Hot extract of Aulosira fertilissima (cyanobacterium) added in different proportions to MS as a liquid culture media for the in vitro propagation of Bacopa monnieri (L.) Pennell. Maximum numbers of shoots were induced from axillary node in MS media (40 ml) + Aulosira extract (60 ml) and maximum shoot multiplication was observed when Kn (1.0 mg/l) was added in the shoot initiation media (mentioned above). Surprisingly rooting was also found to be best in the same combination of MS + cyanobacterial extract that was used for initiation and multiplication of shoots. An experiment was conducted at the Plant Biotechnology Laboratory, Department of Horticulture, Patuakhali Science and Technology University during the period from August 2016 to April 2017 to standardize the protocol for in vitro propagation of banana. The experiment was laid out in completely randomized design with four replications. Three to four months aged field grown rhizome attached shoots were used as explants and cultured on MS medium with different concentrations and combinations of BAP (6-benzylamino purine), BAP + KIN (Kinetin) + NAA (Naphthalene Acetic Acid) and IBA (Indole-3- Butyric Acid) + IAA (Indole-3- Acetic Acid) to observe their efficacy on single shoot initiation, shoot multiplication and root formation respectively. Minimum number of days required for shoot initiation (9.07) with highest shoot initiation percentage (91.14) and the longest shoot (2.23 cm) was found in MS medium supplemented with 5.0 mg/L BAP. On the other hand, highest shoot multiplication percentage (80.99) with maximum number of shoots per explant (4.47), the highest length of shoots (4.17 cm) and maximum number of leaves (4.04)was observed in MS medium supplemented with 4.0 mg/L BAP + 2.0 mg/L KIN + 2.0 mg/L NAA. In case of root regeneration, the best results on days required for root initiation (9.00), the highest root initiation percentage (85.05), maximum number of roots per plantlet (5.83) and the highest length of roots (4.17 cm) was obtained in MS medium supplemented with1.5 mg/L IBA + 0.5 mg/L IAA. In recent years, great strides have been made on the cultivation of spirochetes, and if relatively simple and completely reliable methods of growing them in vitro on both solid and liquid nutrient media have been developed for a number of pathogenic and non-pathogenic spirochetes, then on the issue of With the cultivation of the pale spirochete, we are moving forward very slowly, and the successes achieved recently in this area must be recognized as very modest.

Nodal explants of 12 accessions from four species of yam (Dioscorea spp.) were cultured for six weeks on MS to evaluate the influence of IAA, Kn, NAA and BAP on the production of leaves and microtubers. Four Dioscorea polystachya Turcz., three each of D. bulbifera L. and D. sansibarensis Pax. and two D. japonica Thunb. accessions were used. Five and 10 mg/l of Kn along with IAA and sucrose, and 0.2 and 0.5 mg/l of NAA, sucrose and with or without BAP were used in four treatments. The accessions Yam 23 and Yam 25 of D. sansibarensis failed to initiate any leaf under four treatments. The remaining accessions produced 0.11 to 1.76 leaves per explant. The medium containing IAA with higher concentration of Kn (10 mg/l) and 3% sucrose was found to be best for in vitro production of leaf (0.71/explant) and the most productive species was D. japonica (1.36), followed by D. polystachya (1.19/explant). At the same culture period, Yam 16 of D. bulbifera failed to initiate any microtuber at IAA with Kn, and NAA with or without BAP. The remaining accessions produced 0.09 to 1.15 microtubers per explant. Lower concentration of Kn (5 mg/l) with IAA and sucrose was favourable for producing microtubers (0.61/explant on an average), the best species being D. sansibarensis (1.27) followed by D. japonica (0.59/ explant). Shoot tip explants from in vitro germinated seedlings of Solanum capsicoides All. inoculated on MS containing 2 mg/l BA produced maximum shoot induction response (26 shoots per explant). Rooting of the microshoots (19.4 roots per explant) was obtained better in half strength of MS supplemented with NAA (0.5 mg/l). This study investigated in vitro production of diploid (AS2) and tetraploid (AS4) cytotypes of snowdrop anemone. The effect of plant growth regulators (PGRs) on in vitro shoot multiplication and rooting was investigated. The effect of activated charcoal (AC) on root induction was also studied. Ploidy level affected growth characteristics during multiplication and rooting. Shoot induction in AS4 was higher on medium supplemented with cytokinin (3.2–3.6), while the AS2 clone formed the most shoots on PGR-free medium (3.6). The highest rooting percentage was achieved on PGR-free medium in both genotypes (AS2 clone, 100% and AS4 clone, 93.3%). The addition of AC to the PGR media largely increased root induction and root length. Rooted plantlets were successfully acclimatised in the greenhouse with 100% survival. Thus, the described micropropagation protocol represents a rapid and effective in vitro propagation method for utilisation in horticulture and conservation programmes of snowdrop anemone.

An alternative protocol for in vitro propagation of Picrorhiza scrophulariiflora is described using bavistin and adenine sulphate. The explants differentiated into multiple shoot buds on MS supplemented with various concentrations of bavistin and adenine sulphate ranging from 0 - 400 mg/l either alone or in combination. Maximum number of multiple shoots were obtained on MS containing the combination of bavistin (100 mg/l) and adenine sulphate (100 mg/l). In this combination as high as 28 shoots per explant was achieved and also vetrification of the cultures were not recorded. This study also demonstrates that the bavistin has stronger cytokinin-like activity than adenine sulphate. For instance, it was observed that bavistin alone in the concentration of 300 mg/l produced as high as 24 shoots per explant, however, adenine sulphate (100 mg/l) could produce a maximum of 18 shoots per explant. Moreover, higher or lower concentration did not improve the shoot multiplication. The microshoots were separated from the multiple shoots and transferred to MS containing various concentrations of auxins. Among them, NAA (1 mg/l) produced as high as 6 roots per explant. The regenerated plantlets were hardened in plastic cups (6 x 8 cm) containing 9 : 1 virgin soil and soil at Kyongnosla nursery and acclimated for four weeks.
