Flavobacterium solisilvae sp. nov. and Flavobacterium silvaticum sp. nov., isolated from forest soil

Two Gram-stain-negative, yellow-pigmented and strictly aerobic bacteria, designated strains SE-s27T and SE-s28T, were isolated from forest soil. Both strains were non-motile rods that were catalase-positive and oxidase-negative and grew optimally at 25–30 °C, pH 8.0 and with 0 % (w/v) NaCl. Strain SE-s28T produced flexirubin-type pigments, but strain SE-s27T did not produce them. Both strains contained menaquinone-6 as the sole respiratory quinone and phosphatidylethanolamine as a major polar lipid. As the major cellular fatty acids (>10 %), SE-s27T contained iso-C15 : 1 and iso-C15 : 1G, whereas SE-s28T contained iso-C15 : 0 and summed feature 3 comprising C16 : 1ω7c and/or C16 : 1ω6c and/or iso-C15 : 0 2-OH. The DNA G+C contents of strains SE-s27T and SE-s28T were 33.1 and 44.3 mol%, respectively. The results of phylogenetic analysis based on 16S rRNA gene sequences revealed that SE-s27T and SE-s28T formed respective distinct phylogenetic lineages within the genus Flavobacterium . Strains SE-s27T and SE-s28T were most closely related to Flavobacterium macrobrachii an-8T and Flavobacterium piscinae ICH-30T with 98.0 and 94.5 % 16S rRNA gene sequence similarities, respectively. In conclusion, strains SE-s27T and SE-s28T represent novel species of the genus Flavobacterium , for which the names Flavobacterium solisilvae sp. nov. and Flavobacterium silvaticum sp. nov. are proposed. The type strains of F. solisilvae and F. silvaticum are SE-s27T (=KACC 18802T=JCM 31544T) and SE-s28T (=KACC 18803T=JCM 31545T), respectively.

A strictly aerobic, Gram-staining-negative, rod-shaped, non-motile, yellow-pigmented bacterial strain, designated BX12T, was isolated from soil collected from the peak area of Wudang Mountain in the city of Shiyan, Hubei province, China. Phylogenetic analysis of the 16S rRNA gene sequences showed that strain BX12T was most closely related to Flavobacterium subsaxonicum WB 4.1-42T (95.9 % 16S rRNA gene sequence similarity), followed by Flavobacterium beibuense F44-8T (95.6 %) and Flavobacterium rivuli WB 3.3-2T (94.1 %). The major fatty acids (≥5 %) of strain BX12T were summed feature 3 (comprising C16 : 1ω7c and/or C16 : 1ω6c), iso-C15 : 0, C16 : 0, iso-C17 : 0 3-OH, and C16 : 0 3-OH. The major polar lipid was phosphatidylethanolamine, and the major respiratory quinone was menaquinone-6. The genomic DNA G+C content was 43.9 mol%. On the basis of a high number of phenotypic differentiating properties and phylogenetic uniqueness, strain BX12T represents a novel species of the genus Flavobacterium for which the name Flavobacterium hauense sp. nov. is proposed. The type strain is BX12T ( = CCTCC AB 2012197T = KCTC 32147T). Emended descriptions of Flavobacterium subsaxonicum , Flavobacterium beibuense and Flavobacterium rivuli are also proposed.

A Gram-negative, strictly aerobic, yellow-pigmented rod, designated DK69T, was isolated from soil collected from the waste liquid treatment facility of Bafeng Pharmaceutical Company in the city of Enshi, Hubei Province, China. Phylogenetic analysis based on 16S rRNA gene sequences placed strain DK69T in the genus Flavobacterium of the family Flavobacteriaceae . The highest 16S rRNA gene sequence similarities were found with Flavobacterium cauense R2A-7T (96.9 %), Flavobacterium saliperosum AS 1.3801T (96.3 %) and Flavobacterium suncheonense GH29-5T (95.7 %). The major fatty acids (≥5 %) were iso-C15 : 0, iso-C17 : 1ω9c, C15 : 0, iso-C17 : 0 3-OH and iso-C15 : 0 3-OH. The major polar lipids were phosphatidylethanolamine, one unidentified aminolipid and one unidentified lipid. The major respiratory quinone was menaquinone-6. The genomic DNA G+C content was 34.4 mol%. Strain DK69T represents a novel species of the genus Flavobacterium , for which the name Flavobacterium enshiense sp. nov. is proposed. The type strain is DK69T ( = CCTCC AB 2011144T  = KCTC 23775T). Emended descriptions of the genus Flavobacterium and Flavobacterium cauense , Flavobacterium saliperosum and Flavobacterium suncheonense are also proposed.

A novel bacterium, designated strain F051-1T, isolated from a seawater sample collected from the coast at Damupo beach in Pohang, Korea, was investigated in a polyphasic taxonomic study. Cells were yellow-pigmented, strictly aerobic, Gram-staining-negative and rod-shaped. The temperature, pH and NaCl ranges for growth were 4–30 °C, pH 6.0–9.0 and 1.0–6.0 % (w/v), respectively. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain F051-1T belongs to the genus Psychroserpens in the family Flavobacteriaceae . Its closest relatives were Psychroserpens burtonensis ACAM 188T (96.8 % 16S rRNA gene sequence similarity) and Psychroserpens mesophilus KOPRI 13649T (95.7 %). The major cellular fatty acids were iso-C15 : 0, iso-C15 : 1 G and anteiso-C15 : 0. The polar lipid profile consisted of a mixture of phosphatidylethanolamine, two unidentified aminolipids, one unidentified phospholipid and eight unidentified lipids. The major respiratory quinone was menaquinone-6 and the genomic DNA G+C content of the strain was 33.5 mol%. On the basis of phenotypic, phylogenetic and genotypic data, strain F051-1T represents a novel species within the genus Psychroserpens , for which the name Psychroserpens damuponensis sp. nov. is proposed. The type strain is F051-1T ( = KCTC 23539T  = JCM 17632T).

Three Gram-stain-negative, strictly aerobic, non-motile and rod-shaped bacterial strains, designated as HME9299T, HMF7410T and HMF7856T, were isolated from freshwater and tree bark collected in Yong-in, Republic of Korea. Strains HME9299T, HMF7410T and HMF7856T exhibited the highest 16S rRNA gene sequence similarities of 97.2, 94.4 and 96.4 % to Mucilaginibacter daejeonensis Jip 10T, Mucilaginibacter terrae CCM 8645T and Mucilaginibacter phyllosphaerae PP-F2F-G21T, respectively. Among themselves, the values were 94.1–95.7 %. Phylogenetic analysis of the 16S rRNA gene sequences of the three isolates revealed that they belonged to the genus Mucilaginibacter within the family Sphingobacteriaceae . The predominant fatty acids of three strains were summed feature 3 (comprising C16 : 1  ω7c and/or C16 : 1  ω6c) and iso-C15 : 0. Strain HME9299T contained a relatively large amount of C16 : 1  ω5c. The predominant respiratory quinone was menaquinone-7. The genome sizes of strains HME9299T, HMF7410T and HMF7856T were 4.33, 4.16 and 3.68 Mbp, respectively, and their DNA G+C contents were 41.6, 38.4 and 43.9 mol%, respectively. Based on the results of the phenotypic, genotypic, chemotaxonomic and phylogenetic investigation, three novel species, Mucilaginibacter aquatilis sp. nov, Mucilaginibacter arboris sp. nov. and Mucilaginibacter ginkgonis sp. nov., are proposed. The type strains are HME9299T (=KCTC 42122T=DSM 29146T), HMF7410T (=KCTC 62464T=NBRC 113227T) and HMF7856T (=KCTC 72782T=NBRC 114275T), respectively.

A soil bacterium, designated ZX9611T, was isolated from Taihang Mountain in Henan province, PR China. The strain was Gram-stain-negative and strictly aerobic. The cells were motile, rod-shaped and formed light pink-colored colonies. The 16S rRNA gene sequence of ZX9611T shared the highest similarities with those of Sphingomonas crocodyli CCP-7T (97.0%), Sphingomonas jatrophae S5-249T (96.6%) and Sphingomonas starnbergensis 382T (95.9%). Phylogenetic analyses based on 16S rRNA gene sequences demonstrated that ZX9611T clustered with S. crocodyli CCP-7T, S. jatrophae S5-249T and S. starnbergensis 382T. The average nucleotide identity (ANI) values between ZX9611T and two type strains ( S. crocodyli BCRC 81096T and S. jatrophae DSM 27345T) were 88.3 and 68.6% respectively. ZX9611T exhibited genome-sequence-based digital DNA–DNA hybridization (dDDH) values of 53.3 % and 15.3 %, compared with S. crocodyli BCRC 81096T and S . jatrophae DSM 27345T, respectively. ZX9611T had a genome size of 4.12 Mb and an average DNA G+C content of 64.8 %. ZX9611T had major fatty acids (>5 %) including summed feature 8 (C18 : 1  ω7c and/or C18 : 1  ω6c), C14 : 0 2-OH, C16 : 0 and summed feature 3 (C16 : 1  ω7c and/or C16 : 1  ω6c), and the major polyamine was sym-homospermidine. The only respiratory quinone was ubiquinone-10. The polar lipids were diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylglycerol, phosphatidylcholine and sphingoglycolipid. On the basis of phenotypic, chemotaxonomic and phylogenetic characteristics, strain ZX9611T represents a novel species of genus Sphingomonas, for which the name Sphingomonas montanisoli sp. nov. is proposed. The type strain is ZX9611T (=KCTC 72622T=CCTCC AB 2019350T).

A yellowish-pigmented bacterium, designated strain PLGR-1T, was isolated from the root of Bergenia scopulosa collected from Taibai Mountain in Shaanxi Province, north-west China, and was subjected to a taxonomic study by using a polyphasic approach. Cells of strain PLGR-1T were Gram-stain-negative, strictly aerobic, rod-shaped, non-spore-forming and motile with a single polar flagellum. Growth occurred at 7–33 °C (optimum, 25–28 °C), at pH 5.0–10.0 (optimum, pH 6.0–7.0) and with 0–0.5 % (w/v) NaCl (optimum, 0 %). The predominant respiratory quinone was ubiquinone-8 (Q-8) and the major cellular fatty acids were summed feature 3 (comprising C16 : 1ω7c and/or C16 : 1ω6c), C16 : 0 and summed feature 8 (comprising C18 : 1ω7c and/or C18 : 1ω6c). The major polyamines were putrescine and 2-hydroxyputrescine and the major polar lipids were diphosphatidylglycerol, phosphatidylethanolamine and phosphatidylglycerol. The DNA G+C content was 69.8 mol%. Phylogenetic analyses based on 16S rRNA gene sequences showed that strain PLGR-1T belonged to the class Betaproteobacteria and formed a tight phyletic lineage with members of the genus Rhizobacter . Strain PLGR-1T was most closely related to Rhizobacter dauci DSM 11587T and Rhizobacter fulvus DSM 19916T, with 16S rRNA gene sequence similarities of 98.5 and 98.0 %, respectively. The DNA–DNA relatedness values between strain PLGR-1T and the type strains of Rhizobacter dauci and Rhizobacter fulvus were 46.3 and 14.7 %, respectively. Based on the phenotypic, phylogenetic and genotypic data, strain PLGR-1T is considered to represent a novel species of the genus Rhizobacter , for which the name Rhizobacter bergeniae sp. nov. is proposed. The type strain is PLGR-1T ( = CCTCC AB 2013018T = KCTC 32299T = LMG 27607T).

A Gram-stain-positive, strictly aerobic, yellow colony-forming bacterium, designated strain 3-3T, was isolated from forest soil of Bac Kan Province in Vietnam. Cells were non-motile rods without gliding motility, showing oxidase- and catalase-positive reactions. Growth was observed at 20–37 °C (optimum, 28 °C) and pH 5.5–9.5 (optimum, pH 7.5). The major cellular fatty acids were iso-C15 : 0, iso-C15 : 1 G and summed feature 3 (comprising C16 : 1ω6c and/or C16 : 1ω7c). Strain 3-3T contained phosphatidylethanoamine, three unidentified aminolipids and three unidentified lipids. The G+C content of the genomic DNA was 49.5 mol% and the only isoprenoid quinone detected was menaquinone 7 (MK-7). Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain 3-3T formed a tight phylogenetic lineage with Flavihumibacter petaseus T41T with a bootstrap value of 100 %. Strain 3-3T was related most closely to F. petaseus T41T with 97.3 % 16S rRNA gene sequence similarity and the level of DNA–DNA relatedness between the two was 9.4±1.2 %. Based on phenotypic, chemotaxonomic and molecular features, strain 3-3T represents a novel species of the genus Flavihumibacter , for which the name Flavihumibacter solisilvae sp. nov. is proposed. The type strain is 3-3T ( = KACC 17917T = JCM 19891T).

A novel Gram-stain-negative, strictly aerobic, coccoid and agar-hydrolysing bacterium, designated RQJ05T, was isolated from the marine red algae Gelidium amansii collected from the coastal area of Rizhao, PR China. Cells of strain RQJ05T were approximately 0.8–1.0×1.3–3.0 µm in size and motile by means of a polar flagellum. Growth occurred at 4–33 °C (optimum, 25–30 °C), pH 7.0–8.5 (optimum, pH 7.5–8.0) and in the presence of 1.0–7.0 % (w/v) NaCl (optimum, 2.0–3.0 %). Strain RQJ05T showed oxidase-positive and catalase-negative activities. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain RQJ05T formed a phylogenetic lineage with members of the family Alteromonadaceae and exhibited 16S rRNA gene sequence similarities of 92.6, 91.3, 90.2 and 90.1 % to Catenovulum maritimum Q1T, Catenovulum agarivorans YM01T, Paraphotobacterium marinum NSCS20N07DT and Algicola sagamiensis B-10-31T, respectively. The major cellular fatty acids of strain RQJ05T were summed feature 3 (C16 : 1  ω7c and/or C16 : 1  ω6c), C16 : 0 and summed feature 8 (C18 : 1  ω7c and/or C18 : 1  ω6c). The major polar lipids of strain RQJ05T were phosphatidylethanolamine, phosphatidylglycerol and two aminophospholipids. Strain RQJ05T contained Q-8 as the major respiratory quinone. The genomic DNA G+C content was 39.0 mol%. On the basis of genotypic, phenotypic and phylogenetic evidence, strain RQJ05T is presented as a representative of a novel species in a new genus, for which the name Algibacillus agarilyticus gen. nov., sp. nov. is proposed. The type strain is RQJ05T (=KCTC 62846T=MCCC 1H00352T).

A novel type of freshwater bacterium was isolated from the prealpine mesotrophic Starnberger See (Bavaria, southern Germany). Cells of strain 382T were Gram-negative and rod-shaped, motile and creamy-white. The isolate was strictly aerobic, catalase- and oxidase-positive, and grew at pH values of 6–9 (optimum, pH 7) and temperatures of 10–37 °C (optimum, 28 °C). The genomic G+C content of strain 382T was 64.1 mol%. Based on 16S rRNA gene sequence analyses, strain 382T belongs to the family Sphingomonadaceae and clusters within the genus Sphingomonas . Sphingomonas histidinilytica UM 2T and Sphingomonas wittichii DSM 6014T were the closest relatives, as indicated by the highest 16S rRNA gene sequence similarities (97.1 % and 96.8 %, respectively). Sphingomonas paucimobilis DSM 1098T (the type species of the genus Sphingomonas ) exhibited 95.3 % sequence similarity. This affiliation of strain 382T to the genus Sphingomonas is confirmed by the presence of Q-10 as the major respiratory quinone, two sphingoglycolipids, C14 : 0 2-OH as the major 2-hydroxy fatty acid and sym-homospermidine as the major polyamine. The main cellular fatty acids of strain 382T were C18 : 1ω7c (39 %), C16 : 1ω7c (21 %), C16 : 0 (10 %) and C14 : 0 2-OH (10 %). Based on the phylogenetic distance from other species of the genus Sphingomonas and its unusually high C16 : 1ω7c content, strain 382T represents a novel species of the genus Sphingomonas , for which the name Sphingomonas starnbergensis is proposed. The type strain is 382T ( = DSM 25077T  = LMG 26763T).

A Gram-stain-negative, non-motile, rod-shaped bacterial strain, named SJ-16T, was isolated from desert soil collected in Inner Mongolia, northern PR China. Strain SJ-16T grew at pH 6.0–11.0 (optimum, pH 8.0–9.0), 4–40 °C (optimum, 30–35 °C) and in the presence of 0–8 % (w/v) NaCl (optimum, 0–2 %). The strain was negative for catalase and positive for oxidase. Phylogenetic analyses based on 16S rRNA gene sequences showed that strain SJ-16T clustered with Luteimonas chenhongjianii 100111T and Luteimonas terrae THG-MD21T, and had 98.8, 98.6, 98.3 and <97.9 % of 16S rRNA gene sequence similarity to strains L. chenhongjianii 100111T, L. terrae THG-MD21T, L. aestuarii B9T and all other type strains of the genus Luteimonas , respectively. The major cellular fatty acids were iso-C15 : 0, iso-C16 : 0, summed feature 3 (C16 : 1  ω7c and/or C16 : 1  ω6c) and summed feature 9 (C16 : 0 10-methyl and/or iso-C17 : 1  ω9c). Diphosphatidylglycerol, phosphatidylglycerol and phosphatidylethanolamine were the major polar lipids, and ubiquinone-8 was the only respiratory quinone. The genomic DNA G+C content was 69.3 mol%. The digital DNA–DNA hybridization and average nucleotide identity values of strain SJ-16T to L. chenhongjianii 100111T, L. terrae THG-MD21T, L. rhizosphaerae 4-12T and L. aestuarii B9T were 36.9, 37.5, 24.0 and 21.1 %, and 80.9, 80.6, 80.7 and 76.3 %, respectively. Based on phenotypic, physiological and phylogenetic results, strain SJ-16T represents a novel species of the genus Luteimonas , for which the name Luteimonas deserti is proposed. The type strain is SJ-16T (=CGMCC 1.17694T=KCTC 82207T).
