Among the features of cisplatin chemotherapy-induced peripheral neuropathy are chronic pain and innocuous mechanical hypersensitivity. The complete etiology of the latter remains unknown. Here, we show that cisplatin targets a heterogeneous population of tyrosine hydroxylase-positive (TH+) primary afferent dorsal root ganglion neurons (DRGNs) within the primary afferent dorsal root ganglia in mice, determined using single-cell transcriptome and electrophysiological analyses. TH+ DRGNs regulate innocuous mechanical sensation through C-low threshold mechanoreceptors. A differential assessment of wild-type and vitamin E deficient TH+ DRGNs revealed heterogeneity and specific functional phenotypes. The TH+ DRGNs comprise; fast-adapting eliciting one action potential (AP; 1-AP), moderately-adapting (>=2-APs), in responses to square-pulse current injection, and spontaneously firing (SF). Cisplatin increased the input resistance and AP frequency but reduced the temporal coding feature of 1-AP and >= 2-APs neurons. By contrast, cisplatin has no measurable effect on the SF neurons. Vitamin E reduced the cisplatin-mediated increased excitability, but did not improve the TH+ neuron temporal coding properties. Cisplatin mediates its effect by targeting outward K+ current, likely carried by through K2P18.1 (Kcnk18), discovered through the differential transcriptome studies and heterologous expression. Studies show a potential new cellular target for chemotherapy-induced peripheral neuropathy and implicate the possible neuroprotective effects of vitamin E in cisplatin chemotherapy.

Abstract Purpose: Oxidative stress is known to be a decisive factor in the wide etiopathogenesis of optic neuropathy. This study aimed to comprehensively evaluate the interaction of optic neuropathy’s clinical course with systemic oxidative damage and antioxidant response dynamics in a large series.Methods: This case-controlled clinical study included 33 non-arteritic anterior ischemic optic neuropathy patients and 32 healthy individuals. Extensive systemic oxidation profiles were statistically compared between the two groups, and correlations between the clinical and biochemical data in the patient group were analyzed.Results: Vitamin E and MDA levels were significantly higher in the patient group. Significant correlations were observed in the analyses between clinical findings and oxidative stress parameters. Correlations between vitamin E and intraocular pressure, between B12 and cup-to-disc ratio, between antioxidant glutathione and SOD enzyme systems, and between uric acid and age were found to be very significant. As significant correlations were found in either clinical and biochemical data or in oxidative stress parameters, correlations between vitamin E and cholesterol, MDA was found to be very significant. Conclusions: This study not only supplies significant information regarding oxidative damage and antioxidant response in NAION, but also points out the specific interactions of neuromodulators, like vitamin E, in intracellular signaling pathways and regulation mechanisms. A better reading of these connections may help improve diagnosis, follow-ups and treatment criteria and strategies.

This experiment aimed to investigate effects of the different dietary levels of vitamin E (VE) on the growth performance, immunity and antioxidant capacity of goslings. A total of 240 1-day-old, male Jiangnan white goslings were selected and randomly divided into 6 groups. Each treatment included five replicates. The basal diet was supplemented with 6 concentrations of VE (0, 12, 24, 36, 48 and 60 mg DL-α-tocopherol acetate/kg). The results were as follows: (1) The α-tocopherol content in the serum and liver of goslings increased linearly as supplemental VE increased in diet (p < 0.05). (2) The body weight (BW) and average daily gain (ADG) increased quadratically with increasing dietary VE supplementation (p < 0.05). Dietary VE supplementation could significantly reduce the feed/gain ratio (F/G) (p < 0.05). (3) Dietary supplementation with VE could significantly improve the contents of immunoglobulin A (IgA) and immunoglobulin G (IgG) in the serum of the goslings (p < 0.05). The content of interferon-γ (IFN-γ) in the serum was significantly reduced with VE supplementation (p < 0.05). (4) Dietary supplementation with VE could significantly improve serum and liver catalase (CAT), superoxide dismutase (SOD) activities and liver total antioxidant capacity (T-AOC) (p < 0.05); Serum and liver MDA contents were significantly reduced with VE supplementation (p < 0.05). In summary, dietary supplementation with VE could improve growth performance, immunity and antioxidant capacity. Based on broken-line regression analysis, the dietary VE supplementation level for ADG was 12.51 mg/kg, but higher supplementation level should be considered to improve immunity and antioxidant capacity.

Objective: The study aimed to investigate the structural and conformational changes induced by short-term administration of the amiodarone in the optic nerve besides validating whether vitamin E coadministration with amiodarone will improve these changes. Methods: Thirty New Zealand white rabbits from both sexes were haphazardly categorized into three groups, whereas each group contains ten rabbits (20 eyes). One of these groups served as a control that received an intraperitoneal injection of normal saline. Rabbits in the second group intraperitoneally (ip) injected daily with 160 mg/kg body weight (bw) of amiodarone for two weeks. The last group orally administration 100 mg/kg bw of vitamin E with the 160 mg/kg bw of amiodarone ip daily for two weeks until the time of sacrifice. Fourier transform infrared spectroscopy (FTIR) analysis was conducted on the optic nerve of the all groups. Results: The results obtained from the FTIR spectrum revealed that the short-term administration of amiodarone caused a significant alteration in the stretching NH-OH region. A newly detected component centered at 3739±1 cm-1 was assigned as strO-H. There was a significant decrease (p˂0.05) in the bandwidth and band position of one component of strO-H that centered at 3598±1 cm-1. Moreover, remaining vibrational bands (O-Hasym and O-Hsym) were shifted to higher frequencies. Coadministration of vitamin E with amiodarone reduced the contour to four components as a control with significant increase in the band position of O-Hasym and the bandwidth of one component of str O-H. Amiodarone administrations lead to reducing the area ratio of asymCH2 to symCH2 and elevation of the area ratio of asymCH2 to asymCH3 while the coadministration of vitamin E returned it as the control ratio. The percentage of the β-turn was significantly increased while the α-helix content was decreased due to amiodarone. The contents of both components were considered mimicking the control values when Vitamin E was co-administered with amiodarone. Conclusion: The study stated that amiodarone could change the solubility and folding of the optic nerve proteins. Finally, vitamin E intake with amiodarone turns many of these changes induced by amiodarone to normal levels, which make it a good supplement for amiodarone users.

Encapsulation of vitamin E is the preservation of the biological activities of vitamin E for various applications. In the first part of this research, factors affecting the batch encapsulation of vitamin E, including PCL concentration, the concentration of Tween 20, and the volumetric ratio of aqueous phase to organic phase were experimentally investigated. The Box-Behnken experimental design and response surface methodology were implemented to determine the optimal operating conditions of the batch encapsulation. At the optimal conditions, the percentage of vitamin E encapsulation (%EC) was 98.69%, using the PCL concentration, the Tween 20 concentration, and the volumetric ratio of aqueous phase to organic phase of 3.6 g/L, 0.6 g/L, and 0.9 mL: 1 mL, respectively. The second part is to enhance the productivity by applying the optimized formulation of vitamin E encapsulation in a continuous process using a micro-channel encapsulator. The effect of residence time was investigated. At the residence time of 1 s, the percentage of vitamin E encapsulation of 97.28% and the productivity of 153.61 mg/(mL∙min) were achieved.

Candelilla wax (CW) and 12-hydroxystearic acid (12HSA) are classic solid-fiber-matrix organogelators. Despite the high number of studies using those ingredients in oily systems, there is scarce literature using a mixture of oil and antioxidants. Vitamin E (VE) is an important candidate for its lipophilicity and several applications on pharmaceutical, cosmetics, and food industries. In this work, we investigated the influences of mixtures between vegetable oil (VO) and VE on the microstructures and rheological properties of CW and 12HSA organogels. A weak gel (G′′/G′ > 0.1) with a shear-thinning behavior was observed for all samples. The presence of VE impacted the gel strength and the phase transition temperatures in a dose-dependent pattern. Larger and denser packed crystals were seen for 12HSA samples, while smaller and more dispersed structures were obtained for CW organogels. The results obtained in this work allowed the correlation of the structural and mechanical properties of the organogels, which plays an important role in the physical-chemical characteristics of these materials.
