An alternative protocol for in vitro propagation of Picrorhiza scrophulariiflora is described using bavistin and adenine sulphate. The explants differentiated into multiple shoot buds on MS supplemented with various concentrations of bavistin and adenine sulphate ranging from 0 - 400 mg/l either alone or in combination. Maximum number of multiple shoots were obtained on MS containing the combination of bavistin (100 mg/l) and adenine sulphate (100 mg/l). In this combination as high as 28 shoots per explant was achieved and also vetrification of the cultures were not recorded. This study also demonstrates that the bavistin has stronger cytokinin-like activity than adenine sulphate. For instance, it was observed that bavistin alone in the concentration of 300 mg/l produced as high as 24 shoots per explant, however, adenine sulphate (100 mg/l) could produce a maximum of 18 shoots per explant. Moreover, higher or lower concentration did not improve the shoot multiplication. The microshoots were separated from the multiple shoots and transferred to MS containing various concentrations of auxins. Among them, NAA (1 mg/l) produced as high as 6 roots per explant. The regenerated plantlets were hardened in plastic cups (6 x 8 cm) containing 9 : 1 virgin soil and soil at Kyongnosla nursery and acclimated for four weeks. An efficient protocol for plant regeneration through multiple shoots induction from shoot tips of Polygonum hydropiper (L.) was established. The highest percentage (96.6) of multiple shoot induction and number of shoots (9.0) per culture were found on MS supplemented with 2.0 mg/l Kn. The induced shoots were excised and inoculated on to MS contains different concentrations of IBA or NAA for rooting. The highest percentage (90.0) of root induction and the highest number of roots per shoot (12.0) was found on MS having 1.0 mg/l IBA. A rapid and efficient protocol is outlined for in vitro propagation of muskmelon(Cucumis melo L.) Shoot tips, nodal segments and cotyledonary nodes from invitro grown seedlings were used as explants. The explants were inoculated on MS medium fortified with different combinations and concentrations of growthregulators viz., BAP, NAA, GA3 and IBA for multiple shoot regeneration.Effective result was found on MS medium supplemented with 2.0 mg/l BAP, inwhich 90% and 70% cultures induced multiple shoots from nodal segments andshoot tip explants, respectively. Whereas, 70% cultures of cotyledonary nodeswere found to induced shoots on MS medium with 1.5 mg/l BAP + 0.1 mg/l GA3. In vitro regenerated shoots were subcultured on half strength MS mediumsupplemented with different concentrations of IBA and NAA for successful rootinduction and the effective result (up to 70%) was found in medium with 1 mg/lIBA. Well rooted in vitro grown plantlets were acclimatized in sandy soil, whereas 70% plantlets survived

An efficient protocol of in vitro propagation of Plectranthus bourneae Gamble (Lamiaceae), a valuable medicinal important and endemic Red listed plant of Western Ghats, (Tamil Nadu, India) was standardized by improved shoot multiplication from axillary bud explant. An in vitro propagation system has been reconnoitered on MS with the effective concentration BA (0.7 mg/l) followed by a combination of BA (0.7 mg/l) and TDZ (1.0 mg/l) which promoted high number of shoots. The multiple shoot rate was enhanced further by adding AdS (50 mg/l). Beneficial shoot length was achieved when cultured on MS containing GA3 (0.5 mg/l). Rooting was increased on MS augmented with IBA (1.5 mg/l). Micropropagated plants were acclimatized and the survival rate was 80%. Acclimatized P. bourneae plants can be used as substitute alternative to natural populations. In Vitro Propagation of Kepok Banana var. Unti Sayang Resistant to Blood Disease through Shoot ProliferationABSTRACTKepok is a popular banana variety but sensitive to blood disease caused by Ralstonia solanacearum (Smith). The discovery of a natural mutant of Kepok banana var. Unti Sayang from Sulawesi which male bud falls naturally, is a shortcut to bypass the chains of the spread of blood disease, since the disease is transmitted by insects through the wounds of the male buds. The superior mutant needs to be mass propagated and disseminated to endemic areas to inhibit the spread of blood disease. To achieve that goal, an efficient and effective techniques of in vitro shoot proliferation needs to be developed. Shoot proliferation was performed by addition of BAP, thidiazuron and adenine sulphate. The results showed that the best medium for shoot multiplication was B2T5A (MS+2 mg/L BAP+0,5 mg/L TDZ+20 mg/L adenine sulphate), and for shoot growth was B4A (MS+4 mg/L BAP+20 mg/L adenine sulphate). Rooting was induced on MS medium without hormones. Acclimatization of plantlets on mixed soil, compost and husks with a ratio of 1:1:1 resulted in 92,35% survival rate.Keywords: blood disease, in vitro shoot,  male budless, natural mutant, var. Ditemukannya mutan alami pisang kepok yang jantungnya gugur secara alami yaitu varietas Unti Sayang dari Sulawesi, merupakan jalan pintas untuk memotong rantai penyebaran penyakit darah, mengingat penyakit ini ditularkan oleh serangga melalui luka bekas bunga jantan pada jantung. Mutan unggul tersebut perlu diperbanyak secara massal dan disebarluaskan ke daerah endemik untuk menghambat penyebaran penyakit darah. Untuk mencapai tujuan tersebut, perlu dikembangkan teknik perbanyakan in vitro pisang kepok Unti Sayang yang efektif dan efisien melalui proliferasi tunas. Proliferasi tunas dilakukan dengan penambahan BAP, thidiazuron dan adenin sulfat. Hasil penelitian ini menunjukkan bahwa media terbaik untuk multiplikasi tunas adalah B2T5A (MS+2 mg/L BAP+0,5 mg/L TDZ+20 mg/L adenin sulfat), media terbaik untuk pertumbuhan tunas adalah B4A (MS+4 mg/L BAP+20 mg/L adenin sulfat). Akar dapat diinduksi pada media MS tanpa hormon. Aklimatisasi planlet pada media campuran tanah, kompos dan sekam dengan perbandingan 1:1:1 menghasilkan 92,35% planlet hidup.Kata Kunci: penyakit darah, tunas in vitro, tanpa jantung, mutan alami, var. Aim. Potato (Solanum tuberosum L.) is one of the important crops in Ethiopia which has a crucial role in nutritional security, poverty alleviation, and income generation. The aim of the present investigation is to develop an efficient in vitro propagation protocol for Belete and Gudiene potato varieties by using lateral bud as explants. Materials and Methods. Shoot initiation was achieved by inoculating buds on full-strength MS Murashige and Skoog medium (MS) fortified with variable concentrations of BAP and NAA. Basal MS was used as control throughout the experiment. Results. Results of our study showed that best shoot initiation was obtained on MS medium supplemented with 1.5 mg/l BAP + 3.0 mg/l NAA for Gudiene variety, whereas 1.0 mg/l BAP and 2.0 mg/l NAA produced more shoots in Belete variety. The initiated shoots increased two- to three-fold upon subculture on the MS medium fortified with varying concentrations of BAP and Kinetin. The highest numbers of multiple shoots were obtained in the MS medium containing 2.5 mg/l Kinetin. The combined effect of BAP and Kinetin did not produce any additional positive effect for shoot multiplication. Rooting percentage and number of roots/shoot were found best on the MS medium fortified with 1.0 mg/l IBA + 0.5 IAA. Conclusions. The variety Gudiene was found best for shoot initiation and root formation, while Belete variety proved its superiority for multiple shoot formation. A total number of 82.66% of plantlets were acclimatized under field conditions. This work indicates the practical applicability of plant tissue culture using lateral bud as explants is effective for micropropagation of potato in vitro.

Context: In vitro propagation or tissue culture of plants offers a rapid means of producing large quality of clonal planting stock and propagation that are difficult to establish conventionally. Biotechnological tools are important for multiplication and genetic enhancement of the plants by adopting techniques such as in vitro regeneration and genetic transformations.Objective: Effect of different plant growth regulators (PGRs) and their concentration on multiple shoot regeneration and callus formation was studied in Indigofera viscosa.Materials and Methods: In vitro plant regeneration was achieved in nodal and shoot tip explants. The explants were cultured on MS medium supplemented with BAP and NAA.Results: The nodal explants exhibited a greater number of healthy multiple shoots. The maximum callus induction was observed on MS medium supplemented with 1.5 mg L-1 2,4-D. NAA and BAP combination proved to be the most effective treatment for promoting shoot multiplication. IBA was found to be the best rooting hormone than IAA or NAA. The morphogenetic potential of parval or pointed gourd (Trichosanthes dioica Roxb.) shoot-tip explants was investigated to establish this species as a model tissue culture system. An effective multiple-shoot propagation method is described. Ten-millimeter shoot tips from young branches of greehouse-grown plants served as explants. They were initiated on a MS basal medium. Multiple shoots were encouraged by transferring established explants to a proliferation medium consisting of MSB + 1 mg BAP/liter, because lower concentrations of BAP (0.1 to 0.5 mg–liter–1) inhibited multiple shoot formation; however, the same concentrations promoted rooting in explants. Medium supplemented with 1 mg BAP/liter and 100 mg PVP/liter caused the best proliferation of shoot tips. Upon transferring to fresh medium of the same composition, these shoot tips elongated 24 cm with three to five nodes in 4 weeks of culturing. Shoot multiplication cultures were maintained by transferring segments of multiple-shoot clusters to medium containing 1 mg BAP/liter and 0.5 mg GA3/liter. Medium supplemented with TDZ inhibited the number of regenerating explants but enhanced the number of shoot buds. Eighty percent of these plantlets were successfully rooted on MS medium supplemented with 1 mg NAA/liter. Plantlets survived in potting soil and exhibited normal growth under mist in the greenhouse.

Efforts have been made to establish a protocol for direct multiple shoot regeneration from both in vitro grown seedlings and mature plants of Citrus macroptera. Both nodal and shoot tip explants taken from in vitro grown seedlings were cultured in MS supplemented with different concentrations of BAP and Kn either singly or in combinations. Both these explants are capable to regenerate and produce in vitro multiple shoots. Maximum number of shoots were obtained from nodal explants in MS supplemented with 1.0 mg/l BAP. BAP alone was found superior to Kn. On the other hand, only nodal explants from mature plants were used and 1.0 mg/1 BAP was also found best suitable for shoot induction and multiplication. A simple, repeatable and efficient protocol for direct multiple shoot regeneration from internodal explants has been defined in peppermint ( Mentha x piperita var. Indus). In vitro regenerated shoots of peppermint were excised into 4 to 8 mm long internodes and cultured on Murashige and Skoog's medium supplemented with different cytokinins. In the hormonal assay, 3.0 mg L-l zeatin or 6-isopentenyl adenine independently supplemented to half strength MS medium exhibited multiple shoot regeneration, while thiaduzorn (0.1-3.0 mg L−1) showed no morphogenetic effect. A maximum of 85% in vitro cultured explants showed multiple shoot formation with an average of 7 shoots per explant on MS medium supplemented with zeatin. Multiple shoots were initiated within three weeks of cultivation. Internodes with regenerated multiple shoots were transferred to half - strength MS medium without supplementing with any plant growth hormone for shoot elongation and rhizogenesis. Rooted plants acclimatized and grew to maturity under glasshouse conditions. The plantlets developed were phenotypically identical to the parent plant and exhibited 96 % survival.

An optimal plant propagation method of Physalis angulata L., a medicinally important herbaceous plant species has been developed using axillary meristem explants. Shoot bud proliferation was initiated from axillary meristem explants cultured on MS medium supplemented with various concentrations of 0.5-2.5mg/L/(BAP)/(Zeatin)/(KIN). The maximum in vitro response of shooting frequency of explants (88.1%) and shoots per explant (42) was achieved with medium containing 1.0mg/L BAP. Multiple shoot culture was established by repeated subculturing of the shoot buds of axillary meristems on shoot multiplication medium. Among the subculture media BAP in combination with 1.5mg/L (IAA)+0.25mg/L(GA3) produced maximum shoots per explant (128±0.29) after two weeks of culture. Effective in vitro shoot elongation and rooting was achieved on 1.0mg/L(GA3) and 1.0mg/L(IBA), respectively. Most of the generated shoots were successfully transferred to soil under field conditions. The survival percentage of the transferred plants on soil was found to be 90 per cent.  This protocol can be used for commercial propagation and for future genetic improvement studies.
