The validation of an ovarian biopsy method for in vivo assessment of oocyte maturation in Centropomus parallelus was studied. Diameters of intra-ovarian oocytes siphoned with cannula were analyzed fresh and preserved with 1% formalin in 0.7% NaCl solution. Oocytes in different stages were present along the ovaries, up to the tertiary yolk globule stage, which had a unimodal diameter frequency distribution. The oocyte diameter means were not significantly different at four sites along the ovaries (P > 0.05). Samples obtained with cannula were representative of the ovary central portion, in vivo and in vitro samples of the seven females examined were not significantly different (P > 0.05). An estimate of the coefficient of variation corrected for bias (P < 0.05) for 8 repeated in vivo samples was 1.9 ± 0.6. The results demonstrated that for the species, the biopsy method was satisfactory, providing representative samples of the ovaries.

We examined the relationship between five stages of ovarian growth and development and hepatic microsomal thyroxine (T4) deiodinating activity in rainbow trout, Oncorhynchus mykiss, held in the laboratory at 11.5°C. Thyroxine outer-ring deiodination (T4ORD) activity, which forms biologically active 3,5,3'-triiodothyronine (T3), was low when the gonadosomatic index (GSI = (ovary mass X 100) / body mass) was below 0.2 (stage I, previtellogenesis), but rose sharply between GSI values of 0.2 and 0.3 (stage II), when oocyte Balbiani activity, RNA synthesis, and endogenous vitellogenesis were maximal. Hepatic T4ORD activity was again low during exogenous vitellogenesis at GSI values of 0.3-12, which correspond to stages III-V. Hepatic T4 inner-ring deiodination (T4IRD), which forms biologically inactive 3,3',5'-triiodothyronine, was detected only at stage II. Plasma T3 concentrations were significantly higher at stage II than at stage IV (primary yolk globule formation). Our data indicate changes in systemic thyroidal status at stage II and are consistent with a role for T3 in early oogenesis in trout. Enhanced hepatic T3 production at the late Balbiani stage may be required for oocyte development.

A new type of globular particle, the ‘insoluble yolk globule’, was isolated from the egg yolk of three avian species (hen, duck, and emu) by centrifugation or gel-filtration chromatography. These globules are stable in NaCl and urea solutions at concentrations that dissolve or disrupt other constituents of yolk, The isolated globules are about 1% of the dry yolk of hen's and duck's eggs but about 8% emu's-egg yolk. Most of these globules are less than 2 micrometer in diameter. Electron micrographs of sections show a preponderance of globules in the range 0.125-0.25 micrometer, each with a thick shell surrounding a feature-less anterior. Globules with the same appearance were seen in sections of unfractionated yolk. Two kinds of larger particles were also observed: (i) particles with a distinct outer membrane and a vesiculated interior; (ii) featureless spheres, possibly of lipid. The insoluble yolk globules comprise protein (8-11% by dry wt.), phospholipid (31-35% total lipid), triacylglycerols (49-53%), cholesterol (8%) and cholesteryl esters (2-3%); the variations being among species. The phospholipid is accessible to phospholipase C. The isolated protein is heterogeneous and resembles the apoprotein from the yolk low-density lipoprotein.
