 Technique of tissue culture for Chia (Salvia hispanica) micropropagation was achieved, this study investigated the impact of various concentrations of plant growth regulators on shoot multiplication and root induction with the Chia’s mature seed as a source explant. The highest percentage of shoot formation (80%), shoots number per explant(3.20) and shoot length(3.26 cm), were recorded on MS medium enriched with BAP(1.0 mgl-1) after eight weeks of seed culture. The optimal medium for the rhizogenesis was achieved on half strength MS medium fortified with 1.0 mgl-1 IBA after four weeks of culture, which had the highest rooting percentage (100%) with highest mean of roots number (5.6 roots per shoot) with (3.40 cm root length). The rooted plants were successfully adapted ex vitro with a survival rate of 85%.

The present study was undertaken with a view to develop an efficient protocol for in vitro multiple shoot formation and subsequent root induction considering various cultural aspects using nodal segments of Cucurbita maxima and Benincasa hispida. The best hormone for shoot multiplication of pumpkin was BAP (2 mg/l), incase of ash-gourd BAP was 1.5 mg/l. For callus induction BAP+2,4-D was best combination for pumpkin and it was 2.0+0.1 mg/l when in ash gourd BAP+NAA was the best combination. IBA + ½MS medium were used for induction of shoot bud root. In both of pumpkin and ash gourd 1.5 mg/l IBA was found best for induction of roots. Banana is a fruit crop which has high demand in Ethiopia, but its production is constrained by lack of disease free planting material with conventional propagation methods. For shoot initiation, shoot tip explants were cultured on MS medium supplemented with 0.5, 1.0, 1.5 and 2.0 mg/L BAP. Similarly, MS medium supplemented with BAP at 1.0, 1.5, 2.0 mg/L in combination with IBA at 0.25 and 0.50 mg/L were used for shoot multiplication. Half- strength MS medium augmented with IBA at 1.0, 2.0, 3.0 and 4.0 mg/l were used for root induction. MS medium without PGRs were used as controls. Finally, hardening of the in vitro derived plantlets was carried out in green house both in the primary and secondary acclimatization stages. Results showed that the highest shoot initiation percent (93.40%), highest mean number of shoots per explant (4.67) and lesser day for shoot induction (11.00) were observed in explant cultured on MS + 1.0 mg/L BAP. With shoot multiplication, highest shooting percent (92.60%), maximum number of shoots (7.67) and highest shoot length (5.27 cm) were recorded on MS + 1.5 mg/L BAP + 0.5 mg/L IBA. The highest rooting percent (93.40%), maximum root number per shoot (7.67) and highest root length (11.00 cm) were found on a half strength MS medium + 2.0 mg/L IBA. The survival rate of plantlets were 96.00% in coco peat substrate in primary acclimatization and 97.92% in forest soil, sand and manure substrates mixed at 3:2:1 ratio in secondary acclimatization. Overall, the result showed that the PGRs type, concentrations and combinations used are effective for mass propagation of banana variety studied in this experiment.

A simple, repeatable and efficient protocol for direct multiple shoot regeneration from internodal explants has been defined in peppermint ( Mentha x piperita var. Indus). In vitro regenerated shoots of peppermint were excised into 4 to 8 mm long internodes and cultured on Murashige and Skoog's medium supplemented with different cytokinins. In the hormonal assay, 3.0 mg L-l zeatin or 6-isopentenyl adenine independently supplemented to half strength MS medium exhibited multiple shoot regeneration, while thiaduzorn (0.1-3.0 mg L−1) showed no morphogenetic effect. A maximum of 85% in vitro cultured explants showed multiple shoot formation with an average of 7 shoots per explant on MS medium supplemented with zeatin. Multiple shoots were initiated within three weeks of cultivation. Internodes with regenerated multiple shoots were transferred to half - strength MS medium without supplementing with any plant growth hormone for shoot elongation and rhizogenesis. Rooted plants acclimatized and grew to maturity under glasshouse conditions. The plantlets developed were phenotypically identical to the parent plant and exhibited 96 % survival.

To evaluate an efficient protocol for the micropropagation of Tupistra albiflora K. Larsen, the effects of N6-benzylaminopurine (BA) and naphthalene acetic acid (NAA) concentrations on multiple shoot and root induction were examined. In vitro shoots were used as the explant materials which were cultured on Murashige and Skoog (MS) agar medium supplemented with 0, 1, 2, 3 and 4 mg/L BA for 4 weeks to induce multiple shoots. It was found that the MS medium containing 3 mg/L BA induced 100 % shoot formation with the highest number of 3.2 shoots per explant (2.4-fold significantly higher than the control). For root induction, in vitro shoots were cultured on MS agar medium supplemented with 0, 1, 2, 3 and 4 mg/L NAA for 8 weeks. The results showed that the MS medium containing 1 mg/L NAA induced 100 % root formation with the highest number of 6.6 roots per explant (1.8-fold significantly higher than the control).

In the present study the protocol for callus induction and regeneration in Azima tetracantha has been developed in culture medium. The young apical leaf explants were used for callus induction on MS medium containing BAP and NAA at 1.0 and 0.4mgl-1 respectively showed maximum callus induction (73%). The amount of callus responded for shoot formation (74%) was obtained in the MS medium containing BAP (1.5 mgl-1) and NAA (0.3mgl-1).The elongated shoots were rooted on half strength medium supplemented with IBA (1.5 mgl-1) and Kn (0.4 mgl-1) for shoots rooted. Regenerated plantlets were successfully acclimatized and hardened off inside the culture and then transferred to green house with better survival rate.

Context: Orchid produces a huge number of minute seeds but the seeds can not germinate easily in nature due to the lack of endosperm in the seeds is an incompatibility barrier that limits its propagation in nature. Objectives: To develop in vitro culture techniques for quick propagation of Vanda tessellate, a commercially important orchid species. Materials and Methods: Shoot tips were used as experimental materials. The explants were surface sterilized and the shoot tips were excised. The isolated shoot tips were cultured in MS medium supplemented with different concentration and combinations of auxin and cytokinin. Results: The combination of 1.5 mgl-1 NAA and 1.0 mgl-1 BAP was proved to be the best medium formulation for multiple shoot formation as well as maximum shoot elongation. The single shoots were isolated from the multiple shoots and subcultured in MS medium having NAA and IBA individually and in combinations for root induction. Maximum root induction was obtained in MS agarified medium having 0.5 mgl-1NAA and 1.0 mgl-1IBA. The well rooted plantlets were hardened successfully in the potting mixture containing coconut husk, perlite, charcoal, brick pieces in the ratio of 2:1:1:1 and eventually established under natural condition.Conclusion: An efficient regeneration protocol for micropropagation in V. tessellata through shoot tip culture has been established.Key words: Shoot tip; micropropagation; orchid.DOI: 10.3329/jbs.v17i0.7122J. bio-sci. 17: 139-144, 2009

Anther of five varieties of Brassica species, namely BARI Shariaha-7, Tori-7, Agrani, Daulat and Safal were cultured in vitro to observe their regeneration potentiality. Different concentrations and combinations of growth regulators were supplemented in MS medium. The range of callus induction was 12.50-87.50 %. Maximum callus induction (75.00%) was observed on MS +4 mg/L 2, 4-D + 1.0 mg/L BAP. Among the genotypes, BARI Sharisha-7 showed the highest percentage of callus induction (60.42%). Among the treatments, highest percentage of shoot regeneration (75.00%) was observed on MS + 4 mg/L BAP + 1.0 mg/L NAA. BARI Sharisha-7 also showed the highest rate of plant regeneration (66.67%). Root induction was highest (75%) on half strength MS medium supplemented with 1.0 mg/L IBA and 0.5 mg/L NAA. The plantlets with sufficient roots thus obtained were transferred successfully to plastic pots and subsequently to the field. BARI Sharisha-7 and Tori-7 survived easily in the pots as well as in the field but Safal was very poor in survivability both in the pots and in the field. Micropropagation of three Echinacea species, E. angustifolia, E. pallida, and E. purpurea, was investigated as a potential means of germplasm preservation of species faced with overcollection in the wild and rapid clonal propagation of elite individuals with unique medicinal or ornamental properties. Comparison of explant sources indicated vegetative explants resulted in high contamination rates when collected from shoot-tips (100%),but not when collected from nodal explants (11% to 39). Seed coat removal reduced contamination from 100% in intact seeds to near 0% in excised embryos. Removal of seed coats (pericarp and integument layers) also eliminated dormancy requirement for germination. All species responded with shoot multiplication and loss of rooting when BA or thidiazuron was added to culture medium. Medium with thidiazuron resulted in excessive adventitious shoot formation. Shoot multiplication rates were low (one to three shoots/explant) on medium with BA levels low enough to avoid adventitious shoot formation. Medium containing half-strength MS minerals resulted in more shoots with smaller leaves than full-strength MS minerals. Cultures did not perform well on Woody Plant Medium. Increasing subculture frequency from every 4 weeks to every 2 weeks increased shoot multiplication rates from 1.4 to 1.8 shoots per subculture and total shoots produced after 12 weeks of culturing (per initial explant) from 2.8 to 23.9. Rooting occurred readily on shoots isolated from E. purpurea without addition of IBA. Rooting was low or non-existent on shoots from E. angustifolia and E. pallida, respectively, regardless of IBA level, light conditions, or temperature. Methods described in this study allow rapid multiplication of three Echinacea species and subsequent rooting of E. angustifolia and E. purpurea. Future improvements in root induction treatments will allow more effective use of micropropagation for Echinacea germplasm preservation and multiplication. Abstract Background Moringa stenopetala belongs to the flowering family Moringaceae and genus Moringa. It is often referred to as the East African Moringa tree because it is native only to southern Ethiopia and northern Kenya. The expansion of its cultivation and utilization throughout the world especially in Africa is becoming important. For such expansion, the existing propagation method is limiting, so it needs a good propagation system to supply enough planting material with a uniform genotype. Therefore, the main objective of this study was to optimize an in vitro shoot multiplication protocol for M. stenopetala by using shoot tip as explants. Results Shoots were sterilized and cultured on Muraghige and Skoog (MS) medium for in vitro shoot initiation. For multiple shoot induction, the explants were cultured on MS medium supplemented with different concentrations of kinetin (0.5, 1.0, 1.5, 2.0, 2.5 mg/L) with Indole-3- butyric acid (IBA) or α -naphthalene acetic acid (NAA) (0.01, 0.1, 0.5 mg/L) and maintained at 25 ± 2 °C for four weeks. Rooting was achieved by culturing well developed shoots in half-strength MS medium containing IBA (0.1, 0.5, 1.0, 1.5, 2.0 mg/L), NAA (0.1, 0.5, 1.0, 1.5, 2.0 mg/L), and 0.5 mg/L IBA with NAA (0.1, 0.5, 1.0, 1.5, 2.0 mg/L). Statistical analysis revealed that there was a significant difference among all treatments applied in both shoot multiplication and rooting experiments. The maximum number of shoots per explant (3.43 ± 1.41) and 7.97 ± 4.18 leaves per explant were obtained on MS medium containing 0.5 mg/L kinetin with 0.01 mg/LNAA. The highest mean number of roots per shoot (1.63 ± 1.03) and mean root length (0.87 ± 1.22 cm) were obtained on MS medium containing 1.0 mg/LNAA and 0.1 mg/LIBA alone respectively. After acclimatization, 76% of plants were survived in the greenhouse. Conclusion In general, using NAA with kinetin for shoot multiplication was effective than kinetin with IBA. On the other hand, the application of 1.0 mg/L NAA alone and 1.0 mg/L NAA with 0.5 mg/L IBA were more effective for root induction.

An in vitro regeneration protocol of Adhatoda vasica has been developed using excised nodal segments and juvenile leaves for multiple shoots regeneration directly or through callus induction. Explants were cultured on MS medium with different concentrations of IAA, NAA, BAP, GA3 and Kn singly or in combinations. MS medium supplemented with BAP (10.0 mg/l) was found best for multiple shoot formation, in which 93.33% explants produced multiple shoots. After two months, maximum number of multiple shoots were 10.6 ± 1.82, highest length of plantlets was 5.2 ± 2.20 cm. 100% calli formation were observed on MS medium supplemented with IAA (0.05 mg/l) + NAA (0.05 mg/l) + BAP (1.0 mg/l). Callus initiation started after 14 days and gave light green colored callus. Best callus mediated shoot regeneration was found on MS+10.0 mg/l BAP medium. Root induction of in vitro raised shoots was best on ½ MS + IBA (1.0 mg/l). Well rooted plantlets were transferred to plastic pots containing garden soil and compost in a ratio of 2:1 for hardening.
