The Hirtodrosophila melanderi species group (Diptera: Drosophilidae) from the Huanglong National Nature Reserve, Sichuan, China

The Hirtodrosophila melanderi species group is currently known for thirteen described species, most of which were thought to be fungivorous. More than half known species of this species group were recorded exclusively from high altitude zone to the southeast of the Qinghai-Tibet Plateau in China. In our recent field survey in the Huanglong National Nature Reserve (located to the east of the Qinghai-Tibet Plateau) in Sichuan Province, China, we collected dozens of specimens of the H. melanderi group there. In the present study, these specimens are subjected to species delimitation based on data of not only morphology, but also DNA barcodes (nucleotide sequences of a 658-bp fragment of the mitochondrial cytochrome c oxidase subunit I gene). The five new species thus recognized are described: Hirtodrosophila minshanensis sp. nov., H. lambda sp. nov., H. zhangae sp. nov., H. zouae sp. nov., and H. nigrispina sp. nov. In addition, an updated key to all species of the H. melanderi species group is provided.

A new species of the genus Panolis Hübner, [1821], P. xundian sp. nov., is described and illustrated from southwestern China. It is well-defined morphologically by the male genitalia, with a well-developed pollex at the distal terminal of the sacculus and a broad, ventrally concave cucullus, the female corpus bursae with four long signum-stripes. Based on a 658 bp segment of the mitochondrial cytochrome c oxidase subunit I gene, we report the pairwise genetic distance of 2.5% from its allied species P. exquisita Draudt, 1950. Molecular phylogenetic analyses using three genes (2189 bp in total length) indicate that the new species belongs to the P. exquisita species group.

Woodroaches from the genus Cryptocercus Scudder, 1862 are known to display low levels of morphological divergence, yet significant genetic divergence and variability in chromosome number. Compared with Cryptocercus taxa from North America, the diversity of the genus in Asia has received relatively little attention. We performed morphological and karyotypic examinations of multiple taxa from several previously unsampled mountainous areas of central and south-western China, and identified nine candidate species primarily on the basis of chromosome number. We then investigated diversity across all Asian Cryptocercus, through phylogenetic analyses of 135 COI sequences and 74 28S rRNA sequences from individuals of 28 localities, including species delimitation analysis in General Mixed Yule Coalescent (GMYC) and Automatic Barcode Gap Discovery (ABGD). Phylogenetic results indicated that individuals from the same locality constituted well supported clades. The congruence of GMYC and ABGD results were in almost perfect accord, with 28 candidate species described on the basis of karyotypes (including the nine identified in this study). We provide evidence that each valley population in the Hengduan Mountains contains a separate evolving lineage. We conclude that the principal cause of the rich Cryptocercus diversity in China has been the uplift of the Qinghai-Tibet Plateau.

Neighbor-joining and maximum likelihood analyses of the barcode region of the mitochondrial cytochrome c oxidase subunit I gene (COI DNA barcode) were conducted to aid in species delimitation.

A key to all Asian species (except for the eparmata species complex) of this subgenus was provided. All currently available DNA barcode (partial mitochondrial cytochrome c oxidase subunit I (COI) gene) sequences of this subgenus (217 sequences of 54 species) are employed in a molecular analysis using different species delimitation methods. The results indicate that approximately 68.5% (37 of 54 spp.) of Phortica (s. str.) species could be clearly distinguished from closely related morphospecies or cryptic species.

Five previously known species were attributed to the Lycocerus fainanus species group, including L. inopaciceps (Pic 1926), L. oberthueri (Gorham 1889), L. oudai (Švihla 2004), L. metallipennis (Fairmaire 1887), and L. nigripes (Wittmer, 1995). Four new species of this group were discovered from China and Vietnam, L. binotatus sp. nov., L. testacicollis sp. nov., L. daliensis sp. nov., and L. vietnamensis sp. nov. An updated key to all species was provided. A geographical distribution map is presented, which shows that all the members were located between 18.69041–33.93441° N, and between 98.61413–121.77102° E. The ancestral geographical range was reconstructed based on a phylogeny of morphological data by the Bayesian Binary MCMC method. The result showed that the spatial origin of L. fainanus species group was probably located in northern Vietnam and southwest China. The divergence of the species in southwest China and Taiwan was caused by vicariance about 24 Ma ago, when the latter was separated in the Qinghai-Tibet Plateau, and the remaining species of mainland China all originated from Taiwan after traveling around Southeast Asia and back to China. Nevertheless, this conclusion should be verified when fossil evidence and molecular data are available.

Lithobius (Ezembius) longibasitarsussp. n. and Lithobius (Ezembius) datongensissp. n. (Lithobiomorpha: Lithobiidae), recently discovered from Qinghai-Tibet Plateau, China, are described. A key to the species of the subgenus Ezembius in China is presented. The partial mitochondrial cytochrome c oxidase subunit I barcoding gene was amplified and sequenced for eight individuals of the two new species and the dataset was used for molecular phylogenetic analysis and genetic distance determination. Both morphology and molecular data show that the specimens examined should be referred to Lithobius (Ezembius).
