The validation of an ovarian biopsy method for in vivo assessment of oocyte maturation in Centropomus parallelus was studied. Diameters of intra-ovarian oocytes siphoned with cannula were analyzed fresh and preserved with 1% formalin in 0.7% NaCl solution. Oocytes in different stages were present along the ovaries, up to the tertiary yolk globule stage, which had a unimodal diameter frequency distribution. The oocyte diameter means were not significantly different at four sites along the ovaries (P > 0.05). Samples obtained with cannula were representative of the ovary central portion, in vivo and in vitro samples of the seven females examined were not significantly different (P > 0.05). An estimate of the coefficient of variation corrected for bias (P < 0.05) for 8 repeated in vivo samples was 1.9 ± 0.6. The results demonstrated that for the species, the biopsy method was satisfactory, providing representative samples of the ovaries.

Fluoride conversion coatings on Mg present many advantages, among which one can find the reduction of the corrosion rate under “in vivo” or “in vitro” conditions and the promotion of the calcium phosphate deposition. Moreover, the fluoride ions released from MgF2 do not present cytotoxic effects and inhibit the biofilm formation, and thus these treated alloys are very suitable for cardiovascular stents and biodegradable orthopedic implants. In this paper, the biodegradation behavior of four new magnesium biodegradable alloys that have been developed in the laboratory conditions, before and after surface modifications by fluoride conversion (and sandblasting) coatings, are analyzed. We performed structural and surface analysis (XRD, SEM, contact angle) before and after applying different surface treatments. Furthermore, we studied the electrochemical behavior and biodegradation of all experimental samples after immersion test performed in NaCl solution. For a better evaluation, we also used LM and SEM for evaluation of the corroded samples after immersion test. The results showed an improved corrosion resistance for HF treated alloy in the NaCl solution. The chemical composition, uniformity, thickness and stability of the layers generated on the surface of the alloys significantly influence their corrosion behavior. Our study reveals that HF treatment is a beneficial way to improve the biofunctional properties required for the studied magnesium alloys to be used as biomaterials for manufacturing the orthopedic implants.

The effect of minor stressful stimuli (saline infusions or venepunctures) on lipid mobilization (estimated by plasma free fatty acids (FFA) and glycerol levels) was studied in two strains of conscious rabbits of various ages. Infusions of 0·9% (w/v) NaCl solution induced a prompt increase of FFA levels in naturally obese rabbits while this effect was absent in younger lighter animals. Administration of propranolol (a β-adrenoceptor blocking drug) beforehand did not prevent lipid mobilization. Adipose tissue in vitro was completely unresponsive to the lipid mobilizing action of adrenaline. The initial output of ACTH induced by stressful stimuli was quickly followed by the increase in the plasma concentration of FFA. A highly significant correlation (r = 0·84 or 0·85; P< 0·01) existed between plasma ACTH and FFA levels 30 and 60 min after the start of stress. Dexamethasone completely prevented lipid mobilization while adipose tissue in vivo and in vitro responded strongly to ACTH. Consequently the lack of response of plasma FFA concentrations in corticosteroid-treated rabbits seemed to be linked to the failure of the pituitary gland to mobilize ACTH rather than to the unresponsiveness of adipose tissue to the pituitary hormone. Our data indicate that in the rabbit, under conditions of stress, ACTH is of greater physiological importance than catecholamines in the control of lipolysis.

♦ Background Normal saline (0.9% NaCl) is used during various abdominal surgical interventions and during peritoneal dialysis to rinse the peritoneal cavity. Although no clear clinical evidence exists for the bioincompatibility of normal saline, various experimental studies have suggested that 0.9% NaCl solution can initiate fibrosis of peritoneum. ♦ Material and Methods We review the data derived from in vitro and in vivo experimental studies demonstrating the cytotoxic effect of 0.9% NaCl and its ability to initiate peritoneal adhesions. ♦ Results Normal saline reduces the viability and fibrinolytic activity of peritoneal mesothelial cells. Use of normal saline to wash the peritoneal cavity during abdominal operations or after chronic peritoneal dialysis is more likely to produce adhesions than is no irrigation at all. Chronic exposure of the peritoneum to normal saline causes overgrowth of the connective tissue and formation of new blood vessels within that tissue. Conclusion ♦ Normal saline is a bioincompatible solution that predisposes to the formation of peritoneal adhesions and fibrosis of the peritoneum. A 0.9% NaCl solution should therefore not be used to rinse the peritoneal cavity after interruption of peritoneal dialysis.

В ранних исследованиях была выявлена способность аргининсодержащих пептидов глипролинового ряда в условиях in vitro оказывать антикоагулянтные и суммарные фибринолитические эффекты на плазму крови лабораторных крыс. Цель исследования: изучение действия аргининсодержащих глипролинов HisPheArgTrpProGlyPro (HFRWPGP), ThrLysProArgProGlyPro (TKPRPGP), ArgGluArgProGlyPro (RERPGP) на параметры системы гемостаза крыс в условиях in vivo в норме и сравнительный анализ эффектов исследуемых пептидов с глипролином (PGP). Материалы и методы. В работе использовали пептиды HFRWPGP (АКТГ(69)PGP), TKPRPGP (Селанк), RERPGP и PGP. В опытах 20 белым лабораторным крысам интраназально вводили по 0,02 мл раствора каждого из пептидов в дозе 1 мкг/кг массы тела (3 опытных группы) или 0,85 раствора NaCl (контрольная группа) через каждые 24 ч в течение 7 суток (168 ч). Влияние пептидов на параметры гемостаза исследовали тромбоэластографическим методом, в тестах активированного частичного тромбопластинового времени и агрегации тромбоцитов, а также определяли фибринолитическую активность крови на нестабилизированном фибрине. Результаты. Каждый из исследованных препаратов обнаруживал антикоагулянтную, фибринолитическую, антитромбоцитарную активности. Противосвертывающие эффекты проявлялись через 20 ч после последнего введения и сохранялись на протяжении 168 ч. Наибольшим эффектом обладали пептиды, в состав которых входил аргинин ArgGluArgProGlyPro и Селанк. Заключение. Аминокислота аргинин в составе олигопептидов глипролинового ряда способствует усилению активации противосвертывающей системы. Introduction. Early in vitro studies revealed anticoagulant and fibrinolytic effects of argininecontaining glyprolins on plasma in rats. Aim: to study the effects of argininecontaining glyprolines HisPheArgTrpProGlyPro (HFRWPGP), ThrLysProArgProGlyPro (TKPRPGP), ArgGluArgProGlyPro (RERPGP) on rats hemostasis in vivo in normal conditions and to compare the effects of test peptides with glyproline (PGP). Materials and methods. The following peptides were used: HFRWPGP (ACTH(69)PGP), TKPRPGP (Selank), RERPGP and PGP. In experiments 20 white rats were intranasally injected with 0.02 ml of each peptide solution in a dose of 1 g/kg body weight (3 experimental groups) or 0.85 NaCl solution (control group) every 24 hours for 7 days (168 hours). The effects of peptides on hemostatic parameters were investigated by thromboelastographic method, by activated partial thromboplastin time and platelet aggregation, also fibrinolytic blood activity on nonstabilized fibrin was determined. Results. All tested peptides showed anticoagulant, fibrinolytic, antiplatelet activity. Anticoagulant effects manifested 20 hours after the last injection and persisted for 168 hours. Argininecontaining ArgGluArgProGlyPro and Selank showed the greatest effect. Conclusion. Arginine as a part of argininecontaining glyprolines intensified the activation of anticoagulation system.

Magnetic micro- and nanoparticles (MPs)-based composite materials are widely used in various applications in electronics, biotechnology, and medicine. This group of silicone composites have advantageous magnetic and mechanical properties as well as sufficient flexibility and biocompatibility. These composites can be applied in medicine for biological sensing, drug delivery, tissue engineering, and as remote-controlled microrobots operating in vivo. In this work, the properties of polydimethylsiloxane (PDMS)-based composites with different percentages (30 wt.%, 50 wt.%, 70 wt.%) of NdFeB microparticles as a filler were characterized. The novelty of the work was to determine the influence of the percentage of MP content and physiological conditioning on the properties of the PDMS-MP composites after in vitro incubation. An important essence of the work was a comprehensive study of the properties of materials important from the point of view of medical applications. Materials were tested before and after conditioning in 0.9 wt.% NaCl solution at a temperature of 37 °C. Several studies were carried out, including thermal, physicochemical, and rheological tests. The results show that with an increase of the incubation time, most of the measured thermal and physicochemical parameters decreased. The presence of the magnetic filler, especially at a concentration of 70 wt.%, has a positive effect on thermal stability and physicochemical and rheological properties. The performed tests provided important results, which can lead to further research for a broader application of magnetic composites in the biomedical field.

Abstract Background: As a non-contact, non-invasive medical imaging technique, magnetic induction tomography (MIT) can measure the conductivity distribution inside the human body. Numerical calculation and physical phantom tests have showed the accuracy of MIT in principle. However, very few human studies have limited the development and application. At the key stage before clinical trials, animal experiment will be an effective method to verify the feasibility of in vivo detection the conductivity variation inside of biological body. Methods: An abdominal subcutaneous injection rabbit model was used to simulate the local conductivity perturbation by injecting a 0.9% NaCl solution along with in vitro heparinized blood step by step. An insulated and sliding operation console was built to carry out the animal tests. An improved 16-channel MIT data acquisition system was used to record the data at 13.56 MHz and 4 seconds per frame. A series of time-difference reconstructed images, relative to non-injection, were obtained by the regularized Newton-Raphson algorithm for every 3 mL of injection. Results: 15 rabbits were divided by two groups. Six rabbits were injected with 0.9% NaCl solution and nine rabbits were injected with the in vitro heparinized blood. The target with an increased conductivity distribution can clearly be observed in all the reconstructed images. The maximum target value in each image increased with the injection dosage. The slopes of the regressive line for the mean of maximum target value in two groups were statistically different. Conclusion: Local conductivity perturbation inside of the rabbits is able to be reconstructed. The position and conductivity difference relative to the surrounding tissue of the target can be reflected correctly. This preliminary rabbit test shows the feasibility of the in vivo application for MIT and will be the basis for further animal tests and clinical trials in the future.

SummaryThe aim of this study was to establish a basic manipulation protocol of preantral follicles for deriving developmentally competent oocytes. Primary, early and late secondary follicles retrieved from the ovaries of 14-day-old F1 (C57BL/6 × DBA2) female mice mechanically or enzymatically were cultured singly and in vitro growth of the follicles and maturation of intrafollicular oocytes were subsequently monitored. A mechanical method retrieved more (p < 0.0001) follicles (339 ± 48 vs. 202 ± 28) than an enzymatic method. However, the enzymatic method collected more singly isolated follicles that could be provided for subsequent culture (102 ± 26 vs. 202 ± 28). When an enzymatic method was employed, early and late secondary follicles required 9 and 6 days for reaching the maximal incidence of the pseudoantral stage. However, primary follicles were not possible to develop into the pseudoantral stage. The optimal duration of oocyte maturation from the onset of follicle culture was 7 days and 5–7 days for early and late secondary follicles, respectively. A general decrease in oocyte diameter (65.2–65.53 μm vs. 75 μm) and zona thickness (5.41–5.74 μm vs. 7.76 μm) was detected in in vitro-derived compared with in vivo-derived matured oocytes. Pronuclear formation was detected in 86–94% of mature oocytes after parthenogenetic activation and no significant difference was detected among groups. These results showed that preantral follicles retrieved by an enzymatic method underwent step-by-step growth in vitro, which could yield mature oocytes.

Gonadotropins (GTHs), FSH and LH, play central roles in vertebrate reproduction. Here, we report the production of biologically-active recombinant FSH (r-mtFSH) and LH (r-mtLH) of an endangered salmon species, Manchurian trout (Brachymystax lenok), by baculovirus in silkworm (Bombyx mori) larvae. The biological activities of the recombinant hormones were analyzed using COS-7 cell line transiently expressing either amago salmon FSH or LH receptor. The steroidogenic potency of the r-mtFSH and r-mtLH was examined by a culture system using rainbow trout follicles in vitro. In vivo, bioactivity was assessed by measuring ovarian weight, oocyte diameter, and plasma steroid hormone levels in female rainbow trout. Moreover, inducing potency of milt production were examined in vivo using goldfish. Our results demonstrated that the r-mtFSH and r-mtLH were successfully produced in the baculovirus-silkworm system and recognized by their cognate receptors specifically in vitro. The production of estradiol-17β (E2) and testosterone (T) was stimulated by the r-mtFSH and r-mtLH respectively, from the full-grown follicles of rainbow trout, whereas both E2 and T were increased by relatively higher doses of the recombinant hormones from the follicles of the maturing stage. In in vivo assay, injection of the r-mtFSH but not r-mtLH increased ovarian weight, oocyte diameter, and plasma E2 levels in immature rainbow trout. Injection of both r-mtFSH and r-mtLH induced milt production in male goldfish. In conclusion, the present study strongly suggests that the r-mtFSH and r-mtLH have distinct biological properties, such as a specific responsiveness for the cognate receptor, steroidogenic, and vitellogenic activities for ovarian follicles in salmonids. These recombinant FSH and LH may be applied for future studies on the gonadal development and maturation in fishes as well as the endangered salmon species.

Development of antibiotic-resistant strains of microorganisms is a dangerous phenomenon, actively progressing every year. The uncontrolled use of antibiotics for animals, accumulation in products of animal origin ultimately poses a danger to human health. That is why a decrease in the use of antibiotics and searching alternatives of antibiotic is acute and relevant issues. Therefore, the aim of our research was to study the antimicrobial properties of ozone in relation to pathogens of purulent infection and to show the possibility of using ozone therapy as a potential method of antimicrobial therapy for animals. The materials for determining antimicrobial effect of ozone was 12 samples of purulent exudate in an amount of 2 ml, taken from dogs with purulent wounds before and after sanitation by ozonized 0.87% NaCl solution. Complexity course of wound process with purulent inflammation largely depends on from degree of microbial contamination of the wound and species composition of microorganisms. Therefore, an important aspect in our research was the study of the antimicrobial properties of ozone on its action of purulent exudate «in vitro» and «in vivo» and the determination of the bactericidal effect on microorganisms. The most stable and informative indicator of assessing nature of purulent-inflammatory process is the determination of total number of microorganisms in 1 ml of discharge from a purulent wound. The total microbial number was determined by the method of serial dilutions according to Pasteur. Serial ten-fold dilutions from 10-1 to 10-9 were prepared from purulent exudate in test tubes with sterile MPB (9 ml each). Species composition of microorganisms was determined by cultural and biochemical properties of cultivated microbial colonies, followed microscopy of smears from pure cultures stained using method of Gram. Samples of purulent exudate were subjected to microbiological examination before treatment, and after 30 minutes of washing by ozonized isotonic solution NaCl (ozone concentration of 7 mg/ml). A microbiological study of purulent exudate was also carried out, pre-treated with ozone at a concentration of 7 mg/ml «in vitro» in a test tube in an amount of 2 ml by passing it through exudate (sparging) at a flow rate of 0.5 L/min and a processing time of 10 min. Samples were examined immediately after sampling and sparging. "Microbial landscapes" of purulent wounds were presented by associations of Staph. aureus, Str. faecalis, E. coli. Microbial seeding of purulent exudate for treatment ranged from 6.6 • 10-10 to 3.7 • 10-8 CFU/ml, however, after 10 min of bubbling «in vitro» at an ozone concentration of 7 mg/ml, the degree of microbial seeding of samples did not exceed 10-4 CFU/ml and ranged from 3.1 • 10-4 to 2.3• 10-3 CFU/ml. As shown by the results of microbiological studies, the growth of microorganisms on a nutrient medium in bacteriological plates with purulent exudate samples treated with ozone with concentration of 7 mg/ml was already absent at 10-5 degrees of dilution, which indicates the pronounced antimicrobial properties of ozone. The study «in vivo»also indicates that even after a single use of an ozonized isotonic NaCl solution at an ozone concentration of 7 mg/ml, it completely prevents the growth of Staph. aureus, Str. faecalis, E. coli. and causes 100% death mentioned associations of microorganisms. The results of microbiological studies are confirmed by clinical data. So, on the third day of treatment, the animals in the lesion zone had a small amount of wound exudate, and the microbial number of the latter was 1.4 • 10-4 − 3.1 • 10-³ CFU/ml, below the critical level of contamination and in most cases not leads to the progression of a purulent-inflammatory process. Ozone destroys all types of bacteria, viruses, fungi and protozoa. At the same time, ozone at a concentration of 7 mg/ ml does not have an irritating effect on body tissues, therefore, ozone therapy can be considered as an additional or alternative therapy of bacterial infection. The differentiation and maintenance of many specialized epithelial structures are dependent on the underlying connective tissue stroma and on an intact basal lamina. These requirements are especially stringent in the development and maintenance of the skin and oral mucosa. The keratinization patterns of thin or thick cornified layers as well as the appearance of specialized functional derivatives such as hair and teeth can be correlated with the specific source of stroma which supports these differentiated expressions.
