The experiment was conducted to optimize a suitable protocol for in vitro regeneration in cole crops. Callus initiation was excellent in the variety Early Tropical. Highest percentage of callus proliferation was observed in Early Tropical (75.0%) followed by Tangail Special Pauslali (55.0%) and the lowest in Tara (40.0% ). Maximum callus proliferation (68.5%) was observed in MS + 3.0 mgL-1 BAP + 0.1 mgL-1 2,4-D + 2.0 mgL-1 AgNO3. Callus proliferation was lowest (40.0%) in MS + 2.5 mgL-1 BAP + 0.1 mgL-1 2,4-D + 2.0 mgL-1 AgNO3. MS medium supplemented with 3.0 mgL-1 BAP + 1.0 mgL-1 2,4-D + 2.0 mgL-1 AgNO3 was the best for shoot initiation & plantlet regeneration. The highest number of shoots per vial was 7.20 and the lowest number of shoots per vial was 4.40. Among the concentration MS + 3.0 mgL-1 BAP + 0.1 mgL-1 2,4-D + 2.0 mgL-1 AgNO3 showed the highest performance of shoots per vial. The experiment was conducted to develop an efficient protocol for in vitro regeneration of mungbean (Vignaradiata) on the aspect of regeneration potentiality of two mungbean varieties (BINA mung 5 and BINA mung 7) as influenced by different combinations of growth regulators supplemented with MS medium. Cotyledon explant of both varieties was used for the present study. Data were collected for various characters of callus initiation, shoot regeneration and root proliferation. Initiation of callus (%) and required days for its initiation and weight of callus were influenced significantly due to the effect of varieties where BINA mung 5 produced more callus induction (40.36%) at minimum requiring time (18.27 days) including heavier sizes of callus (1.54 g) than BINA mung 7 when BINA mung 5 further recorded the longest root (2.92 cm) compare to BINA mung 7. Effect of treatments of the present study were significantly influenced the whole characters regarding callus culture, shoot regeneration and root proliferation. The highest percentage of callus (88.44%) within minimum time (12.53 days) including larger sizes callus (3.521 g) were produced in 1.0 mg L–1 BAP + 2.5 mg L–1 NAA among the treatments while the highest percentage of regenerated shoot (83.44%) at minimum requiring time (17.59 days) and more shoots (7.69 callus–1) were obtained in 1.0 mg L–1 BAP + 2.0 mg L– 1 NAA. Root induction (82.50%), number of roots plantlet–1 (8.469) with minimum requiring time for initiation (14.13 days) and root length (5.250 cm) were the highest in 0.2 mg L–1 IAA + 1.0 mg L–1 kinetin + 0.2 mg L–1 BAP. Incase of interaction, percentage of callus initiation (89.38 %) was the highest in BINA mung 5 treated by 1.0 mg L–1 BAP + 2.5 mg L–1 NAA at requiring minimum time (12.38 days) while same treatment produced the larger callus (3.581 g) among the interactions. The highest percentage (84.38%) and number (7.813 callus–1) of shoot with minimum requiring time (17.50 days) were found from BINA mung 5 treated by 1.0 mg L–1 BAP + 2.0 mg L–1 NAA. Similarly, the longest shoot (5.58 cm) was produced from the BINA mung 5 treated by 1.0 mg L–1 BAP + 2.0 mg L–1 NAA. However, root induction (%), roots plantlet–1, days required for root initiation and root length were statistically similar among the whole interaction treatments due to non significant variation. This result mentioned that the variety BINA mung 5 was better than BINA mung 7 for callus induction, shoot regeneration and root initiation while 1.0 mg L–1 BAP + 2.5 mg L–1 NAA, 1.0 mg L–1 BAP + 2.0 mg L–1 NAA and 0.2 mg L–1 IAA + 1.0 mg L–1 kinetin + 0.2 mg L–1 BAP supplemented with MS medium were the best combinations for better callusing, higher ability of shoot regeneration and root proliferation.DOI: http://dx.doi.org/10.3329/jesnr.v7i2.22203 J. The experiment was carried out during the period from June 2016 to January, 2017 at the Plant Biotechnology Laboratory, Department of Horticulture, Patuakhali Science and Technology University (PSTU) for in vitro regeneration of sweet potato. In this study, the nodal segment explants and different combinations of growth regulators were used for in vitro regeneration of sweet potato. BAP (6-benzylamino purine) and KIN (Kinetin) were used for in vitro shoot regeneration and IBA (Indole-6- Butyric Acid) with NAA (Naphthalene Acetic Acid) were used for in vitro root regeneration. The highest percentage of shoot initiation (91.30), the minimum number of days (9.00) for shoot initiation, the highest number of shoot/plantlet (11.00) and the highest shoot length (4.38 cm) was observed when nodal segment explants cultured on MS medium supplemented with BAP 1.5 mg/L + KIN 0.1 mg/L. On the other hand, the highest root initiation percentage (94.12), the minimum number of days (6.00) for root initiation, the highest number of root/plantlet (9.33) and the highest root length (11.13 cm) was observed in the MS medium supplemented with IBA 0.5 mg/L + NAA 0.1 mg/L. An experiment was conducted on in vitro regeneration and multiplication of Dendrobium bensoniae. Different concentrations of BA and IBA alone or combination of both hormones were used as treatment for regeneration.  It was revealed that shoot regeneration from node was the best at 2.0 mg/l BA supplemented to MS medium. It gave better responses than all other concentrations and combinations of BA and BA+IBA, used in the present study. The highest number of shoots and leaves were found when 1.0 mg/l BA with 1.5 mg/l IBA was supplemented into MS medium.  For rooting, 0.5 mg/l BA with 1.0 mg/l IBA was found to be the most effective. The well-rooted plantlets were successfully acclimatized under 70-80% humidity and planted in pots and transferred to the shade house for establishment. Around 85% of plantlets survived in the field. From the present result, it may be recommended that MS medium supplemented with 2.0 mg/l BA may be used for rapid shoot induction and regeneration of D. bensoniae.The Agriculturists 2016; 14(2) 24-31

An efficient in vitro regeneration system was developed for Rauvolfia serpentina L. through direct and indirect organogenesis from nodal and leaf explants. Among the different growth regulators, MS medium supplemented with 2.0 mg/l BAP, 0.5mg/l IAA and 0.02mg/l NAA found best for the multiple shoot formation from nodal segments. In this combination 98% explants produced multiple shoots and the average number of shoots per explants is 13∙4. The frequency of callus induction and multiple shoot induction from leaves was highest 88% in MS medium supplemented with 2.0 mg/l BAP, where mean number of shoots/explants was 12.5. The highest frequency of root induction (80%) and mean number of roots/plantlets (10) were obtained on half strength of MS medium containing 0.2 mg/l IBA. The rooted plantlets were transferred for hardening following acclimatization and finally were successfully established in the field.Bangladesh J. Sci. Ind. Res.53(2), 133-138, 2018

The effect of cytokinins on callus proliferation from cotyledons and plantlet development was studied in cotton. The frequency of callus induction was observed on MS medium enriched with a variety of cytokinins in different concentrations. With the increase of cytokinin concentration, the percentage of callus formation, percentage of shoot developing calli and number of shoots/calli were increased. Among the three different cytokinins studied, BA showed the highest performance. The highest percentage of callus (6.55%) and shoot developing calli (5.87%) was obtained on MS with 1.0 mg/l BA. Highest number of shoots (3.02) per calli was observed on MS media supplemented with 1.0 mg/l Kn. The rooting media composed of MS medium, 0.6% agar, sucrose and fortified with 2.0 mg/l NAA induced root development at the highest percentage (41.23%) with maximum number of roots (3.61) per cutting and length of root (3.62 cm) per culture. The plantlets were acclimatized in natural conditions. Chinese fever vine (Paederia foetida L.), a valuable medicinal plant has been greatly utilized in therapeutic purposes throughout the world. Since conventional propagation techniques of P. foetida are very slow, inefficient and cannot cope with the increasing demand, in-vitro regeneration through tissue culture could be an alternative means of rapid propagation. Therefore, the efforts were made to develop a suitable protocol through direct organogenesis of P. foetida. After surface sterilization, the nodal explants were cultured in Murashigue and Skoog (MS) medium and MS medium supplemented with different concentrations and combinations of plant growth regulators. MS medium supplemented with 6-benzylaminopurine; BAP (2.0 mg L-1) produced the maximum number of shoots; 4.40 ± 0.98 and 5.40±1.12 after 15 and 30 days of culture respectively. The number of shoots gained by 15 days was found to be the highest; 1.20±0.80 at BAP (4.0 mg L-1) followed by 1.00±0.55 at BAP (2.0 mg L-1). Although the combination of BAP + Kinetin (2 mg L-1 +2 mg L-1) showed the highest shoot growth (3.40 ± 1.08 cm) by 15 days, sole application of BAP (2.0 mg L-1) or Kn (0.5, 1.0, 2.0 and 3.0 mg L-1) showed similar responses. BAP (2.0 mg L-1) showed the best responses for developing the highest number of leaves; 18.60 ± 2.42 and 29.20 ± 2.73 respectively after 15 and 30 days of culture. Similarly, development of the maximum number of leaves (10.60 ± 0.68) was reported by 15 days at BAP (2.0 mg L-1). Rooting was significantly induced in indole-3-acetic acid (IAA) supplemented to 1/2 strength MS medium as compared to control (only ½ strength MS medium). The best performance of rooting was observed by 0.5 mg L-1 IAA which produced average 4.33 roots per shoot after 21 days of culture. The regenerated plants showed similar morphology to the mother plants. Thus, a suitable protocol for successful multiplication of P. foetida in vitro was established using nodal explants. Ann. In vitro regeneration of memory enhancing plant Bacopa monneiri was observed by using nodal explant. In present study, shoot initiation was best seen in MS media in 3mg/l BAP concentration. Combination of BAP and IAA (1mg/l+3mg/l concentration) was best suited for root initiation. The rooted plantlets were hardened and successfully established in soil. The experiment was conducted on in vitro regeneration of tomato at the Plant Biotechnology Laboratory, Department of Horticulture, Patuakhali Science and Technology University, Patuakhali. The objective was to develop an efficient regeneration protocol in tomato through callus induction for subsequent plantlet regeneration. Seeds were inoculated on MS medium where germination rate was 78.4%. The stems of in vitrocultured seedlings were used as explants. Different concentrations and combinations of growth regulators were added to MS medium to observe their efficacy on callus induction, shoot initiation and root formation. Stem explants cultured on MS medium fortified with 2 mg/L BAPgave the highest number of shoots (3.0) at 45 DAC. Among the concentrations of PGRs, 0.25 mg/L IAA produced the highest length (4.064 cm) of plantlets, number (5.0) of leaves and fresh weight (0.663 g) of plantlets with the stem explants at 45 DAC. The concentration of 0.5 mg/L IAA produced the highest number (21.00) of roots/plantlet, length (7.676 cm) of roots at 45 DAC, from the same explants. The highest survival rate of in vitro regenerated plantlets in the pot was 70.00 % with the stem explants. The results of the current study showed significant increase in the growth of callus of Solanumlycopersicon Mill. Indicating a good efficiency of the optimized media composition and the experimental model used in comparison to other studies of similar nature.Asian J. Med. Biol. Res. An efficient and reproducible in vitro regeneration protocol was established for two varieties of ginger (Z. officinale Roscoe) namely, BARI Ada-1 and Chinese ginger accession number SG876). In case of BARI Ada-1 best result was obtained on MS supplemented with 2.0 mg/l BAP, 0.5 mg/l Kn and 0.5 mg/l NAA. In this combination, 95% rhizome bud explants responded within 6 - 8 days and mean number of shoots per explant was 8.79 ± 0.42. On the other hand, Chinese ginger showed best (90%) shoot regeneration response from the same explants on the same medium and hormonal combinations but in exchange of 0.25 mg/l NAA. In this hormonal composition shoot initiation started within 7 - 8 days of culture and mean number of shoots/explant was 6.83 ± 0.71 after 24 - 27 days of culture. Maximum root induction (90 and 80%) was found on MS supplemented with 0.5 mg/l IBA and 0.5 mg/l NAA in case of BARI Ada-1 and Chinese ginger, respectively. The in vitro regenerated plantlets were successfully transplanted into the soil after acclimatization.

Full potential of grass pea has not been utilized because of the presence of the neurotoxin amino acid β-N-oxalyl-L-αβ -diaminopropionic acid (ODAP/BOAA). Conventional breeding and other approaches have not been successful in reducing the toxin. Integration of in vitro techniques can contribute significantly to meet the challenge. Therefore, this study was carried out to evaluate the in vitro regeneration capacity of grass pea genotypes. Shoot initiation, multiplication and rooting of IVAT-LS-690 were conducted using completely randomized design with five replications. Genotypes were treated with BAP and NAA for shoot initiation while BAP and Kn Combination were used for multiplication. Different concentrations of IBA and IAA were used for rooting. Shoot proliferation percentage was the highest (100%) for IVAT-LS-690,on Murashige and Skoog (MS) medium augmented with 2.0 mg/l BAP +0.1 mg/lNAA.For in vitro shoot multiplication, best results were obtained on concentrations of 3mg/l BAP+1mg/l Kn with maximum shoot number per explants (11.5). High number of roots per shoot (6) and percent of rooted shoot (86.66%) were obtained from ½ MS medium supplemented with 0.5 mg/l IBA. This study inferred that both genotype and BAP levels play a crucial role for shoot regeneration capacity and the optimum hormonal combination for grass pea is genotype specific.
