Natural pigments improve aesthetic value as well as antioxidant potential of a food product. This study was designed to determine the effects of green extraction techniques on carotenoids, polyphenols and antioxidant activities of pulp and peel of two varieties of pumpkin (Cucurbita maxima). Innovative green extractions (IGE; Ultrasound and Microwave-Assisted Extractions) synergised with corn oil (used as green solvent) were compared with conventional extraction (CE; hexane/isopropyl alcohol; 60:40, v/v). Results showed total carotenoids to be almost double on employing IGE (PM2-UAE-peel = 38.03 ± 4.21; PM4-UAE-peel = 33.78 ± 1.76 µg/g) when compared to conventional extraction (PM2-CE-peel = 19.21 ± 4.39; PM4-CE-peel = 16.21 ± 2.52 µg/g). Polyphenolic contents ranged between 510.69 ± 5.50 and 588.68 ± 7.26 mg GAE/100 g of extract in IGE, compared with conventional extracts (269.50 ± 2.17 to 318.46 ± 6.60 mg GAE/100 g) and percent inhibition of 2,2-Diphenyl-1-picrylhydrazyl (DPPH) ranging between 88.32 ± 1.51 and 93.53 ± 0.30% in IGE when compared with conventional extraction (50.61 ± 1.44 to 57.79 ± 2.09%). Further, oxidative stability of carotenoids extracts from IGE (protection factor = 1.59 ± 0.01 to 1.81 ± 0.05) were found to be significantly higher (p < 0.05) than conventional extracts. Based on results, this study supports the use of innovative green extraction techniques to obtain bioactive pigments like carotenoids. It is anticipated that results generated will find potential applications in food, pharmaceutical and cosmetic industries.

Introduction. Dust is one of the main harmful factors in underground coal mining. The dust concentration in the coal mines remains high due to the imperfection of the used technologies, increasing the pneumoconiosis risk in miners. Respirators are the last means of protection, but information about their effectiveness is contradictory. Aim. Assess the effectiveness of individual respiratory protective devices in the coal mines. Material and methods. The protection factor (PF), the ratio of the dust concentration outside the mask to the concentration under the mask) is used to assess respirators’ effectiveness. We measured the PFs in the laboratory and the workplaces. We also measured the time the miners did not use respirators. The measurements’ results were used as input data for mathematical modeling to reduce dust load reduction. Results. The concentration of dust under the mask exceeded the Occupational Exposure Limit (OEL) in most cases. Information about the high efficiency of the negative pressure half-mask respirators (in case of continuous use) was not confirmed. The PFs measured under laboratory conditions reached 6.2, and in the workplace were from 5 to 31. The miners did not use respirators from 10 to 45% of the shift duration (15 measurements). The measurements and information on the concentration of dust in 5 mines were used to simulate the impact of the respirators’ usage on the dust load. Calculations performed by different methods, and using various programs, gave the same result. The negative pressure half-mask respirators cannot reduce the dust’s concentration in the inhaled air to the OEL even with continuous use. Conclusion. The results showed that the respirators’ usage could not prevent developing incurable pneumoconiosis in miners.

KeponeR (decachlorooctahydro-1,3,4-metheno-2H-cyclobuta[cd]pentalen-2-one) is an insecticide effective against ants and roaches. It can cause severe toxicity in fishes, birds, rodents and man. Prominent effects include hepatic lipid deposition and hypertrophy, impairment of reproductive capacity and neurological disorders. Mitochondrial oligomycin-sensitive Mg2+-ATPase is also inhibited. The present study is a preliminary investigation of tissue ultrastructural changes accompanying physiological signs of acute toxicity, which after two days treatment include: pronounced hypersensitivity and tremor, various degrees of anorexia and adipsia, and decreased weight gain.Three different series of adult male Sprague-Dawley rats (Charles River or CD-I) were treated by intubation with Kepone in corn oil at a dose of 50 mg per kg for 3 successive days or at 200 ppm in food for 8 days. After ether anesthesia, rats were immediately perfused via a cannula in the left ventricle with 4% p-formaldehyde and 0.5% glutaraldehyde in Millonig's phosphate buffer at pH 7.2 for 20-30 min at 22°C.
