Lily (Lilium longiflorum) is a perennial herbaceous plant with white trumpet-shaped flowers, fragrant and bulbous. In vitro culture through bulbs is one of way propagation of lily plants, but it requires a long time and only produces limited plants. In vitro propagation is a very promising technique for plant propagation because it can produce a lot of plant seeds in a short time. Bulbs are one of the fastest explants for growing shoots in lilies, but it is not known for certain which cuts of explants from bulb scales are best for multiplying in vitro. This study aims to determine the effect of lily bulb explants and the concentration of NAA and BAP growth regulators on the growth of lily bulb explants. The best results were obtained on the base and middle cuts explant of bulb scales compared to the tip cuts explant ones. The best results of the growing percentage, the number of shoots and the best growing time are shown in the combination treatment of growth regulator 1 mg L−1 NAA and 1 mg L−1 BAP. The optimum results on the number of micro bulbs were found in the treatment of growth regulators 0.5 mg L−1 NAA and 1 mg L−1 BAP. The best results of the average time formed micro bulb was in the treatment of 1 mg L−1 NAA and BAP with middle explant cuts, and treatment concentrations of 0.5 mg L−1 NAA and BAP in the base explant section. The base and middle bulb explants are able to regenerate or grow higher shoots. This is caused by the presence of endogenous natural auxin and the spread of auxin in plant parts not in the same amount. Therefore when added to the exogenous growth regulator such as auxin or cytokines to culture media will further trigger the formation of micro tubers more quickly,. It can increase the concentration of endogenous growth regulators in cells, help growing process and developing tissue.   Keywords: Bulb, lily, micro bulbs, in vitro, shoots

Marumi kumquat (Fortunella Japonica) is culture for its valuable nutritional value and medicinal importance in many regions of the world. The current study aimed to evaluate the effect of two types of media enriched with different concentrations of fructose and different plant growth regulators and different fructose concentration on in vitro propagation of Fortunella Japonica. The findings showed that the most effective treatment for explant surface sterilization was by using 0.1% HgCl2 for ten minutes which give best results for production contamination-free explants at the initiation cultures. At multiplication stage, WPM medium gave better results at all tested BA levels as compared with MS medium. No significant differences were showed by using BA alone or in combination with GA3 in the measured parameters. It has been observed that WPM medium supplemented with 0.5mgl-1 BA with the presence of 30mgl-1 fructose was able to give the highest shoot length (1.56cm) with maximum shoots number/explant 9.0 and highest leaves number/explant (21.0). The proliferated shoots were exposed to full strength MS medium salts supplemented with 2mgl-1 NAA which showed the highest ratio of rooting. In vitro rooted plantlets were gradually acclimatized and transferred to open air conditions, which recorded a high survive rate reached to 92%

Abstract The aim of this study is to develop and improve methods of in vitro propagation of representatives of Dactylorhiza: D.baltica , D. fuchsii. For the study, we used protocorms obtained by the asymbiotic germination of seed during 90 days. It has been established that half-strength of Murashige and Skoog (1962) medium (½ MS) supplemented with 1-2 mg/l 6-Benzylaminopurine(6-BAP), potato puree (20g/l), and charcoal (1g/l) effectively influenced the development of protocorms, and seedlings formation in the studied species. The result of the study showed that the survival rate of protocorms was high in all experimental culture media, but in D. fuchsii it was better at a concentration 2mg/l of 6-BAP (95.4%), while in D. baltica it was high at 1mg/l (87.0%). The highest percentage of multiple protocorms (68%) and the formation of new secondary protocorms in D. fuchsii (5,5±0,3 units) were observed on a culture medium containing 2 mg/l 6-BAP. The highest percent of rooting of D. fuchsii protosoms (78%) and length of roots (0.9cm) observed in ½ MS medium without growth regulators. During the development of D. baltica protosoms, the culture medium of ½ MS containing 1 mg/l 6-BAP had the best effect on the number of roots (1.8±0.1root/protosom), while the medium supplemented with 2mg/l of 6-BAP contributed to the formation of a larger number of new secondary protocorms (3,2±0,1protocorm/unit). During the subsequent cultivation of protosoms of D. baltica on a culture medium containing 1 mg/l it was observed an increase in the height of shoots (4,8±0,3 см), and the length of roots (2,2±0,1 см), wherein the number of newly formed protocorms was higher by 30% on the medium supplemented with 2 mg/l 6-BAP. Tanaman lili (Lilium longiflorum Thunb.) merupakan tanaman hias dan bunga potong. Permintaan bunga potong lili sangat menjanjikan tetapi ketersediaan bibit masih terbatas. Penelitian ini terdiri dari perkecambahan biji lili secara in vitro pada media ½ Murashige & Skoog (MS), multiplikasi bagian eksplan pangkal dan ujung tunas pada berbagai perlakuan media MS kombinasi ZPT BAP dan NAA.  Tujuannya untuk mendapatkan kombinasi media yang tepat dan mengetahui persentase tertinggi bagian eksplan yang mampu tumbuh tunas, akar dan kalus. Kombinasi media MS dengan BAP dan NAA yaitu (a) kontrol 0:0 mg L-1, (b) 0:0.1 mg L-1, (c) 0:1 mg L-1, (d) 0.1:0 mg L-1, (e) 0.1:0.1 mg L-1, (f) 0.1:1 mg L-1, (g) 1:0 mg L-1, (h) 0.1:1 mg L-1, (i) 1;1 mg L-1. Penelitian ini disusun dalam RAL faktorial dua faktor yaitu kombinasi pemberian BAP dan NAA, multiplikasi eksplan bagian pangkal dan ujung tunas. Biji lili mampu berkecambah sebanyak 58%. Media c dan i yang mampu menginduksi eksplan untuk tumbuh tunas, akar dan kalus. Media c mampu menginduksi tunas, akar dan kalus, media i mampu menginduksi akar dan kalus. Persentase respon pertumbuhan eksplan bagian pangkal tunas yaitu pertumbuhan tunas 11.1% dan akar 13.3%, sedangkan eksplan bagian ujung tunas menunjukkan pertumbuhan akar 6.7% tanpa pertumbuhan tunas. Pertumbuhan kalus menunjukkan hasil yang sama pada eksplan pangkal dan ujung tunas yaitu 4.4%.

Nodal explants of 12 accessions from four species of yam (Dioscorea spp.) were cultured for six weeks on MS to evaluate the influence of IAA, Kn, NAA and BAP on the production of leaves and microtubers. Four Dioscorea polystachya Turcz., three each of D. bulbifera L. and D. sansibarensis Pax. and two D. japonica Thunb. accessions were used. Five and 10 mg/l of Kn along with IAA and sucrose, and 0.2 and 0.5 mg/l of NAA, sucrose and with or without BAP were used in four treatments. The accessions Yam 23 and Yam 25 of D. sansibarensis failed to initiate any leaf under four treatments. The remaining accessions produced 0.11 to 1.76 leaves per explant. The medium containing IAA with higher concentration of Kn (10 mg/l) and 3% sucrose was found to be best for in vitro production of leaf (0.71/explant) and the most productive species was D. japonica (1.36), followed by D. polystachya (1.19/explant). At the same culture period, Yam 16 of D. bulbifera failed to initiate any microtuber at IAA with Kn, and NAA with or without BAP. The remaining accessions produced 0.09 to 1.15 microtubers per explant. Lower concentration of Kn (5 mg/l) with IAA and sucrose was favourable for producing microtubers (0.61/explant on an average), the best species being D. sansibarensis (1.27) followed by D. japonica (0.59/ explant).
