate represents a novel species, Paenibacillus relictisesami sp. nov.; the type strain is KB0549T ( = JCM 18068T = DSM 25385T).

A facultatively alkaliphilic actinomycete strain, designated EGI 80088T, was isolated from a saline-alkali soil sample from Xinjiang province, north-west China, and subjected to a polyphasic taxonomic characterization. Strain EGI 80088T formed fragmented aerial hyphae and short spore chains, and rod-like spores aggregated at maturity. Whole-cell hydrolysates of the isolate contained ll-diaminopimelic acid as the diagnostic diamino acid, and glucosamine, mannose, galactose, glucose and rhamnose as the marker sugars. The major fatty acids identified (>5 %) were anteiso-C15 : 0, iso-C15 : 0, summed feature 4 (iso-C17 : 1I/anteiso-C17 : 1B), iso-C16 : 0 and anteiso-C17 : 0. The predominant menaquinone was MK-9(H4). The G+C content of the genomic DNA of strain EGI 80088T was 70.6 mol%. EGI 80088T showed the highest 16S rRNA gene sequence similarity to its closest phylogenetic neighbour Haloactinopolyspora alba YIM 93246T (98.5 %). The DNA–DNA relatedness value of the strain EGI 80088T and H. alba YIM 93246T was 59.3±5.2 %. On the basis of morphological, chemotaxonomic and phylogenetic characteristics and DNA–DNA hybridization data, strain EGI 80088T represents a novel species of the genus Haloactinopolyspora , for which the name Haloactinopolyspora alkaliphila sp. nov. (type strain EGI 80088T = BCRC 16946T = JCM 19128T) is proposed. The description of the genus Haloactinopolyspora has also been emended.

An aerobic, Gram-stain-positive, endospore-forming, rod-shaped and moderately halophilic strain SKP4-6T, was isolated from shrimp paste (Ka-pi) collected from Samut Sakhon Province, Thailand. Phylogenetic analysis revealed that strain SKP4-6T belonged to the genus Halobacillus and was most closely related to Halobacillus salinus JCM 11546T (98.6 %), Halobacillus locisalis KCTC 3788T (98.6 %) and Halobacillus yeomjeoni KCTC 3957T (98.6 %) based on 16S rRNA gene sequence similarity. The digital DNA–DNA hybridization (dDDH) and average nucleotide identity (ANI) values between strain SKP4-6T and its related species were 18.2–19.3 % and 69.84–84.51 %, respectively, which were lower than the threshold recommended for species delineation. The strain grew optimally at 30–40 °C, at pH 7.0 and with 10–15 % (w/v) NaCl. It contained l-Orn–d-Asp in the cell wall peptidoglycan. The DNA G+C content was 44.8 mol%. The major fatty acids were iso-C15 : 0, anteiso-C15 : 0 and anteiso-C17 : 0. The predominant isoprenoid quinone was MK-7. Phosphatidylglycerol and diphosphatidylglycerol were present as major polar lipids. Based on this polyphasic approach, digital DNA–DNA relatedness and ANI values, strain SKP4-6T represents a novel species of the genus Halobacillus , for which the name Halobacillus fulvus sp. nov. is proposed. The type strain is SKP4-6T (=JCM 32624T=TISTR 2595T).

A Gram-staining-positive, aerobic organism, isolated from the Red Sea sponge Spheciospongia vagabunda was investigated to determine its taxonomic position. On the basis of results of 16S rRNA gene sequence analysis strain EG49T was most closely related to Actinokineospora cibodasensis and Actinokineospora baliensis (both 97.3 % similarity) and Actinokineospora diospyrosa and Actinokineospora auranticolor (both 97.0 % similarity). The 16S rRNA gene sequence similarity to all other species of the genus Actinokineospora was <97.0 %. The quinone system of strain EG49T contained the menaquinones MK-9(H4) (47 %), MK-9(H6) (27 %) and MK-9(H2) (15 %) in major amounts. Minor amounts of MK-7(H4) (2 %), MK-9(H0) (1 %), MK-9(H8) (3 %) and MK-10(H4) (3 %) were detected as well in addition to MK-8(H4), MK-8(H6), MK-10(H2) and MK-10(H6) (all <1 %). The diagnostic diamino acid of the peptidoglycan was meso-diaminopimelic acid. In the polar lipid profile, diphosphatidylglycerol, phosphatidylethanolamine and hydroxyphosphatidylethanolamine were predominant. Phosphatidylinositol-mannoside, two unidentified phospholipids and two glycoglipids as well as one aminoglycolipid, one aminolipid and one unidentified lipid were found in addition. The fatty acid profile was composed of mainly iso-branched fatty acids: iso-C16 : 0, iso-C14 : 0, iso-C15 : 0 and iso-C16 : 1H. All these findings clearly supported the classification of the strain as representing a member of the genus Actinokineospora . In addition, the results of physiological and biochemical tests also allowed phenotypic differentiation of strain EG49T from the most closely related species of the genus Actinokineospora . Strain EG49T represents a novel species of the genus Actinokineospora , for which we propose the name Actinokineospora spheciospongiae sp. nov., with strain EG49T ( = DSM 45935T = CCM 8480T = LMG 27700T) as the type strain.

The isoprenoid quinones were MK-7 (98 %) and MK-8 (2 %). Strain G8BT contained a peptidoglycan of type A4β, l-Orn–d-Asp. All these features confirmed the placement of isolate G8BT within the genus Ornithinibacillus . DNA–DNA hybridization experiments revealed a low level of relatedness (6 %) between strain G8BT and Ornithinibacillus bavariensis DSM 15681T. On the basis of evidence from this study, a novel species of the genus Ornithinibacillus , Ornithinibacillus halophilus sp. nov., is proposed, with strain G8BT ( = IBRC-M 10683T = KCTC 13822T) as the type strain.

Strain VN07A0015T was isolated from soil collected on Cat Ba Island, Vietnam. The taxonomic position of strain VN07A0015T was near Streptomyces aomiensis M24DS4T (98.5 % 16S rRNA gene sequence similarity) and Streptomyces scabrisporus NBRC 100760T (95.6 %), and it clustered within them; however, this cluster was distant from the type strains of other species of the genus Streptomyces . The aerial mycelia of strain VN07A0015T were greyish and formed imperfect spiral spore chains (retinaculiaperti type) with smooth-surfaced spores. The morphological features of strain VN07A0015T were different from those of the type strains of S. aomiensis and S. scabrisporus . The chemotaxonomic characteristics of strain VN07A0015T were typical for all members of the genus Streptomyces , which possessed ll-type diaminopimelic acid, menaquinone MK-9(H6, H8) and the major fatty acids iso-C16 : 0 and iso-C15 : 0. DNA–DNA relatedness between strain VN07A0015T and S. aomiensis NBRC 106164T was less than 30 %. In addition, some physiological and biochemical traits differed from those of S. aomiensis . Therefore, we propose that strain VN07A0015T be classified in the genus Streptomyces as a representative of Streptomyces catbensis sp. nov. (type strain VN07A0015T = VTCC-A-1889T = NBRC 107860T).

Cells of strain X4BT were motile rods and formed ellipsoidal endospores at a terminal or subterminal position in swollen sporangia. Strain X4BT was a strictly aerobic bacterium, catalase- and oxidase-positive. The strain was able to grow at NaCl concentrations of 0.5–22.5 % (w/v), with optimum growth occurring at 7.5 % (w/v) NaCl. The optimum temperature and pH for growth were 35 °C and pH 7.0. Analysis of 16S rRNA gene sequence revealed that strain X4BT is a member of the family Bacillaceae , constituting a novel phyletic lineage within this family. Highest sequence similarities were obtained with the 16S rRNA gene sequences of the type strains of Sediminibacillus albus (96.0 %), Paraliobacillus ryukyuensis (95.9 %), Paraliobacillus quinghaiensis (95.8 %) and Sediminibacillus halophilus (95.7 %), respectively. The DNA G+C content of this novel isolate was 35.2 mol%. The major cellular fatty acids of strain X4BT were anteiso-C15 : 0 and anteiso-C17 : 0 and its polar lipid pattern consisted of diphosphatidylglycerol, phosphatidylglycerol, two aminolipids, an aminophospholipid and an unknown phospholipid. The isoprenoid quinones were MK-7 (89 %) and MK-6 (11 %). The peptidoglycan contained meso-diaminopimelic acid as the diagnostic diamino acid. On the basis of 16S rRNA gene sequence analysis in combination with chemotaxonomic and phenotypic data, strain X4BT represents a novel species in a new genus in the family Bacillaceae , order Bacillales for which the name Saliterribacillus persicus gen. nov., sp. nov. is proposed. The type strain of the type species (Saliterribacillus persicus) is X4BT ( = IBRC-M 10629T = KCTC 13827T).

The taxonomic position of a novel actinomycete isolated from soil in Fars Province (Iran) was determined using a polyphasic approach. Phenotypic characterization and 16S rRNA gene sequence analysis of the isolate matched those described for members of the genus Streptomyces . On ISP2 medium, strain HM 1154T produced a dark cream, branched substrate mycelium and Retinaculiaperti aerial hyphae that in some images also appeared spiral and that developed into greyish-white spore chains with a smooth surface. The isolate showed optimal growth at 28 °C and pH 6–9 with 0–4 % (w/v) NaCl. Whole-cell hydrolysates contained ll-diaminopimelic acid as diagnostic diamino acid, ribose and glucose. The main phospholipids were phosphatidylethanolamine, diphosphatidylglycerol, phosphatidylinositol, three unknown phospholipids and an unknown aminophospholipid; MK-9(H4) and MK-9(H2) were the predominant menaquinones. The major cellular fatty acids were the branched saturated iso-C16 : 0 and anteiso-C15 : 0. Strain HM 1154T exhibited the highest 16S rRNA gene sequence similarities to Streptomyces coerulescens DSM 40146T (99.4 %), Streptomyces varsoviensis DSM 40346T (99.3 %), Streptomyces youssoufiensis DSM 41920T (99.2 %), Streptomyces abikoensis DSM 40831T (99.2 %), Streptomyces rimosus subsp. rimosus DSM 40260T (99.1 %), Streptomyces luteireticuli DSM 40509T (99.1 %), Streptomyces thioluteus DSM 40027T (99.1 %), Streptomyces blastmyceticus DSM 40029T (99.0 %) and Streptomyces hiroshimensis DSM 40037T (99.0 %). DNA–DNA hybridization studies showed relatedness values of 11.0–35.8 % with the closest related species. Based on these results, strain HM 1154T is considered to represent a novel species within the genus Streptomyces , for which the name Streptomyces zagrosensis sp. nov. is proposed. The type strain is HM 1154T ( = DSM 42018T = UTMC 1154T = CECT 8305T).
