Cooling and freezing processes cause physical and chemical damage to sperm by cold shock and oxidative stress. This study aimed to evaluate the effect of two antioxidants on sperm parameters of cooled and frozen-thawed ram semen diluted in an egg yolk-based extender. Semen was collected from 30 rams and processed in two consecutive experiments to test the inclusion of different concentrations of quercetin and butylated hydroxytoluene (BHT) in an egg yolk-based semen extender. Dimethyl sulfoxide (DMSO) was added as a solvent to the semen extender in a ratio of 1 mL DMSO for 90 mg of quercetin and 1 mL DMSO for 880 mg of BHT. After collection, semen was diluted at 200 × 106 motile sperm/mL (control) and split into different groups in each experiment. In experiment 1, semen was diluted with the extender containing quercetin (Q5, 5 μg/mL; Q10, 10 μg/mL; Q15, 15 μg/mL) or DMSO alone (DMSO1, 0.055 μL DMSO per mL; DMSO2, 0.165 μL DMSO per mL). In experiment 2, semen was diluted with the extender with BHT (BHT1, 0.5 μg/mL; BHT2, 1 μg/mL; BHT3, 1.5 μg/mL) or DMSO alone (DMSO3, 0.375 μL DMSO per mL; DMSO4, 1.125 μL DMSO per mL). After dilution, the semen was divided into two aliquots. Treated ram sperm samples were also subjected to different storage methods. The first set of samples was cooled at 5 °C for 24 h, whereas the second set of samples was frozen-thawed. Sperm motility parameters and plasma membrane integrity (PMI) were evaluated immediately after dilution (0h) and 24 h after cooling and in the frozen-thawed samples via computer-assisted sperm analysis and epifluorescence microscopy, respectively. The inclusion of quercetin or BHT did not affect sperm motility parameters or PMI of fresh, cooled, or frozen-thawed sperm in this study (P < 0.05). However, further studies are needed to test the effects of these antioxidants on the fertility of cryopreserved ram semen.

Pickering high internal phase emulsions (HIPEs) are gel-like concentrated emulsions that have the potential to be an alternative to partially hydrogenated oil (PHO). In this study, egg yolk granules (EYGs), natural complexes of protein and lipid isolated from egg yolk, were used as an emulsifier to prepare Pickering HIPEs. Gel-like HIPEs with an oil phase volume fraction of 85% and with an emulsifier concentration of only 0.5% could be prepared by using EYGs as an emulsifier. The EYGs were able to form stable HIPEs at NaCl ionic strengths over 0.2 M and at pH over 5.0 with NaCl ionic strength of 0.3 M. The EYGs, which could stabilize HIPEs, were easily to adsorb and cover the oil-water interface to form emulsion droplets with small particle size. In addition, interacting EYGs in the aqueous phase formed a continuous network structure, and the oil droplets packed closely, exhibiting high elasticity and shear thinning behavior. Furthermore, the formed HIPEs had suitable storage stability with no significant changes in appearance and microstructure after storage for 60 days. This work can transform traditional oils from liquid-like to solid-like by using EYGs to enrich food processing diversity and improve the storage stability of oils while reducing the intake of PHO and providing a healthier diet for consumers.

Quail (Coturnix coturnix japonica L.) egg yolk is one of the high-fat foods which can trigger hyperlipidemia. The condition of hyperlipidemia can have an oxidative stress effect on the brain. Superoxide dismutase (SOD) is a natural antioxidant that acts as a defense mechanism against oxidative stress. The inhibition rate of SOD decreases when oxidative stress occurs. This study aims to determine the effect of quail egg yolk on the SOD inhibition rate of brain organs on a rat. This study used male Wistar rats aged 2-3 months with 200-300 grams of weight. The rats were divided into two groups. Each group was fed with ad libitum for two weeks. The A groups as control continued ad libitum consumption, and the B group was given additional quail egg yolk 5 ml / 200 g BW for 2 weeks. At the end of the study, the rats were terminated. The brain organs were examined for SOD inhibition rate with spectrophotometry. The mean SOD inhibition rate in the A and B groups was 74.14% ± 6.16 and 24.14% ± 5.65, respectively. The independent t-test showed significant differences in SOD inhibition rate between groups (p 0.001). Furthermore, quail egg yolk significantly reduced the SOD inhibition rate in the brain organ of the rat.

The purpose of this study was to investigate whether the addition of chlorogenic acid (CGA) to a sheep semen extender could improve the quality of chilled sheep sperm. Ejaculates (n = 80) were collected from five Hu rams with an artificial vagina. The ejaculates were mixed and divided into five equal parts, diluted with a CGA-free Tris–egg yolk extender (control), or supplemented with 0.2, 0.4, 0.8, and 1.2 mg/mL. The sperm kinematic parameters (viability, progressive motility), functional integrity of plasma membrane and acrosome, adenosine triphosphate (ATP) concentration and antioxidant parameters (Catalase (CAT), Superoxide dismutase (SOD) activity, total antioxidant capacity (T-AOC), ROS level and Malondialdehyde (MDA) content) were evaluated during storage of the semen. The results indicated that: PM, plasmatic membrane integrity and acrosomal integrity in 0.8 mg/mL CGA were higher (p < 0.05) from day 1 to 5. The ROS level in CGA groups was lower than the control (p < 0.05). CAT, SOD, ATP, and T-AOC were highest at 0.8 mg/mL concentration within 1 to 5 days. The above results indicated that the right concentration of CGA improved the quality of Hu ram sperm during chilling storage.

In this modern era of changing lifestyles, people and children cannot follow the rules of Dincharya and Ritucharya. Children have insufficient nutrients due to irregular food habits, which are triggering factors that cause childhood Karshya, resulting in mental and physical conditions that will not adequately develop. Biological and mentally healthy children are vital parts of a well-developed Nation, and this can be done by supplying nutritional requirements. The research was conducted in Bal Rog OPD, SAC and Hospital Lucknow. In this clinical trial total of 50 patients were selected and divided into group A and group B, having 25 in each group. Group A was treated with Egg Yolk Basti, and group B was treated with Vidarikandadi Granules for three months. In group A, 14 (56%) patients were relieved, 7 (28.0%) patients were moderately improved, 3 (12.0%) patients were mildly improved, and one patient did not show any response. In group B, 13 (52%) patients were relieved, 8 (32.0%) patients were moderately improved, 3 (12.0%) patients were mildly improved, and one patient did not show any response. This study indicates that Egg Yolk Basti and Vidarikandadi Granules can be effective in childhood Karshya.

The objective of this study was to determine the immunomodulatory effects of egg yolk protein–water extract (EYW) on splenocyte proliferation, cytokine secretion, immunoglobulin production, and NK cell cytotoxic activity in BALB/c mice. The forced swimming test (FST) was used to provide a model for suppressing immune regulation. The proliferation of B cells in the EYW supplementation group was significantly increased from the level to which it was reduced by the FST (from 40.9% to 81.8%, p < 0.05). EYW supplementation affected cytokine secretion of splenocytes. Levels of interleukin (IL)-2 and IL-10—as Th1 and Th2 cytokines, respectively—were decreased after the FST. However, EYW supplementation showed that secretion levels of these cytokines were significantly increased to pre-FST levels (p < 0.05). The production of immunoglobulins (IgA and IgG) was increased abnormally after the FST, whereas EYW supplementation significantly decreased it to pre-FST levels (p < 0.05). EYW supplementation also improved NK cell cytotoxic activity against YAC-1 tumor cells compared to the PC group (p < 0.05). These data suggest that EYW has potential as an immunomodulatory agent in the food and/or pharmaceutical industries.

Purpose: Conducting a honey diluent test for creeples of sperm of a drone honey bee.Materials and methods. The material for the research served a sperm of the milled drone drums of the "Prioksky" type of the Midway breed of bees. The selection of sperm was carried out in June-July 2020 g by the method of artificial stimulation of the turning of the endofalosha in half-armed drones aged 25-30 days. The rock type "Prioksky" of the middle Russian breed of bees. Before freezing, the sperm was stored in glass capillaries in the cooled state at 3 ° C for 2 months. The following composition of the diluent was tested - 10% honey, lactose, sucrose, egg yolk and dimethyl sulfoxide.Results. Studies have shown the viability of sperm at 64.0 ± 1.8% (41.5-83.7), and a total mobility of 2.2 ± 0.6% (0-11.5). To evaluate the fertilizing ability of sperm, carried out artificial insemination of 10 bee modules. In 4 seeded bees dykens, the presence of sperm in a seed-hearter with a concentration of sperm from 0.22-4.4 million / μl is revealed. In paired eggs of three other seeded matters, the presence of sperm and the complete absence of spermatozoa in the seed-receptionist are recorded.Conclusion. Tests of the honey diluent for deep freezing sperm of the drone honey bees in liquid nitrogen confirmed its cryophylactic properties.

The aim of the research is to determine the effect of changes in the concentration of estradiol-17β on the concentration of vitellogenin at the stage of egg shell formation and preparation of the body of meat and egg breed chickens (for example, the Pushkin breed) for ovulation.Materials and methods. The material for the study was the Pushkin breed chickens, in the amount of 28 heads. at the age of 48 weeks. Chickens were kept in individual cages. Light mode - 14C:10T. Drinking - nipple, temperature in the house and feeding - according to the standards adopted in the bioresource collection of RRIFAGB "Genetic collection of rare and endangered chicken breeds." Within 10 days prior to blood sampling, an ultrasound scan of the ovary and oviduct was performed from the studied group of chickens, which made it possible to determine the stage of egg formation. Blood from the studied chickens was taken in the morning on an empty stomach for the simultaneous determination of the concentration of estradiol and vitellogenin at the stage of egg shell formation. The concentration of estradiol and vitellogenin in the blood plasma of chickens was determined by ELISA. To clarify the effect of the concentration of estradiol (E2) on vitellogenin (VTG), the studied flock was divided into 2 groups according to the concentration of vitellogenin: 1st gr. – > Mav. (> 20 μg / ml) and 2nd gr. – < Mav. (<20 μg / ml). During the study period, eggs of chickens were evaluated according to the following morphometric parameters: the weight of the egg, yolk, protein and shell, elastic deformation, shape index, density of protein fractions, thickness of the shell and subshell, marbling. The number of eggs laid was determined in 10-day trial period.Results. It has been established that the duration of the ovulatory cycle in Pushkin breed chickens is 27-29 hours. As a result of such a long ovulatory cycle in the following days, laying of eggs occurs later with a progressive shift, as a result of which the laying cycle on average for the studied group of chickens is 3 days. The E2 level in the 1st group was 258,48 ± 30,60 pg/ml (p < 0,05), which significantly differed from the 2nd group – 181,45 ± 13,81 pg/ml. A positive correlation was found between VTG and E2, r = 0,5 (p < 0,001).Conclusion. The VTG level significantly increases in Pushkin chickens at the age of 48 weeks with an increase in the concentration of E2 in the blood. A positive correlation was noted between these features. According to the results of our research, the high content of estradiol and vitellogenin in the chickens blood serum did not have a significant effect on egg productivity.
