ABSORPTION AND FLUORESCENCE SPECTRA OF THE COMPLEXES OF METHYLENE BLUE AND ACRIDINE ORANGE WITH POLY(rA)-POLY(rU)

The interaction of methylene blue (MB) and acridine orange (AO) with poly(rA)-poly(rU) was studied using absorption and fluorescence spectroscopy methods. The absorption and fluorescence spectra of complexes of these ligands with a polynucleotide were obtained, similar to those of complexes of these ligands with double-stranded (ds-) DNA. It was revealed that the isosbestic point in the spectra of the AO-poly(rA)-poly(rU) and MB-poly(rA)-poly(rU) complexes is not formed, although the binding of AO and MB with ds-RNA by intercalation mode is not excluded.

Absorption and fluorescence spectroscopy techniques were applied to investigate the photophysical characteristics of acridine orange (AO) dye in solvents that included distilled water, dimethyl sulfoxide (DMSO), acetone and ethanol in various concentrations (1×10-4–1×10-6) M. All of the samples were served at room temperature. The relationships between various parameters describing the strength of optical transitions in atoms and molecules were reviewed. This study expresses various viewpoints by describing how concentration and solvent affect the dye's absorption and fluorescence spectra. The absorption spectra of AO exhibit a band at (490 nm), except for DMSO, which shifts more towards red by 5 nm. The fluorescence spectra show a blue shift in AO aqueous solution around 6 nm until (0.5×10-4) M, followed by a red shift at around 7 nm at (1×10-6) M. There is a blue shift in (1×10-5) M for DMSO at around 4 nm, then a 10 nm red shift in higher concentrations as well as a 9 nm red shift in acetone and 6 nm in ethanol. Adding magnesium oxide nanoparticles (MgO NPs) quenched AO in both absorption and fluorescence spectra, whereas maximum fluorescence and intensity increased when aluminium oxide nanoparticles (Al2O3 NPs) were added to the solution. Study on the interaction of DNA-specific ligands – classical intercalator acridine orange (AO) and groove binding compound Hoechst 33258 (H33258) with poly(rA)-poly(rU), being a model for double-stranded (ds-) RNA, has been carried out. The absorption and fluorescence spectra of the complexes of these ligands with ds-polynucleotide were obtained. It was revealed that the optic and fluorescent characteristics of the complexes of both ligands with ds-RNA are similar with those at the complex-formation with DNA.

Sensitive and selective detection of antibiotic residues is an important issue for human health, the environment and public safety. In this work, a study of the spectral-luminescent properties of the complex of sulgin and methylene blue in water was carried out. The methods of spectroscopy of electronic absorption and fluorescence are used to assess the properties of the studied molecules, which enter into specific and van der Waals, hydrophobic and electrostatic interactions between sulgin and methylene blue. It has been shown that the constant of interaction between sulgin and methylene blue is 2.5×103 M-1.
