Glutathione system activity in the blood of overweight postmenopausal women

One of the important components of the antioxidant defense system is the glutathione system, the activity of which, when overweight, changes direction depending on gender and ethnicity. The results of studies involving overweight menopausal women are mixed. The study involved 61 postmenopausal women, who, after clinical and anamnestic examination, were divided into 2 groups: control (BMI = 19-24.9 kg / m2) and overweight group (BMI = 25-29.9 kg/m2). The use of hormone replacement therapy; the use of antioxidant drugs; diseases of endocrine genesis; exacerbation of chronic diseases; premature early menopause; surgical menopause was the exclusion criteria for women from the study. The lipid profile parameters with the calculation of the atherogenic coefficient; reduced and oxidized glutathione levels with the calculation of their ratio, the glutathione S-transferase and glutathione reductase activities were determined in the blood. Overweight women showed an increase in the triacylglycerols (p = 0.041) and cholesterol in very low density lipoproteins levels (p = 0.044). When assessing the glutathione system activity in women of the main group, compared with the control, an increase in the glutathione-S-transferase (p = 0.023) and glutathione reductase (p = 0.022) activities was noted, however, the reduced and oxidized glutathione levels, as well as their ratio did not differ from the control values. The results obtained indicate the activation of the glutathione system enzymatic link in response to changes in lipid status in postmenopausal women with overweight.

The article  deals with the results of experimental research data of relationship between the antioxidant defense system and lipid body peroxidation of high calves cows  under  the influence of nanpreparation Germakap conducted  with simultaneous vaccination with inactivated formol vaccination with repeated  in two weeks later  at the same doses as their  stability and immunity to Salmonella. It was found the likely increase in activity of glutathione peroxidase and glutathione reductase and at the same time, reduction of malon dialdehyde and hydroperoxides lipids and superoxidimutase. These changes in animals  body occur due to complex components adaptive nan preparation Germakap that lead to the normalization of metabolic and free radical processes in animals.  However, the increase in the catalytic activity of glutathione peroxidase and glutathione reductase activity in plasma of high calves cows  from research group can be explained by increasing intensity of synthesis in which these enzymes by introducing nan preparation Germakap closely associated with the regeneration of glutathione in the cell, and also the activity of glutathione peroxidase.  Through interaction with restoration of glutathione and glutathione peroxidase, glutathione reductase it was formed glutathione system that protects cells from stress peroxidation

The state of the glutathione-dependent link of the antioxidant blood system of rats under obesity development and with the compatible consumption of the kidney bean (P. vulgaris) pods extract was studied. Glutathione system forms functional basis of organism antioxidant defense system and disturbances in its work can lead to a number of serious cytotoxic and destructive lesions. Glutathione system constituent elements has its own glutathione and enzymes, which catalyze the reaction of its reverse transformation. These include glutathione peroxidase, glutathione transferase, and glutathione reductase. It is known that the water kidney bean (P. vulgaris) pods extract has hypoglycemic properties. However, complex studies of the polyfunctional action of this extract in obesity are absent. The obtained results indicate a decrease in the overall potential of the glutathione system in animals on a high-calorie diet. It has been established that glutathione peroxidase activity (GP) decreases in the serum of rats under experimental obesity, also glutathione transferase (GT) and glutathione reductase (GR) activity increase compared to the corresponding values in control animals. Our results demonstrated the concentration decrease of recovered glutathione (GSH) under consumption of high-calorie diet compared to the control. The findings after 6-th weeks of kidney bean (P. vulgaris) pods extract consumption indicate the positive modulating effect of this extract on the content of GSH and the activity of GP, GT and GR in rats with the obesity development. Probably, the consumption of this extract leads to attraction of the investigated components of the antioxidant system to the reactions of neutralization of toxic free radical compounds that were formed against the background of the development of obesity. Further studies may serve as the basis for the development of new therapeutic or medicinal preparations that based on the kidney bean (P. vulgaris) pods extract, because of still no consensus on the treatment of obesity.

Aim. To study the polymorphism of GSTM1, GSTT1 genes. To study the activity of enzymes encoding these genes in long-livers with arterial hypertension (AG) and osteoarthritis (OA). Methods. The determination of the frequency of polymorphous variants of gene deletions GSTT1 and GSTM1 and the activity of glutathione S-transferase (GST), glutathione reductase (GRD), glutathione peroxidase (GPO) was performed in long-livers, who were separated depending on the presence AG and OA. Results. In the group of patients with hypertension and OA genotype GSTM1«+»/GSTT1«+» met the GSTM1«+»/GSTT1«-» 3.0 times more often. The genotype GSTM1«-»/GSTT1«+» in comparison with GSTM1«-»/GSTT1«-» in this study group was 3.17 times. The study of GPO, GRD, and GST enzymes has shown a tendency to depend on its level from the health of long-livers. Conclusions. The advantage of genotypes with the active form of the gene GSTT1«+» was spotted among healthy long livers and patients with hypertension and OA. It indicates a better functioning of the antioxidant defense system. The activity of the enzymes of the glutathione system GPO, GRD, and GST was higher in healthy long-livers livers than in patients with comorbid pathology. The chronic effect of environmental methomyl on the antioxidant system in testis of Nile tilapia (Oreochromis niloticus) and its recovery pattern was investigated. Tilapia were exposed to sublethal concentrations of 0.2, 2, 20 and 200 μgL−1 methomyl for 30 days and thereafter moved to methomyl-free water for 18 days. Antioxidant levels in testis, including glutathione peroxidase, catalase, glutathione-S-transferase, glutathione reductase, superoxide dismutase, reduced glutathione, oxidized glutathione were measured every 6 days during the period of exposure, and at 18 days after being transferred to methomyl-free water. The results showed that lower methomyl concentration (0.2 μgL−1) had no effect on the above antioxidants, thus 0.2 μgL−1 could be seen as NOAEL for methomyl to tilapia. However, higher methomyl concentration of 2, 20 and 200 μgL−1 could significantly influence the above antioxidants. Glutathione peroxidase and oxidized glutathione increased significantly. On the contrary, reduced glutathione decreased significantly. Catalase, superoxide dismutase, glutathione reductase, glutathione-S-transferase increased at lower methomyl (2 and 20 μgL−1), but decreased at higher methomyl (200 μgL−1). The recovery test showed that oxidative damage caused by lower methomyl of 2 and 20 μgL−1 was reversible, and oxidative damage caused by higher methomyl of 200 μgL−1 was irreversible within 18 days of recovery period.

Abstract. Background: Aortic dilatation in Marfan syndrome (MFS) is progressive. It is associated with oxidative stress and endothelial dysfunction that contribute to the early acute dissection of the vessel and can result in rupture of the aorta and sudden death. We evaluated the participation of the glutathione (GSH) system, which could be involved in the mechanisms that promote the formation and progression of the aortic aneurysms in MFS patients. Patients and methods: Aortic aneurysm tissue was obtained during chest surgery from eight control subjects and 14 MFS patients. Spectrophotometrical determination of activity of glutathione peroxidase (GPx), glutathione-S-transferase (GST), glutathione reductase (GR), lipid peroxidation (LPO) index, carbonylation, total antioxidant capacity (TAC), and concentration of reduced and oxidized glutathione (GSH and GSSG respectively), was performed in the homogenate from aortic aneurysm tissue. Results: LPO index, carbonylation, TGF-β1, and GR activity were increased in MFS patients (p < 0.04), while TAC, GSH/GSSG ratio, GPx, and GST activity were significantly decreased (p < 0.04). Conclusions: The depletion of GSH, in spite of the elevated activity of GR, not only diminished the activity of GSH-depend GST and GPx, but increased LPO, carbonylation and decreased TAC. These changes could promote the structural and functional alterations in the thoracic aorta of MFS patients.
