In root explants of a long-day plant, Cichorium intybus L., grown in vitro, inflorescences were produced under short-day conditions (9 h) if long days were applied on days 10 to 18 of culture. Long days consisted of either 16 h fluorescent light (5.6 W∙m−2) or daily cycles of 9 h white fluorescent plus 15 h red (660 nm, 0.3 W∙m−2) or blue (440 nm, 0.5 W∙m−2) light. The structural changes in the meristems of buds regenerated under these conditions were studied. The influence of photoperiod appeared to be critical between the 10th and 16th days, during the progressive transition from vegetative to prefloral stage, which occurred more rapidly under photoperiodic conditions with red or blue light. In noninductive short days, intermediate meristems could be observed tardily on day 24, but no flower formation took place after transfer to long days. Increasing the quantity of light under short-day conditions up to a level comparable to long-day conditions resulted in a rise in mitotic activity, mainly in the peripheral zone of the meristem, but the vegetative zonation was retained. These data therefore indicate that the duration of light really commits the meristem to the subsequent formation of flower primordia; in contrast, the quantity of light controls the bud formation during the first part of development. Several selected S. armeria lines differing in their reaction to GA3 were treated with GA3 at various concentrations under short-day (SD) or long-day conditions. With SD treatment one application of GA3 at high concentration (10 000 p.p.m. or greater) induced flower formation in certain lines. Stem elongation increased with GA3 concentration and with plant age and was much greater on flowering plants than on non-flowering ones. [For previous related work see HcA 41, 4400.]. Siberian hamsters ( Phodopus sungorus) rely on photoperiod to coordinate seasonally appropriate changes in physiology, including immune function. Immunity is regulated, in part, by the sympathetic nervous system (SNS), although the precise role of the SNS in regulating photoperiodic changes in immunity remains unspecified. The goal of the present study was to examine the contributions of norepinephrine (NE), the predominant neurotransmitter of the SNS, to photoperiodic changes in lymphocyte proliferation. In experiment 1, animals were maintained in long [16:8-h light-dark cycle (16:8 LD)] or short days (8:16 LD) for 10 wk, and splenic NE content was determined. In experiment 2, in vitro splenocyte proliferation in response to mitogenic stimulation (concanavalin A) was assessed in spleen cell suspensions taken from long- or short-day hamsters in which varying concentrations of NE were added to the cultures. In experiment 3, splenocyte proliferation was examined in the presence of NE and selective α- and β-noradrenergic receptor antagonists (phenoxybenzamine and propranolol, respectively) in vitro. Short-day animals had increased splenic NE content compared with long-day animals. Long-day animals had higher proliferation compared with short-day animals independent of NE. NE (1 μM) further suppressed splenocyte proliferation in short but not long days. Last, NE-induced suppression of proliferation in short-day hamsters was blocked by propranolol but not phenoxybenzamine. The present results suggest that NE plays a role in photoperiodic changes in lymphocyte proliferation. Additionally, the data suggest that the effects of NE on proliferation are specific to activation of β-adrenergic receptors located on splenic tissue. Collectively, these results provide further support that photoperiodic changes in immunity are influenced by changes in SNS activity.

Abstract This study combined in vitro production of bovine blastocysts, multiple embryo transfer techniques, and a conceptus-endometrial explant co-culture system to test the hypothesis that bovine endometrium exposed to long vs. short day 15 conceptuses would exhibit a different transcriptome profile reflective of potential for successful pregnancy establishment. Bovine endometrial explants collected at the late luteal stage of the estrous cycle were cultured in RPMI medium for 6 h with nothing (control), 100 ng/mL recombinant ovine interferon tau (IFNT), a long day 15 conceptus, or a short day 15 conceptus. Transcriptional profiling of the endometrial explants found that exposure of endometrium to IFNT, long conceptuses, or short conceptuses altered (P < 0.05) expression of 491, 498, and 230 transcripts, respectively, compared to the control. Further analysis revealed three categories of differentially expressed genes (DEG): (i) commonly responsive to exposure to IFNT and conceptuses, irrespective of size (n = 223); (ii) commonly responsive to IFNT and long conceptuses only (n = 168); and genes induced by the presence of a conceptus but independent of IFNT (n = 108). Of those 108 genes, 101 were exclusively induced by long conceptuses and functional analysis revealed that regulation of molecular function, magnesium-ion transmembrane transport, and clathrin coat assembly were the principal gene ontologies associated with these DEG. In conclusion, bovine endometrium responds differently to age-matched conceptuses of varying size in both an IFNT-dependent and -independent manner, which may be reflective of the likelihood of successful pregnancy establishment.
