In this study, two isomeric impurities were identified in cefotiam hydrochloride injection preparation and were characterized. Column-switching HPLC-MS and NMR techniques were used to identify the impurity 1 as the Δ3(4) isomers of cefotiam. Using software-based calculations, it was predicted that neither of the isomeric impurities was embryotoxic. This study provides a reference for the production, storage, and quality control of cefotiam and related cephalosporin antibiotics.

Elsholtzia ciliata (Thunb.) Hyl, family Lamiaceae, is an important and popular anti-bacterial and anti-inflammatory Traditional Chinese Medicine (TCM). However, there are limited scientific studies on its anatomy and pharmacological activities. Moreover, the information of chemical constituents in relation to its non-volatile constituents are still missing. The current study aimed to evaluate the anatomic, pharmacological and phytochemical profile of Elsholtzia ciliata, providing means for the quality control of this herbal drug. The methodology designed for this study included the preparation of anatomic sections and their description, extraction, chromatography, structural elucidation of isolated compounds by NMR techniques and their quantification by HPLC using pharmacological assays (Formalin, hot plate, DPPH, antimicrobial-Gram positive, Gram Negative and fungus, and MTT assays) to confirm the activities described for this species. Results of the anatomic study are aligned with the pattern expected for plants belonging to the Lamiaceae family; Ursolic acid and Oroxylin were isolated from this plant species. The findings observed in this study indicate that Elsholtzia ciliata possess anti-inflammatory, antinociceptive, antioxidant, antimicrobial and anticancer activities. The chemical compounds isolated from its leaves and the anatomy profile of its parts provide the basis for further quality control for this plant.

Methods for spectrophotometric determination of cephalosporin antibiotics ceftazidime and ceftriaxone using azocoupling reactions with 8-hydroxyquinoline have been developed. Optimal conditions for the diazotization of antibiotics in hydrochloric acid (CHCl = 12 M) and subsequent azocoupling with 8-hydroxyquinoline in alkaline medium (0.16 M NaOH in the final volume) were established. The of azocopling products of violet and light red colors are formed, which are characterized by absorption maxima at 553 nm (ε553 = 6.31·103 M-1 cm-1) and 550 nm (ε550 = 1.45·104 M-1 cm-1) for ceftazidime and ceftriaxone, respectively. Stoichiometric ratios of the azo compounds components were determined using continuous variations methods and in both cases are 1:1. Sensitive methods of spectrophotometric determination of ceftazidime (LOD = 1.81∙10-6 M) and ceftriaxone (LOD = 5.92·10-7 M) have been developed. The selectivity of cephalosporin antibiotics spectrophotometric determination in the presence of auxiliary substance (sodium carbonate) has been investigated. The elaborated methods has been approved during the analyses of model solutions and the single component drug. The recommended procedure is well suited for the ceftazidime and ceftriaxone assay in medicine to assure high standards of quality control.

In the past most studies of the control of energy metabolism focus on the role of the phosphorylation potential ATP/ADP.Pi on the regulation of respiration. Studies using NMR techniques have demonstrated that the concentrations of these compounds for oxidation phosphorylation do not change appreciably throughout the cardiac cycle and during increases in cardiac work. Hence regulation of energy production by calcium ions, present in the mitochondrial matrix, has been the object of a number of recent studies.Three exclusively intramitochondnal dehydrogenases are key enzymes for the regulation of oxidative metabolism. They are activated by calcium ions in the low micromolar range. Since, however, earlier estimates of the intramitochondnal calcium, based on equilibrium thermodynamic considerations, were in the millimolar range, a physiological correlation was not evident. The introduction of calcium-sensitive probes fura-2 and indo-1 made monitoring of free calcium during changing energy metabolism possible. These studies were performed on isolated mitochondria and extrapolation to the in vivo situation is more or less speculative.
